Zygomycetes are saprophytic fungi found ubiquitously in nature. Generally, fungi are considered as the most common microbes encountered by mammalian hosts due to its ubiquity in nature. Among the fungi, the Zygomycetes represent the most basal terrestrial lineage which can cause infections in humans. They comprise two orders, the Mucorales and the Entomophthorales, which contain human pathogenic species. Members of the Mucorales are responsible for mucormycosis; the second most common mould fungi infection in the world and infection with members of the Entomophthorales can result in basidiobol

Drosophila melanogaster offers a powerful model system for interrogating interactions between host cells and human bacterial pathogens. Brucella, a gram-negative, facultative intracellular bacterium is the causative agent of brucellosis, a zoonotic disease of global consequence. Over the past several decades, pathogen factors that mediate Brucella infection have been identified. However, host factors that mediate infection have remained obscure. We have used the power of the Drosophila S2 cell system to identify and characterize host factors that support infection by Brucella melitensis. Host

Flavivirus is a genus of the family Flaviviridae. It includes West Nile virus (WNV), dengue virus (DENV), yellow fever virus (YFV), Japanese encephalitis virus (JEV), tick-borne encephalitis virus (TBEV), and several other viruses which lead to extensive morbidity and mortality in humans. To establish infection and replication in the hosts, flaviviruses have evolved a variety of strategies to modulate the host's immune responses. In this review, the strategies employed by flaviviruses to evade the innate and adaptive immunity of host are summarized based on current studies, with a major focus

Helicobacter pylori is a serious chronic transmissible pathogen that causes gastric structural and functional damage and is a major cause of morbidity and mortality worldwide. H. pylori is one of the digestive tract pathogens that has traveled with mankind since before humans moved out of Africa. Some have suggested that the long association of H. pylori with humans means there must be a benefit and suggest dire consequences will follow its eradication. Fortunately, there are now sufficient experiences with the outcome of treatment and with populations where the infection disappeared long ago

Chlamydia is an obligate intracellular pathogen that develops in the host cell in a vacuole termed the chlamydial inclusion. The prevailing concept of the chlamydial inclusion is of a parasitophorous vacuole. Here, the inclusion is the recipient of one-way host-pathogen interactions thus draining nutrients from the cell and negatively impacting it. While Chlamydia orchestrates some aspects of cell function, recent data indicate host cells remain healthy up until, and even after, chlamydial egress. Thus, while Chlamydia relies on the host cell for necessary metabolites, the overall function of …

Intracellular plant-microbe interactions rely on host-derived interface membranes: as sites for reciprocal nutrient and signal exchange during symbiosis, or as conduits for asymmetrical nutrient acquisition and effector delivery by pathogens. Sustaining these dynamic structures places substantial metabolic and vesicular trafficking demands on host cells. This review examines how cell cycle reprogramming may help plants meet these demands. During plant-pathogen interactions, biotrophic pathogens can reprogram host cell cycle pathways to establish metabolically favorable niches, whereas plant im

Virus-virus interactions can profoundly shape viral ecology and evolution, yet they remain poorly understood, particularly in haloarchaea. Upon characterization of a spindle-shaped virus, Tebenquiche spindle-shaped virus 1 (Tebi-SV1), isolated from Halorubrum strain TLS-6, we detected the presence of a second virus, Tebenquiche pleomorphic virus 1 (Tebi-PV1), that coinfects host cells. Gene sharing network analysis revealed several genomes related to Tebi-SV1 in public databases, most of which also co-occur with pleolipoviruses in their host genomes. Attempts to separate the two viruses were u

Despite growing interest in bat-virus interactions, experimental infection studies (EIS) remain taxonomically and geographically restricted, limiting inference about bat immunology and zoonotic risk. We performed a systematic literature search (Jan 1900-Jan 2025) and manually curated peer-reviewed primary studies of viral experimental infections in live bats and bat-derived cell lines (BdCL). From 545 candidate records we retained 127 papers (83 live-bat, 44 BdCL), comprising 315 unique bat-virus trials. We categorized host taxonomy, viral family (ICTV), and biogeographical congruence, to quan

Grade C molar-incisor pattern periodontitis (C-MIP) is a rapidly progressive form of periodontal disease affecting young individuals that is often linked to a highly virulent genotype of Aggregatibacter actinomycetemcomitans (Aa). Although Aa is present in the healthy oral microbiome, its transition into subgingival tissue correlates with the conversion from healthy to diseased status within the periodontal pocket. These changes may be due to immune evasion strategies attributed to Aa exotoxins. We previously demonstrated that a host cell protein, cellugyrin, plays a critical role in exotoxin

Chlamydia trachomatis has a significant impact on public health, especially among adolescents and young women; it primarily affects urogenital epithelial cells, leading to cervicitis and urethritis, with >90% of cases showing no symptoms. Consequently, chlamydial infections are commonly misdiagnosed, and, if untreated, they may result in severe reproductive sequelae including infertility. A better understanding of C. trachomatis cell biology and bacterial-host cell interactions may be helpful to identify strategies able to counter its transmission among the population, as well as its dissemina

An essential property associated with leptospiral virulence is the pathogen's ability to translocate across host cells, enabling Leptospira to evade the host immune response, disseminate, and establish infection. The cell monolayer translocation assay allows for the quantification of Leptospira strain's competence to cross cell barriers while measuring the integrity of the polarized eukaryotic cell monolayer during this process.

Metabolomics, a critical tool for analyzing small-molecule metabolites, integrates with genomics, transcriptomics, and proteomics to provide a systems-level understanding of fungal biology. By mapping metabolic networks, it elucidates regulatory mechanisms driving physiological and ecological adaptations. In fungal pathogenesis, metabolomics reveals host-pathogen dynamics, identifying virulence factors like gliotoxin in Aspergillus fumigatus and metabolic shifts, such as glyoxylate cycle upregulation in Candida albicans. Ecologically, it highlights fungal responses to abiotic stressors, includ

Bacterial viruses, or bacteriophages, are the most abundant viruses on Earth, and their hosts are the most widespread living organisms in the biosphere. They are found in a free state, as virions, but are also very abundant in bacterial genomes, as prophages. Bacteriophages are present in all biotopes colonized by bacteria, such as water, soils, extreme environments, human, animal, and plant microbiomes, where they contribute to genetic exchanges. Therefore, all factors affecting these environments have a major impact on the dynamics of bacterial populations and their viruses.

The nervous system, like the immune system, constantly interfaces with the environment, encountering threats, including pathogens. Recent discoveries reveal an emerging role for sensory neurons in host defense and immunity. Sensory neurons detect infections either by directly sensing microbial signals or through immune mediators. Beyond pathogen detection, they modulate immune responses and local inflammation by interacting with immune cells, influencing inflammation and pathogen clearance. Additionally, sensory neurons trigger protective reflexes - such as pain, coughing, sneezing, and itchin

Infections with Mycobacterium tuberculosis (Mtb) resulted in over one million deaths in 2024, the highest number for any infectious disease. With no vaccines that protect against pulmonary tuberculosis (TB) and the challenges associated with antibiotic therapy, there is a critical need to better understand host-Mtb interactions to help curb this major public health problem. Mtb is arguably the most successful human pathogen, and it survives in diverse environments, resulting in heterogeneous disease outcomes in patients. Five years ago, in my commentary in mSphere, I discussed how Mtb virulenc

The adverse outcomes of fungal infection in mammalian hosts depend on the complex interactions between the host immune system and pathogen virulence-associated traits. The main clinical problems arise when the host response is either too weak to effectively eliminate the pathogen or overly aggressive, resulting in host tissue damage rather than protection. This article will highlight current knowledge regarding the virulence attributions and mechanisms involved in the dual-sided role of the host immune system in the immunopathogenesis of the thermally dimorphic fungus Talaromyces marneffei thr
