Auxin protects Arabidopsis thaliana cell suspension cultures from programmed cell death induced by the cellulose biosynthesis inhibitors thaxtomin A and isoxaben.

BACKGROUND: Thaxtomin A (TA) is a natural cellulose biosynthesis inhibitor (CBI) synthesized by the potato common scab-causing pathogen Streptomyces scabies. Inhibition of cellulose synthesis by TA compromises cell wall organization and integrity, leading to the induction of an atypical program of cell death (PCD). These processes may facilitate S. scabies entry into plant tissues. To study the mechanisms that regulate the induction of cell death in response to inhibition of cellulose synthesis, we used Arabidopsis thaliana cell suspension cultures treated with two structurally different CBIs,

The Production of Human β-Glucocerebrosidase in Nicotiana benthamiana Root Culture.

Gaucher disease is caused by a deficiency of the enzyme glucocerebrosidase (GCase). Currently, enzyme-replacement therapy using recombinant GCase produced in mammalian cells is considered the most effective treatment. Plants are an attractive alternative host for recombinant protein production due to the low cost of large-scale production and lack of risk of contamination by human pathogens. Compared to whole plants, root cultures can grow faster. Therefore, this study aimed to produce recombinant GCase in a Nicotiana benthamiana root culture. Root culture of a GCase-producing transgenic plant

Endosulfan Degradation by Selected Strains of Plant Growth Promoting Rhizobacteria.

Sixty endosulfan tolerant bacterial strains were isolated from pesticide stressed agricultural soils. Five most tolerant strains were tested for plant growth promoting (PGP) activities and endosulfan degradation under different optimizing conditions in broth and soil. The strains PRB101 and PRB77 were the most efficient in terms of endosulfan degradation and PGP activities and showed solubilization indexes of 3.3 and 3.1 mm, indole acetic acid production of 71 and 68 μg mL-1, siderophore zones of 13 mm each at the recommended dosage, respectively. Hydrogen cyanide and ammonia production remain

The auxin conjugate indole-3-acetyl-aspartate affects responses to cadmium and salt stress in Pisum sativum L.

The synthesis of IAA-amino acid conjugates is one of the crucial regulatory mechanisms for the control of auxin activity during physiological and pathophysiological responses. Indole-3-acetyl-aspartate (IAA-Asp) is a low molecular weight amide conjugate that predominates in pea (Pisum sativum L.) tissues. IAA-Asp acts as an intermediate during the auxin degradation pathway. However, some recent investigations suggest a direct signaling function of this conjugate in various processes. In this study, we examine the effect of 100 μM IAA-Asp alone and in combination with salt stress (160 mM NaCl)

Structures and biological activities of three synaptic antagonists from orb weaver spider venom.

The venom of Argiope aurantia, an orb weaver spider, contains a mixture of low molecular weight "argiotoxins", which block neuromuscular transmission in insects. Complete structure elucidation of three argiotoxins reveals common features; a hydrophilic, basic domain of arginine, a polyamine and asparagine is connected to an aromatic moiety contributed either by 4-hydroxyindole-3-acetic acid or 2,4-dihydroxyphenylacetic acid. Structural assignments of two argiotoxins are verified by chemical synthesis. The argiotoxins cause reversible paralysis when injected into insects and this is correlated

Proline/serine-rich coiled-coil 1 alleviates atherosclerosis via remodeling tryptophan metabolism mediated by Akkermansia muciniphila.

Genome-wide association studies have implicated proline/serine-rich coiled-coil 1 (PSRC1) in coronary artery disease (CAD) pathogenesis. Our previous studies demonstrated that Psrc1 deficiency accelerates atherosclerosis via gut microbial dysbiosis, characterized by a substantial depletion of Akkermansia muciniphila. Recent studies implicate microbiome-dependent tryptophan metabolism as a novel checkpoint in atherosclerosis, with specific microbial taxa regulating metabolite-driven immune responses. The mechanism by which Psrc1 modulates atherosclerosis through A. muciniphila and its regulatio

Urinary 5-hydroxytryptophol following acute ethanol consumption: clinical evaluation and potential aviation applications.

HYPOTHESIS: The unknown prevalence of alcohol use and misuse among aviation pilots, crewmembers and associated support personnel call for continuous improvement of methods for detecting recent alcohol use. Early detection is essential to proper treatment and prevention of potentially catastrophic mishaps. Urinary 5-hydroxytryptophol (5HTOL), a serotonin (5HT) metabolite, has shown promise in the clinical setting as a noninvasive marker of recent alcohol consumption. METHODS: The urinary 5HTOL concentrations of 11 male and female subjects were followed for approximately 24 h following dosing w

Structural insights into Arabidopsis thaliana CTS-mediated fatty acid and hormone metabolism.

In plants, peroxisomal β oxidation is essential for energy production, sugar metabolism and the regulation of hormonal signaling. The Arabidopsis COMATOSE (CTS) gene, which encodes the peroxisomal ATP-binding cassette transporter ABCD1 (also known as CTS or PAX1), mediates the import of various fatty acyls and hormones into peroxisomes for β oxidation. Although physiological evidence has demonstrated that defects in CTS impair plant development and growth, the underlying molecular mechanisms remain poorly understood. In this study, biochemical assays confirm that CTS transports a broad range o

Metabolite-Mediated Alleviation of Iron Deficiency and Growth Promotion of Malus hupehensis by Bacillus licheniformis LCDD6 in Calcareous Soil.

Calcareous soils are typically deficient in essential nutrients such as iron, phosphorus, and potassium, which frequently results in nutrient deficiency in fruit trees. Bacillus licheniformis LCDD6 markedly enhanced Malus hupehensis seedling growth and plant iron nutrition in calcareous soil. This study aimed to elucidate the mechanism underlying these beneficial effects of strain LCDD6 under iron deficiency. Transcriptomic analysis revealed that iron deficiency induced metabolic reprogramming in strain LCDD6, characterized by a significant upregulation of genes involved in the biosynthesis of

Evidence That PbrSAUR72 Contributes to Iron Deficiency Tolerance in Pears by Facilitating Iron Absorption.

Iron is an essential trace element for plants; however, low bioactive Fe in soil continuously places plants in an Fe-deficient environment, triggering oxidative damage. To cope with this, plants make a series of alterations to increase Fe acquisition; however, this regulatory network needs further investigation. In this study, we found notably decreased indoleacetic acid (IAA) content in chlorotic pear (Pyrus bretschneideri Rehd.) leaves caused by Fe deficiency. Furthermore, IAA treatment slightly induced regreening by increasing chlorophyll synthesis and Fe2+ accumulation. At that point, we i

In vitro antagonistic activity, plant growth promoting traits and phylogenetic affiliation of rhizobacteria associated with wild plants grown in arid soil.

The role of plant growth-promoting rhizobacteria (PGPR) in adaptation of plants in extreme environments is not yet completely understood. For this study native bacteria were isolated from rhizospeheric arid soils and evaluated for both growth-promoting abilities and antagonistic potential against phytopathogenic fungi and nematodes. The phylogentic affiliation of these representative isolates was also characterized. Rhizobacteria associated with 11 wild plant species from the arid soil of Almadinah Almunawarah, Kingdom of Saudi Arabia (KSA) were investigated. From a total of 531 isolates, only

Mutation of Mediator subunit CDK8 counteracts the stunted growth and salicylic acid hyperaccumulation phenotypes of an Arabidopsis MED5 mutant.

The Mediator complex functions as a hub for transcriptional regulation. MED5, an Arabidopsis Mediator tail subunit, is required for maintaining phenylpropanoid homeostasis. A semidominant mutation (ref4-3) that causes a single amino acid substitution in MED5b functions as a strong suppressor of the pathway, leading to decreased soluble phenylpropanoid accumulation, reduced lignin content and dwarfism. By contrast, loss of MED5 results in increased concentrations of phenylpropanoids. We used a reverse genetic approach to identify suppressors of ref4-3 and found that ref4-3 requires CDK8, a kina

Long-term potentiation in the presence of NMDA receptor antagonist arylalkylamine spider toxins.

The role of the NMDA receptor (NMDAR) in long-term potentiation (LTP) is now well established. All potent NMDAR antagonists known to date inhibit the induction of LTP at the Schaffer collateral-CA1 pyramidal cell synapse in rat hippocampus, regardless of their site and mechanism of action. Arylalkylamine toxins are noncompetitive NMDAR antagonists in the mammalian central nervous system (CNS). The synthetic toxins argiotoxin-636 (Arg-636), Joro spider toxin (JSTX-3), alpha-agatoxin-489 and -505 (Agel-489 and Agel-505) and philanthotoxin-433 (delta-PhTX) were found in the present study to have

Cellular specificity of the gravitropic motor response in roots.

A number of features of the gravitropic response of roots are not readily accounted for by the classical Cholodny-Went theory. These include the observations that (i) in the later stages of the response the growth gradient is reversed with no evident reversal of the auxin gradient; (ii) a major component of the acceleration of growth along the upper side occurs in the distal elongation zone (DEZ), a group of cells located between the meristem and the main elongation, not within the central elongation zone; and (iii) the initiation of differential growth in the DEZ appears to be independent of

Honeybee pollination enhances pollen germination, pollen tube growth, phytohormone responses and gene expression in pear pistils.

BACKGROUND: Honeybee pollination significantly enhances the yield of pear (Pyrus spp.), yet the early physiological mechanisms of pollination and fertilization remain underexplored. In this study, pollen germination, pollen tube growth, and ovule development were analyzed, along with the phytohormones in pistil and the gene expression associated with phytohormone biosynthetic and signal transduction pathways to the differential mechanisms after honeybee and artificial pollination. RESULTS: The results demonstrated that, compared to artificial pollination, honeybee pollination induced more eff

Pleiotropic Phenotypes of the Tomato diageotropica Mutant Enable Resistance to Ralstonia solanacearum.

Quantitative disease resistance (QDR) is the most common form of disease resistance in crops, but it is challenging to understand at the cellular level due to the involvement of many genes and biological processes. Ralstonia solanacearum, the causal agent of bacterial wilt disease, is a destructive plant pathogen of Solanaceous species that is best controlled by quantitatively resistant varieties, but few QDR genes are known. We previously found that a tomato auxin pathway mutant known as diageotropica (dgt) has enhanced resistance to R. solanacearum. Here, we show that, as with wild-type quan

Kidney clearances of protein-bound uremic toxins predict outcomes in chronic kidney disease: a prospective cohort study.

This prospective cohort study investigated the kidney clearance of protein-bound uremic toxins (PBUTs)-specifically, indole sulfate (IS), p-cresol sulfate (pCS), and indole-3-acetic acid (IAA)-across various stages of chronic kidney disease (CKD) stages and their associations with adverse clinical outcomes. From July 2018 to December 2020, 186 non-dialysis CKD patients were enrolled and followed until June 2025. Serum and 24-hour (24 h) urine PBUT levels were measured, and kidney clearances were analyzed. The findings indicated a decrease in the 24 h kidney clearances of IS (Cis), pCS (Cpcs),

Mining and identification of factors influencing multi-branch plasticity in ornamental kale.

Transcriptome-revealed plant hormones and nutrients are key factors influencing branching in ornamental kale. Topping treatment and exogenous hormones application revealed that auxin and SLs inhibited lateral buds outgrowth, respectively. Plant architecture is a crucial horticultural characteristic in ornamental kale as the variety of branching patterns significantly enhances the esthetic appeal of garden plants. The factors influencing multi-branch plasticity in ornamental kale are yet to be elucidated. In this study, we mined the key branching genes by comparing the transcriptomes of a singl
