Neurodegenerative diseases are becoming prevalent as the population ages. Geniposide could inhibit oxidative stress, reduce apoptosis, protect neuron, and has been used for therapy of the neurodegenerative diseases. The bioavailability of geniposide by nasal route is greater than that by oral administration. However, mucociliary clearance is a rate-limiting factor for nasal route administration. The objective of this study was to develop and evaluate a mucoadhesive, thermoreversible in situ nasal gel of geniposide. The poloxamers (P407, P188) and the hydroxypropyl methylcellulose were used as

Human immunity involves both innate and adaptive defence mechanisms, with inflammation playing a central role in responding to cellular injury, pathogenic infections, and allergic stimuli. Reactive oxygen species (ROS) are closely associated with the onset and progression of inflammation. While moderate ROS levels function as crucial signalling molecules, excessive ROS can damage cellular components. This study aimed to evaluate the anti-inflammatory and antioxidant potential of plant-derived bioactive compounds including chlorogenic acid, oleuropein, tomatine, and tyrosol using human monocyti

It is known that the Mediterranean diet is effective in reducing the risk of several chronic diseases, including cancer. A critical component of the Mediterranean diet is olive oil, and the relationship between olive oil consumption and the reduced risk of cancer has been established. Oleuropein (OL) is the most prominent polyphenol component of olive fruits and leaves. This compound has been shown to have potent properties in various types of cancers, including breast cancer. In the present study, the molecular mechanism of OL was examined in two racially different triple-negative breast canc

Amyloid-ß (Aß) fragments, oligomeric Aß aggregates, and pyroglutamylated-Aß peptides, as well as epigenetic mechanisms and autophagy dysfunction all appear to contribute in various ways to Alzheimer's disease progression. We previously showed that dietary supplementation of oleuropein aglycone, a natural phenol abundant in the extra virgin olive oil, can be protective by reducing Aß42 deposits in the brain of young and middle-aged TgCRND8 mice. Here, we extended our study to aged TgCRND8 mice showing increased pE3-Aß in the brain deposits. We report that oleuropein aglycone is active against g

Recent advances in nerve repair technology have focused on finding more biocompatible, non-toxic materials to imitate natural peripheral nerve components. In this study, casein protein cross-linked with naturally occurring genipin (genipin-cross-linked casein (GCC)) was used for the first time to make a biodegradable conduit for peripheral nerve repair. The GCC conduit was dark blue in appearance with a concentric and round lumen. Water uptake, contact angle and mechanical tests indicated that the conduit had a high stability in water and did not collapse and cramped with a sufficiently high l

Catalpol, an iridoid glucoside, separated from the root of Rehmannia glutinosa Libosch, has been known to show various neuroprotective effects. In humans and rodents, MPTP is well known to produce clinical, biochemical and neurochemical changes similar to those which occur in Parkinson's disease (PD). Furthermore, the accumulated evidence suggests that MPP(+), conversed by monoamine oxidase type B (MAO-B) in astrocytes principally, is the active metabolite of MPTP and the major cause to PD associated with mitochondrial dysfunction. In this study, we treated mesencephalic neuron-astrocyte and a

Loss of β-cell function and β-cell death is the key feature of type 2 diabetes mellitus (T2DM). One hypothesis for the mechanism of this feature is amyloid formation by the human islet amyloid polypeptide (hIAPP). Despite the global prevalence of T2DM, there are no therapeutic strategies for the treatment of or prevention of amylin amyloidosis. Clinical trials and population studies indicate the healthy virtues of the Mediterranean diet, especially the extra virgin olive oil (EVOO) found in this diet. This oil is enriched in phenolic compounds shown to be effective against several aging and li

This study addresses the need for effective natural inhibitors of enzymatic browning in fresh-cut produce by exploring plant-derived compounds with activity against apple polyphenol oxidase (PPO). Extracts from 26 wild plant species were screened using thin-layer chromatography (TLC)-coupled bioautographic assays specifically adapted to apple PPO. Among these, D. fullonum showed clear inhibitory activity and was selected for further investigation. Extracts from different plant organs revealed that PPO inhibition was localized in the leaves, while stems and flower receptacles were inactive. Bio

Galium odoratum (L.) Scop., commonly known as sweet woodruff, is a perennial European woodland herb traditionally used as a medicinal and aromatic plant. Its characteristic sweet, hay-like scent is mainly linked to coumarin, which is formed from glycosidically bound melilotoside precursors during wilting and drying. However, the phytochemistry of G. odoratum extends beyond coumarin. The species also contains iridoid glycosides, coumarin-related melilotosides, caffeoylquinic acids, flavonoids, and volatile constituents. This review summarizes current knowledge on traditional use, phytochemical

Loganic acid is an iridoid compound extracted from Gentianaceae plant Gentiana macrophylla Pall. It can effectively inhibit inflammation and tumor migration and has antioxidant activity. In this paper, we establish a simple, fast, sensitive and validated LC-MS method with the purpose of quantification of loganic acid in rat plasma with gliclazide as an internal standard (IS). Methanol was used to precipitate the protein in the plasma sample, and a C18 column (2.1 × 50 mm, 1.7 μm) was used for the separation of the target compound. Meanwhile, 0.1% formic acid water-methanol was employed as the

Autosomal dominant polycystic kidney disease (ADPKD) is characterized by progressive enlargement of kidney cysts, leading to chronic kidney disease. Since the available treatment for ADPKD is limited, there is emerging interest for natural compounds as potential therapeutic candidates. The aim of our study was to investigate whether an olive leaf extract may be able to counteract the cyst growth in an in vitro model of ADPKD. We treated WT9-12 cells with an olive leaf extract (OLE). In monolayer culture we evaluated cell viability by the MTT assay, protein expression by western-blot analysis a

Olive leaves are an abundant agro-industrial by-product rich in phenolic secoiridoids and a constant source of bioactive compounds. An ultrasound-assisted extraction of ambient air-dried Olea europaea leaves from the "Lianolia" variety with water at room temperature followed by ethyl acetate partitioning, afforded an oleacein-rich fraction. HPLC-DAD identified oleacein as the major constituent, while LC-HRMS (Orbitrap) analysis revealed a prominent oleacein-like metabolite bearing one additional oxygen atom among the eluting peaks. Guided fractionation by MPLC and preparative HPLC led to the i…

Fractionation is key for enriching bioactive compounds in traditional Chinese medicine, yet systematic studies on chemical differences across polarity-based fractions are limited. This study systematically compared the chemical profiles of four polarity fractions from a Gentiana scabra Bunge ethanol-water extract to elucidate the distribution patterns of its major active constituents. An integrated approach of untargeted LC-MS and quantitative HPLC was employed. The contents of gentiopicroside, isoorientin, and gentisin generally increased with ethanol concentration, with gentisin showing a mo
