Antibiotic release from keratin hydrogels for use in wound healing applications.

Skin injuries, particularly severe injuries, such as full-thickness wounds or surgical sites, are susceptible to bacterial infection. Materials composed of keratin proteins have been investigated for use in wound dressings, and at least two products based on these materials have received FDA clearance for treating radiation dermatitis via the 510(k) mechanism. We hypothesized that hydrogels composed of oxidatively extracted keratins, known as keratose, could achieve sustained release of several classes of antibiotic drugs that might improve the performance of keratins as wound dressing materia…

Colloidal stability of nanocellulose hydrogels in physiological and cell culture environments.

Nanocellulose is a promising alternative to animal-derived matrices for tissue engineering, yet its colloidal stability and rheological performance are heavily influenced by the ions and proteins in physiological environments. Here, we systematically investigated the colloidal stability and interfacial behavior of native (nCNF) and anionic (aCNF) cellulose nanofibrils in physiologically relevant media using rheology, zeta potential, static multiple light scattering (SMLS), and multi-parameter surface plasmon resonance (MP-SPR). Increasing ionic strength compressed the electrical double layer,

From flow to form: structuring and patterning hydrogels using microfluidic approaches.

Three-dimensional (3D) cell culture can leverage the precise arrangement of materials, known as patterning, to generate physiologically relevant tissue-like structures. Hydrogels are widely used in 3D cell culture due to their ability to mimic the properties of biological extracellular matrix networks. In this tutorial review, we discuss the use of microfluidic systems to control fluid movement and placement within fabricated microchannels to pattern hydrogel precursors in 3D through the use of capillary flow. Such systems offer unique advantages in their ability to create complex biomimetic s…

Tissue-derived extracellular matrix hydrogels instruct epigenetic adaptation in metastatic colonization.

The extracellular matrix (ECM) plays a central role in regulating tumor progression and metastatic colonization by providing biochemical and mechanical signals that shape cancer cell fate. However, most organoid culture systems rely on basement membrane extracts that fail to reproduce the tissue-specific extracellular environments encountered during metastasis. Here, we develop tissue-derived decellularized matrix hydrogels to reconstruct organ-specific microenvironments and investigate epigenetic adaptation to ECM cues during metastatic colonization. Patient-derived colorectal cancer organoid…

Multifunctional Hydrogels Loaded with Mn Single-Atom Nanozymes: Advancing Endometrial Repair.

Irregular endometrial damage often evolves into chronic conditions, such as endometrial thinning, functional tissue fibrosis, and intrauterine adhesions. These conditions pose significant challenges to female reproductive health. Addressing this, we developed a dynamic double-cross-linked hydrogel for the repair of fragile and dynamic endometrial tissue. This hydrogel integrates natural bioactive components (chitosan, protocatechuic aldehyde, and Fe(III)) and serves as a scaffold for Mn single-atom nanozymes (Mn SAN). Through dynamic covalent and hydrogen bonding, the hydrogel exhibits excepti

Vascularized skin tissue models featuring adipose cell spheroid-laden GelMA hydrogels.

The multifaceted tissue interplay between skin and adipose structures is increasingly recognized to play crucial roles in antimicrobial defense, hair cycling, wound healing, and thermogenesis. However, the technical challenges associated with the development of an in vitro model of such complex tissues include the difficulties of integrating tissues with diverse characteristics. Here, we present a method using a gelatin methacryloyl (GelMA) hydrogel to establish a microenvironment that hosts connected composite tissues: a vascularized skin layer and a subcutaneous adipose layer. When adipogene

4D Bioprinting of Self-Morphed Corneal Equivalents Using Smart Hybrid Hydrogels.

The curvature of the cornea is intrinsically linked to its topographical features, which are critical determinants of its optical and mechanical performance. While conventional bioprinting enables the fabrication of static, complex geometries, this study introduces a novel 4D bioprinting strategy that allows for the autonomous transformation of flat constructs into anatomically curved shapes, closely mimicking native corneal architecture. By engineering a concentration-dependent swelling gradient in gelatin methacrylate (Gel-MA), controlled, directional folding is achieved through differential

Multi-step functionalization of hydrogels through mechano- and photo-responsive linkages.

Patterning soft materials with cell adhesion motifs can be used to emulate the structures found in natural tissues. While patterning in tissue is driven by cellular assembly, patterning soft materials in the laboratory most often involves light-mediated chemical reactions to spatially control the presentation of cell binding sites. Here we present hydrogels that are formed with two responsive crosslinkers-an anthracene-maleimide adduct and a disulfide linkage-thereby allowing simultaneous or sequential patterning using force and UV light. Hydrogels were formed using poly(ethylene glycol)-based…

PDGF-Releasing Hydrogels for Enhanced Proliferation of Human Nucleus Pulposus Cells.

Hydrogels offer a promising solution for sustained and controlled drug delivery and cell-tissue biocompatibility. In the intervertebral disc (IVD), delivering growth factors faces challenges due to the antagonistic inflammatory environment and continuous mechanical stress, which can degrade biological agents and may reduce their local activity. To address this, we investigated the prolonged release of platelet-derived growth factor isoforms BB (PDGF-BB) and AB (PDGF-AB) by using N-desulfated heparin methacrylamide (Hep-N) crosslinked within matrix-metalloproteinase sensitive poly(ethylene glyc…

Viscoelasticity of Fibrous Hydrogels Driven by Dynamic Intrafibrillar Imine Cross-Linking.

Viscoelasticity of biological fibrous networks impacts cell fates and may reflect pathological conditions in vivo. Imine-cross-linked fibrous hydrogels can serve as effective in vitro models for studying viscoelastic properties of biological tissues; however, the specific role of intrafibrillar dynamic covalent bonds in governing hydrogel elasticity and stress relaxation remains unexplored. Here, for fibrous hydrogels derived from cellulose nanocrystals and polyethylene glycol, we systematically varied the content of intrafibrillar imine cross-links to explore their impact on hydrogels' elasti…

Toughness of Double Network Hydrogels: The Role of Reduced Stress Propagation.

Double network hydrogels show remarkable mechanical performance, combining high strength and fracture toughness with sufficient stiffness to bear load, despite containing only a low density of cross-linked polymer molecules in water. We introduce a simple mesoscale model of a double network material, detailed enough to resolve the salient microphysics of local plastic bond breakage, yet simple enough to address macroscopic cracking. Load sharing between the networks results in a delocalization of stress such that the double network inherits both the stiffness of its stiff-and-brittle sacrifici…

Injectable myocardium-derived hydrogels with SDF-1α releasing for cardiac repair.

Myocardial infarction (MI) is a predominant cause of morbidity and mortality globally. Therapeutic chemokines, such as stromal cell-derived factor 1α (SDF-1α), present a promising opportunity to treat the profibrotic remodeling post-MI if they can be delivered effectively to the injured tissue. However, direct injection of SDF-1α or physical entrapment in a hydrogel has shown limited efficacy. Here, we developed a sustained-release system consisting of SDF-1α loaded poly(lactic-co-glycolic acid) nanoparticles (PLGA NPs) and an injectable porcine cardiac decellularized extracellular matrix (cdE
