Dimethyl sulfoxide was found to inhibit histamine release induced by compound 48/80 at concentrations ranging from 0.6 to 10%. At higher concentrations (20 and 40%) the substance caused histamine release by itself. A new method is described for assessing changes in vascular permeation by albumin in multiple tissues of the same animal in response to intravascular injection of vasoactive agents. Total and specific IgE anti-schistosome antibodies were quantitated in 31 patients with chronic schistosomiasis and in 15 controls. Both levels of total and specific IgE were significantly increased in sera of 74 and 68% of infected patients respectively. Histamine release from basophils by specific antigens was assessed using a spectrofluorimetric method. This test was found to be highly significant in all the patients studied. There was a significant correlation between specific IgE levels and histamine release (R = 0.43, p less than 0.05).

To detect a potential defect in immunoregulatory function in atopic subjects, we studied histamine-induced suppressor-T-cell activity and histamine Type 1 and Type 2 receptors on T cells. The delayed skin-test response to four antigens was assessed before and after 6 weeks' cimetidine therapy in patients with duodenal-ulcer disease. In the eight patients who received cimetidine there was a significant increase in both erythema and induration after six weeks' cimetidine therapy. In contrast, the intensity of delayed-hypersensitivity reactions at 6 weeks was not increased in eight control patients who did not receive cimetidine. These studies indicate that therapy with an H2-receptor antagonist may be associated with an augmentation of delayed hypersensitivity responses.
