A heteromeric Texas coral snake toxin targets acid-sensing ion channels to produce pain.

Natural products that elicit discomfort or pain represent invaluable tools for probing molecular mechanisms underlying pain sensation. Plant … like proteins that together function as a potent, persistent and selective agonist for acid-sensing ion channels (ASICs), showing equal or greater efficacy compared with acidic pH. MitTx is highly selective for the ASIC1 subtype at neutral pH; under more acidic conditions (pH < 6.5), MitTx massively potentiates (>100-fold) proton-evoked activation of ASIC2a ch …

Novel ideas of local anaesthetic actions on various ion channels to ameliorate postoperative pain.

This review considers the ion channels that underlie transduction of nociceptive energies in the periphery, that are involved in impulse initiation and propagation in peripheral sensory neurones, and that participate in pre- and post-synaptic actions in the spinal cord dorsal horn, in light of their susceptibility to local anaesthetics. Although there are results from experiments on isolated cells and tissues ex vivo that support the hypothesized actions, their extrapolation to actions in vivo and the consequences for peri- and postoperative pain control are largely speculative.

Mechanosensitive ion channels and the peptide inhibitor GsMTx-4: history, properties, mechanisms and pharmacology.

Sensing the energy from mechanical inputs is ubiquitous--and perhaps the oldest form of biological energy transduction. However, the tools available to probe the mechanisms of transduction are far fewer than for the chemical and electric field sensitive transducers. The one pharmacological tool available for mechansensitive ion channels (MSCs) is a peptide (GsMTx-4) isolated from venom of the tarantula, Grammostola spatulata, that blocks cationic MSCs found in non-specialized eukaryotic tissues. In this review, we summarize the current knowledge of GsMTx-4, and discuss the inevitable crosstalk…

Effects of iron-chelators on ion-channels and HIF-1alpha in the carotid body.

Acute hypoxia instantaneously increases the chemosensory discharge from the carotid body, increasing ventilation mostly by inhibiting the oxygen sensitive ion channels and exciting the mitochondrial functions in the glomus cells. On the other hand, Fe2+-chelation mimics hypoxia by inhibiting the prolyl hydroxylases and the degradation of HIF-1alpha in non-excitable cells. Whether Fe2+-chelation can inhibit the ion channels giving rise to the sensory responses in excitable cells was the question. We characterized the responses to Fe2+-chelators on excitable glomus cells of the rat, and found th…

Mutations that affect ion channels change the sensitivity of Drosophila melanogaster to volatile anesthetics.

We have quantitated the response of D. melanogaster to general anesthetics with a device, the inebriometer, that assays the fly's geotactic and postural behavior. When alleles of several loci that encode or regulate subunits of ion channels were compared with control stocks, several ion channel mutants clearly increased the anesthetic sensitivity of Drosophila. The effects were specific in that: a) for several alleles, genetic tests indicated that the anesthesia phenotype was due to the ion channel mutation and not to extraneous genetic differences between the stocks; b) a given ion channel mu…

Effect of anthrax toxin's lethal factor on ion channels formed by the protective antigen.

Protective antigen (PA), a component of anthrax toxin, mediates translocation of the toxin's lethal and edema factors (LF and EF, respectively) to the cytoplasm, via a pathway involving their release from an acidic intracellular compartment. PA63, a 63-kDa proteolytic fragment of PA, can be induced to form ionconductive channels in the plasma membrane of mammalian cells by acidification of the medium. These channels are believed to be comprised of dodecyl sulfate-resistant oligomers (heptameric rings) of PA63 seen by electron microscopy of the purified protein. Here we report that the PA63-med

GABA- and glutamate-gated ion channels as molecular sites of alcohol and anesthetic action.

The evidence presented above indicates that GABA- and glutamate-activated ion channels are molecular sites of alcohol and anesthetic action. In view of the important role that these channels play in CNS excitability, it seems likely that the actions of alcohol and anesthetics on these channels contribute significantly to the behavioral effects of these agents. Although the behavioral effects of alcohol and anesthetics may well result from a combination of actions on different ion channels and other molecular sites in the CNS, it is of interest to consider whether the actions of these agents on…

Induction of ion-permeable channels by the venom of the fanged bloodworm Glycera dibranchiata.

Venom from the poison glands of the polychaete annelid Glycera convoluta has been reported to dramatically increase the frequency of miniature end-plate potentials at the frog and crayfish neuromuscular junctions, without causing detectable ultrastructural changes. We report here that addition of venom from the related annelid Glycera dibranchiata to one side of a lipid bilayer results in the formation of ion-permeable channels in the membrane. The channel forming activity was found in the void volume of a Sephadex G-25 column (mol. wt. greater than 5000). The conductance of a single channel i

Effect of bradykinin and histamine on the membrane voltage, ion conductances and ion channels of human glomerular epithelial cells (hGEC) in culture.

The effects of bradykinin (BK) and histamine (Hist) on the membrane voltage (Vm), ion conductances and ion channels of cultured human glomerular epithelial cells (hGEC) were examined with the nystatin patch clamp technique. Cells were studied between passage 3 and 20 in a bath rinsed with Ringer-like solution at 37 degrees C. The mean value of Vm was -41 +/- 0.5 mV (n = 189). BK (10(-6) mol/l, n = 29) and Hist (10(-5) mol/l, n = 55) induced a rapid transient hyperpolarization by 15 +/- 1 mV and 18 +/- 1 mV, respectively. The hyperpolarization was followed by a long lasting depolarization by 6 …

Purinergic ion channel-type P2X7 receptor regulates SK3 channel to promote the progression of gastric cancer.

The purpose of this study was to investigate the effect of the purinergic ion channel-type P2X7 receptor (P2X7R) on the progression of gastric cancer (GC). Therefore, in this study, in vivo and in vitro experiments were performed to investigate the molecular mechanism of P2X7R on the progression of GC. The results showed that adenosine triphosphate (ATP) and benzoyl ATP (BzATP) activate P2X7R and increased the intracellular calcium concentration of 7901 and 803 cells, and enhanced the migration abilities of GC cells. While P2X7R antagonists (A438079 and AZD9056) decreased the ATP-induced calci

Transmembrane channel-like 4 and 5 proteins at microvillar tips are potential ion channels and lipid scramblases.

Microvilli-membrane bound actin protrusions on the surface of epithelial cells-are sites of critical processes including absorption, secretion, and adhesion. Increasing evidence suggests microvilli are mechanosensitive, but underlying molecules and mechanisms remain unknown. Here, we localize transmembrane channel-like proteins 4 and 5 (TMC4 and 5) and calcium and integrin binding protein 3 (CIB3) to microvillar tips in intestinal epithelial cells, near glycocalyx insertion sites. We find that TMC5 colocalizes with CIB3 in cultured cells and that a TMC5 fragment forms a complex with CIB3 in vi

Acid-sensing ion channel 1a activates IKCa/SKCa channels and contributes to endothelium-dependent dilation.

Acid-sensing ion channel 1a (ASIC1a) belongs to a novel family of proton-gated cation channels that are permeable to both Na+ and Ca2+. ASIC1a is expressed in vascular smooth muscle and endothelial cells in a variety of vascular beds, yet little is known regarding the potential impact of ASIC1a to regulate local vascular reactivity. Our previous studies in rat mesenteric arteries suggest ASIC1a does not contribute to agonist-induced vasoconstriction but may mediate a vasodilatory response. The objective of the current study is to determine the role of ASIC1a in systemic vasodilatory responses …

Tailoring Fluorinated Chain Positioning in Anion Exchange Membranes for Boosted Ion and Water Transport Channels.

Anion exchange membranes (AEMs) have advanced anion exchange membrane water electrolyzers (AEMWEs) by achieving high ionic conductivity and controlled swelling, yet balancing hydration-enhanced electrolytic performance and swelling-induced dimensional instability remains a key challenge for scalable application. In this work, we advance beyond conventional cationic grafting modification strategies via the rational design of fluorinated alkyl side chains on hydrophobic polymer backbones, thereby enhancing amphiphilic self-assembly. Such structural engineering facilitates the formation of interc

Concluding remarks: Molecular and ion flows through angstrom-scale channels: bridging theory, simulation, and experiment.

These concluding remarks report on a Faraday Discussion in which angstrom-scale confinement emerged not as a geometric extension of bulk behaviour but as a qualitatively distinct thermodynamic and kinetic regime, with emerging evidence that some of its anomalies may have quantum origins. Opened by the Spiers Lecture of Prof. Lydéric Bocquet, the Discussion covered four sessions: structure and dynamics of confined molecules; stimuli-responsive transport; molecular and ion sieving; and iontronics and emergent neuromorphic effects. Machine-learning molecular dynamics at first-principles accuracy

Modulatory Effects of Bioactive Phytoconstituents on the Amplitude and Gating Properties of Membrane Ion Channels.

This review provides a comprehensive overview of the modulatory actions of plant-derived constituents on membrane ion channels in various cell types. Among their diverse bioactivities, ion channel regulation-governing membrane excitability, signal transduction, and cellular homeostasis-has emerged as a critical mechanistic basis for their pharmacological effects. Twenty-four representative phytoconstituents are discussed and classified into five major categories based on their structural features: alkaloids, terpenoids, lignans and acetogenins, polyphenols, and other aromatic and conjugated co…

PLASTID ENVELOPE ION CHANNELS (PEC1/2) link Ca2+ and jasmonic acid signaling in plant cells.

Plants constantly encounter adverse environmental interactions. One organelle is particularly specialized in stress signaling and phytohormone synthesis: the plastid. Calcium (Ca2+), a key second messenger, is known to intersect with cellular phytohormone signaling networks. While cytosolic Ca2+ dynamics have been studied extensively, the physiological relevance of stromal Ca2+ transients and the identity of channels mediating rapid Ca2+ flux into plastids remain largely unexplored. In this study, we provide evidence for PLASTID ENVELOPE ION CHANNELS (PECs) as long-sought mediators of fast-act…

Circadian dysregulation of voltage-gated ion channels orchestrates cross-organ electrophysiological remodeling and therapeutic timing.

Disruption of intrinsic circadian rhythms alters cellular electrophysiology, promoting disease across multiple organ systems. We show that dysregulation of core clock genes (BMAL1, CLOCK, PER, CRY) remodels voltage-gated ion-channel (VGIC) expression, leading to sustained depolarization, impaired repolarization, and oxidative stress, unifying malignant and degenerative phenotypes. Integrative transcriptomic analysis across multi-organ datasets (GSE130408, GSE62944, TCGA-BRCA, TCGA-LAML, GSE56473) revealed consistent signatures: upregulation of sodium channels (SCN family), suppression of potas

Piezo1 Ion Channels Regulate the Formation and Spreading of Human Endometrial Mesenchymal Stem Cell Spheroids.

Mesenchymal stem cells obtained from desquamated endometrium (eMSCs) are considered as reliable and promising objects for stem cell-based therapy. eMSCs aggregated into three-dimensional (3D) spheroids demonstrate greater efficiency compared to monolayer 2D eMSCs. However, molecular processes and specific mechanisms regulating the effectiveness of spheroids remain unknown. Regulation of a number of physiological reactions in MSCs is associated with the functioning of Ca2+-permeable mechanosensitive Piezo1 channels. In our previous study, we showed that selective Piezo1 activation by its select

The crucial role of potassium ion channels in diabetes mellitus and its complications: A review.

Potassium ion channel (K+ channel) is a crucial transmembrane protein found on cell membranes that plays a pivotal role in regulating various physiological processes such as cell membrane potential, action potential formation, and cellular excitability. Diabetes, a chronic metabolic disorder characterized by elevated blood glucose levels, can cause abnormal changes in the sensitivity and functioning of K+ channels over time. This can lead to an increase in intracellular K+ and Ca2+, disrupting normal cellular function and metabolism and resulting in a range of physiological and metabolic issue
