Evaluation of Efficacy and Safety of Olaparib in Combination With Abiraterone for the Treatment of Metastatic Castration-Resistant Prostate Cancer (mCRPC) With Homologous Recombination Repair Gene Mutations.

INTRODUCTION: Metastatic castration-resistant prostate cancer (mCRPC) with homologous recombination repair (HRR) gene mutations remains a therapeutic challenge, particularly after progression on prior androgen receptor pathway inhibitors. This study aimed to evaluate the efficacy and safety of olaparib plus abiraterone versus olaparib monotherapy in this specific population. PATIENTS AND METHODS: This single-center retrospective cohort study included patients with HRR-mutated mCRPC who received olaparib-based therapy between January 2017 and December 2025. Patients were divided into the olapa…

Targeting homologous recombination deficiency with intensified chemotherapy versus standard chemotherapy followed by olaparib in stage III breast cancer (SUBITO): an open-label, randomised, controlled, phase 3 trial.

BACKGROUND: Patients with stage III, human epidermal-growth-factor-receptor 2 (HER2; also known as ERBB2)-negative breast cancer with homologous recombination deficiency (HRD) had a 4-year overall survival of 35% after anthracycline-based chemotherapy versus 78% after intensified alkylating chemotherapy with autologous stem cell rescue (IACT) in a post-hoc analysis of an earlier randomised controlled trial. In this study, we aimed to prospectively assess 4-year overall survival with IACT and establish whether this approach remains superior to a contemporary HRD-targeting regimen in patients wi…

A Podcast on Integrating Genetic Testing for Homologous Recombination Repair Gene Alterations in Patients with Prostate Cancer in the USA: a Multidisciplinary Approach to Overcoming the Obstacles.

Homologous recombination repair (HRR) gene alterations in prostate cancer predispose patients to more aggressive disease and a poorer prognosis. The presence of these HRR gene alterations may inform patient eligibility for clinical trials and targeted therapies, and importantly, through cascade testing, help identify family members at risk of developing an inherited cancer. This finding highlights the importance of testing for HRR gene alterations in patients with prostate cancer. Despite the potential implications of such testing to patient care, in a cross-sectional retrospective study in th…

Novel recombinant vaccinia virus-vectored vaccine affords complete protection against homologous Borrelia burgdorferi infection in mice.

The rising prevalence of Lyme disease (LD) in North America and Europe has emerged as a pressing public health concern. Despite the availability of veterinary LD vaccines, no vaccine is currently available for human use. Outer surface protein C (OspC) found on the outer membrane of the causative agent, Borrelia burgdorferi, has been identified as a promising target for LD vaccine development due to its sustained expression during mammalian infection. However, the efficacy and immunological mechanisms of LD vaccines solely targeting OspC are not well characterized. In this study, we developed a

Characterization of ectromelia virus deficient in EVM036, the homolog of vaccinia virus F13L, and its application for rapid generation of recombinant viruses.

The orthopoxvirus (OPV) vaccinia virus (VACV) requires an intact F13L gene to produce enveloped virions (EV) and to form plaques in cell monolayers. Simultaneous introduction of an exogenous gene and F13L into F13L-deficient VACV results in expression of the foreign gene and restoration of plaque size. This is used as a method to rapidly generate VACV recombinants without the need for drug selection. However, whether other OPVs require the orthologs of F13L to generate EV and form plaques, whether F13L orthologs and EV are important for OPV pathogenesis in natural hosts, and whether a system b

A recombinant insect-specific alpha-toxin of Buthus occitanus tunetanus scorpion confers protection against homologous mammal toxins.

We have constructed a cDNA library from venom glands of the scorpion Buthus occitanus tunetanus and cloned a DNA sequence that encodes an alpha-toxin. This clone was efficiently expressed in Escherichia coli as a fusion protein with two Ig-binding (Z) domains of protein A from Staphylococcus aureus. After CNBr treatment of the fusion protein and HPLC purification, we obtained approximately 1 mg recombinant apha-toxin/l bacterial culture. The toxin, called Bot XIV, displays no toxicity towards mammals but is active towards insects as shown by its paralytic activity against Blatella germanica co

Homologous recombination repair and breast cancer gene testing in prostate cancer: expert perspectives and practical guidance on best practice from sample acquisition to biomarker-informed clinical decision-making.

The prostate cancer (PCa) treatment landscape, particularly metastatic PCa, is rapidly evolving, with an increase in personalised treatment approaches and therapy selection being guided by tumour genomics. However, PCa remains the third leading cause of death among male-specific cancers in the European Union. Nearly a quarter of patients with metastatic PCa harbour homologous recombination repair (HRR) pathway mutations, with approximately 12% harbouring alterations in the BRCA1 or BRCA2 genes. These genomic alterations are associated with poorer prognosis and increased sensitivity to targeted…

Association of Prior PARP Inhibitor Exposure with Clinical Outcomes after 177Lu-PSMA-617 in Men with Castration-resistant Prostate Cancer and Mutations in DNA Homologous Recombination Repair Genes.

BACKGROUND AND OBJECTIVE: The prostate-specific membrane antigen (PSMA) radioligand 177Lu-PSMA-617 (LuPSMA) is approved for treatment of metastatic castration-resistant prostate cancer (mCRPC). PARP inhibitors (PARPi) are approved for patients with mCRPC and mutations in homologous recombination repair (HRR) pathway genes. Both modalities induce DNA damage and therefore may share mechanisms of resistance. We investigated whether PARPi exposure would reduce the subsequent efficacy of LuPSMA. METHODS: This retrospective study included 100 patients with a PARPi-qualifying HRR alteration treated …

Exploring the efficacy of PARP inhibitors in metastatic castration-resistant prostate cancer with homologous recombination repair alteration: a meta-analysis based on subgroups and reconstructed individual patient data.

BACKGROUND: Treatment for metastatic castration-resistant prostate cancer (mCRPC) harboring homologous recombination repair (HRR) alteration remains a challenge. Recently published trials have evaluated the poly (ADP-ribose) polymerase inhibitors (PARPIs) in mCRPC. However, the efficacy in subgroup with specific HRR gene mutation and treatment protocol requires further elucidation. This meta-analysis aims to explore the efficacy of PARPIs based on subgroups and reconstructed individual patient data (IPD). METHODS: Literature was searched using PubMed, Embase, Cochrane Library, and ClinicalTri…

The peroxidoxin 2 protein of the human parasite Onchocerca volvulus: recombinant expression, immunolocalization, and demonstration of homologous molecules in other species.

The peroxidoxin protein of the filarial parasite Onchocerca volvulus (OvPXN-2) belongs to a group of highly conserved antioxidant molecules. For a more detailed characterization of this protein and for determination of its expression pattern the OvPXN-2 protein was recombinantly expressed as a His-tagged protein. Under reducing conditions the recombinant protein had an apparent molecular mass of 28 kDa. Considering the size of the His-tag and the FLAG epitope introduced to the recombinant protein, this size is in agreement with that of the native protein identified in O. volvulus extract. Anti

Generation of a human embryonic stem cell line (SMUDHe010-A-82) carrying a homozygous c.1538G > A (p.G513D) mutation in the OSMR gene by CRISPR/Cas9-mediated homologous recombination.

Mutations in the tumor suppressor M receptor (OSMR) gene are associated with primary localized cutaneous amyloidosis (PLCA). Recently, we confirmed that OSMR loss-of-function mutations enhance epidermal keratinocyte differentiation via inactivation of the STAT5/KLF7 signaling. However, no disease model was available for PLCA. Accordingly, we generated an OSMR c.1538G > A mutant human embryonic stem cell line (SMUDHe010-A-82) using CRISPR/Cas9-mediated homologous recombination. The cell line preserves normal karyotype, pluripotency and the ability to differentiate into all three germ layers. Mo…

Simultaneous Inhibition of PARP-1 and CK2α by Novel Benzothieno[3,2-d]pyrimidin-4(1H)-one Derivatives: A Promising Strategy to Combat Homologous Recombination Deficiency-Independent Tumors and Metastasis.

Poly(ADP-ribose) polymerase 1 (PARP-1) inhibitors yield robust clinical benefits yet only work for homologous recombination deficiency (HRD) patients. To expand their utility and resolve clinical drawbacks, we synthesized dual PARP-1/casein kinase II alpha (CK2α) inhibitors built on benzofuran/benzothieno[3,2-d]pyrimidin-4(1H)-one cores with methyl (Z)-carbamohydrazonothioate pharmacophores. Among them, NJT-12 exhibited the optimal activity, with IC50 values of 3.40 and 4.31 nM against PARP-1 and CK2α. In non-HRD A2780 cells, NJT-12 modulates the Akt1Ser129-GSK-3βSer9-Wnt/β-catenin pathway to …

Impact of adjuvants on a recombinant SARS-CoV-2 spike vaccine: Protective efficacy and recall antibody response following homologous challenge.

Immune imprinting, or original antigenic sin, challenges the control of evolving severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). How the quantity and quality of pre-existing immunity modulate the recall antibody response upon re-exposure remains poorly understood. We immunized hamsters with a recombinant ancestral spike protein (S-2P) formulated with one of four distinct adjuvants to assess the impact of adjuvant-induced immunity on protective efficacy. Furthermore, we analyzed the modulatory effect of the adjuvant on the potency, breadth, and evolution of antibody responses afte
