Inadequate booster uptake threatens the success of immunization campaigns as seen with the recently rolled-out R21 malaria vaccine. The ability to administer both prime and boost immunizations with a single injection would therefore save lives and alleviate health care burdens. We present a platform for delayed delivery of the booster dose that is scalable with existing technology, easily injectable, and protective against malaria in vivo. Using chip-based microfluidics, we encapsulated the R21 malaria vaccine in polymer microcapsules that release their content weeks to months postinjection. C

A method for active immunisation of ducklings during the raising period (between the first and 20th days of age) was devised in another attempt to control erysipelas septicaemia of duck on freeland fattening sites. Spirovak erysipelas vaccine "Dessau" was aerogenically applied through vortex injector nozzles, when the animals were between five and 20 days of age. Immunisation took place in groups of 5,000 animals each. The vaccine concentrations were four doses per cubic metre in the first immunisation and eight doses per cubic metre in the second. Exposures were 30 minutes in the first immuni

Sensitized spleen and peripheral lymph node lymphocytes were tested after different types of immunization with allogeneic tumour cells for their capacity to induce macrophage cytotoxicity in vitro. The macrophages were rendered cytotoxic either by direct contact with lymphocytes and tumour cells (activation of macrophages) or by a factor (macrophage arming factor, MAF), released by the sensitized lymphocytes incubated with tumour cells (arming of macrophages). Both types of reactions are T-cell dependent. Macrophage activation is a more sensitive way to detect lymphocytes with the capacity to

It is well known that newborns and infants respond poorly to immunization with influenza virus vaccines. The poor response of neonates may be related to restricted B cell repertoire and high susceptibility of neonates to high dose tolerance. Protective antibody response against hemagglutinin (HA) of influenza virus is a T-dependent response. While the immunization of neonates with live virus caused a long lasting unresponsiveness, the immunization with a plasmid containing influenza virus HA circumvents the neonatal unresponsiveness. Genetic immunization primes efficiently neonatal HA-specific

BACKGROUND: In the context of ongoing debate about whether a single dose of the yellow fever (YF) vaccine provides lifelong protection, addressing key unanswered questions-such as the extent to which revaccination enhances humoral and cell-mediated immune (CMI) responses, and the true duration of immunity-is particularly important for protecting high-risk groups. This systematic review evaluated the immunogenicity of YF revaccination, to support evidence-based vaccination policies.
METHODS: A systematic search in electronic databases was conducted to identify relevant studies that evaluated h

Specific antibody responses were assessed in pigs immunized with the Taenia solium vaccine TSOL18. Anti-TSOL18 responses were compared 2 weeks after secondary immunization, where the interval between primary and secondary immunization was 4, 8, 12, 16 or 20 weeks. All animals responded to the vaccine and there was no diminution in antibody responses in animals receiving their second injection after an interval up to 20 weeks. Pigs receiving vaccinations at an interval of 12 weeks developed significantly increased antibody responses compared with animals receiving immunizations 4 weeks apart (P

Respiratory syncytial virus (RSV) is the leading cause of childhood hospitalizations due to bronchiolitis in children under 5 years of age. Moreover, severe RSV disease requiring hospitalization is associated with the subsequent development of wheezing and asthma. Due to the young age in which viral protection is needed and risk of vaccine enhanced disease following direct infant vaccination, current approaches aim to protect young children through maternal immunization strategies that boost neutralizing maternal antibody (matAb) levels. However, there is a scarcity of studies investigating th

Streptococcus pneumoniae (Spn) is a common pathogen causing a secondary bacterial infection following influenza, which leads to severe morbidity and mortality during seasonal and pandemic influenza. Therefore, there is an urgent need to develop bacterial vaccines that prevent severe post-influenza bacterial pneumonia. Here, an improved Yersinia pseudotuberculosis strain (designated as YptbS46) possessing an Asd+ plasmid pSMV92 could synthesize high amounts of the Spn pneumococcal surface protein A (PspA) antigen and monophosphoryl lipid A as an adjuvant. The recombinant strain produced outer m

BACKGROUND: Childhood immunization is among the most cost-effective public health strategies for reducing under-five morbidity and mortality. However, full immunization coverage for age remains suboptimal in many low- and middle-income countries due to factors related to caregivers, children, health systems, and sociocultural contexts. The study aimed to assess the prevalence of full immunization for age and identify its associated determinants across multiple domains.
METHODS: A community-based cross-sectional study was conducted among 364 caregiver-child pairs. Full immunization for age was

In the intraerythrocytic protozoan parasites of the genus Babesia both innate and adaptive immune responses are necessary to confer protection against clinical disease. In particular, the adaptive immune response involves the production of neutralizing antibodies as well as the presentation of parasite antigens to CD4+ T lymphocytes by professional antigen-presenting cells. Therefore, the development of alternative vaccines that replace the use of live attenuated strains should include relevant epitopes targeting both B and T cell responses. The aim of this study was to design new Babesia bige

Advances in vaccine technology over the past two centuries have facilitated far-reaching impact in the control of many infections, and today's emerging vaccines could likewise open new opportunities in the control of several diseases. Here we consider the potential, population-level effects of a particular class of emerging vaccines that use specific viral vectors to establish long-term, intermittent antigen presentation within a vaccinated host: in essence, "self-boosting" vaccines. In particular, we use mathematical models to explore the potential role of such vaccines in situations where cu

Tuberculosis incidence has made re-emergence due to multidrug resistance of the bacilli and for acquired immunodeficiency syndrome. Of all vaccines BCG is most widely used. It protects children from secondary forms of tuberculosis. Several authors recommend a second dose to boost the waning effect of BCG. The vaccine at present needs to be improved to make it more efficacious. Vaccines which involve the protective immune responses are ideal.

Eight rhesus monkeys immunized with a partially purified preparation of Plasmodium knowlesi gametes were monitored for over 6 years to determine the extent of transmission blocking immunity. Monkeys were challenged regularly, and anti-gamete antibodies were assayed by in vivo and in vitro mosquito feedings. Transmission blocking immunity persisted at high levels in most of the monkeys. In those animals in which protection waned between challenges, a challenge infection provided a sufficient booster effect to prevent infection of mosquitoes. Immunity to other stages of malaria (i.e., sporozoite

The authors present the results of studies on obtaining and comparative assessment of experimental anthrax diagnostic sera in experiments on various animals. Donkeys, sheep, horses, rabbits and monkeys (Papio hamadryas) were immunized with the STI-I vaccine by a single scheme. The activity of the obtained sera was tested in the diffuse precipitation reaction by the amount of the detected antibodies and the titre. The most active sera were obtained from donkeys and sheep: their titre was 5.5 and 4 times greater and amount of the detected antibodies 2.6--2 times greater than in the sera of horse

Immunization with vaccinia virus (VACV), the virus comprising the smallpox vaccine, induces memory CD8(+) T cells that protect from subsequent infections with smallpox in humans or the related ectromelia virus (ECTV) in mice. Memory CD8(+) T cells largely mediate these effects by expanding into secondary effectors that secrete the antiviral cytokine interferon-γ (IFN-γ) and induce cytolysis via releasing factors such as perforin, which permeabilizes target cells. We show that protection from ECTV infection after VACV immunization depends on the initial memory cell frequency and ability of expa

B1 cells, a subset of B lymphocytes whose developmental origin, phenotype, and function differ from that of conventional B2 cells, are the main source of "natural" IgM but can also respond to infection by rapidly producing pathogen-specific IgM directed against T-independent antigens. Francisella tularensis (Ft) is a Gram-negative bacterium that causes tularemia. Infection with Ft Live Vaccine Strain activates B1 cells for production of IgM directed against the bacterial LPS in a process incompletely understood. Here we show that immunization with purified Ft LPS elicits production of LPS-spec

INTRODUCTION: Vaccine-specific immune responses vary between populations and are often impaired in low income, rural settings. Drivers of these differences are not fully elucidated, hampering identification of strategies for optimising vaccine effectiveness. We hypothesise that urban-rural (and regional and international) differences in vaccine responses are mediated to an important extent by differential exposure to chronic infections, particularly parasitic infections.
METHODS AND ANALYSIS: Three related trials sharing core elements of study design and procedures (allowing comparison of out

Given that TB still constitutes a tremendous public health problem at the start of the 21st Century, it may come as a surprise that Bacillus Calmette-Guérin (BCG), developed nearly 100 years ago, is today still the only vaccine available against TB. Owing to its limited efficiency in controlling TB, much effort has been deployed to develop new, improved vaccines, with initial preclinical models showing encouraging results. However, since most individuals worldwide have been vaccinated with BCG, new vaccine developments have to be placed in that context. Consequently, several approaches explore
