Possible aetiological factors in leukaemias in Africa.

The hypothesis that the true incidence of c-ALL is relatively uniform throughout the world is not supported by experience in tropical Africa, where ALL is uncommon under five years of age. A high rate of spontaneous somatic mutation in pre-B cells may initiate the development of c-ALL, but its progress could be determined by (i) a leukaemogenic agent causing a second genetic event, (ii) the effects of intense antigenic barrage, either stimulating or suppressing pre-B-cell mitosis, or (iii) genetic determinants. Epidemiological patterns in populations of low, intermediate and high socio-economi

Novel insights into the pathogenesis of the Graffi murine leukemia retrovirus.

The Graffi murine leukemia virus (MuLV) was isolated in 1954 by Arnold Graffi, who characterized it as a myeloid leukemia-inducing retrovirus. He and his team, however, soon observed the intriguing phenomenon of hematological diversification, which corresponded to a decrease of myeloid leukemias and an increase of other types of leukemias. Recently, we derived two different molecular clones corresponding to ecotropic nondefective genomes that were named GV-1.2 and GV-1.4. The induced leukemias were classified as myeloid based on morphological analysis of blood smears. In this study, we further

Minimally differentiated acute myelogenous leukemia (AML-M0) granulocytic sarcoma presenting in the oral cavity.

Acute myelogenous leukemia with minimal differentiation (AML-M0) is a rare subtype of acute leukemia in which blasts fail to show morphologic differentiation and conventional cytochemical stains and myeloid markers are negative. Acute myelogenous leukemia (AML) presents primarily with peripheral blood and/or bone marrow involvement. Presentation in extramedullary sites, including the head and neck region, is not uncommon. Acute myelomonocytic leukemia (AML-M4) and acute monocytic leukemia (AML-M5) have had the highest incidence of associated oral infiltrates. We report a case of a 58-year-old

Polygenic Risk, Agent Orange Exposure, and Lymphoid Neoplasms in the Veterans Affairs Million Veteran Program.

IMPORTANCE: Agent Orange (AO), a toxic herbicide mixture contaminated with carcinogenic dioxin, was widely used during the Vietnam War. Although epidemiologic evidence suggests AO exposure is associated with increased lymphoid malignant neoplasm risk, the details of this association and its potential interaction with genetic factors remain unclear. OBJECTIVE: To evaluate (1) whether AO exposure was associated with increased risk of lymphoid malignant neoplasms, (2) associations of polygenic risk scores (PRSs) in specific lymphoid malignant neoplasms, and (3) whether there was a genetic compon

Hypermethylation of the imprinted NNAT locus occurs frequently in pediatric acute leukemia.

Recent studies have demonstrated imprinting of the human neuronatin (NNAT) gene. NNAT maps to 20q11.2-q12, a region exhibiting loss of heterozygosity in acute myeloid leukemia and myelodysplastic/myeloproliferative disease. To investigate possible epigenetic dysregulation of genes in this region relevant to leukemogenesis, we analyzed methylation of the NNAT gene in normal tissues and in leukemias. We found a differential methylation pattern, typical of imprinted genes, at sites in the CpG island containing NNAT exon 1 in normal pituitary, peripheral blood cells and bone marrow-derived CD34-po

5q deletion in childhood T-acute lymphoblastic leukemia at diagnosis: a case report.

BACKGROUND: We present the case of a 6-year-old Moroccan male patient of Berber ethnic origin, diagnosed with T-cell acute lymphoblastic leukemia, who exhibited a deletion of the 5q region. CASE PRESENTATION: The patient initially presented with classic symptoms of T-cell acute lymphoblastic leukemia, including bone pain, hepatosplenomegaly, and lymphadenopathy. Laboratory tests revealed anemia, hyperleukocytosis, and a high percentage of lymphoid blasts in both the blood and bone marrow. Immunophenotyping results confirmed that these blasts were of T-cell origin. Cytogenetic analysis identif

Frequent microsatellite instability and BAX mutations in T cell acute lymphoblastic leukemia cell lines.

Allelic status of the BAT26 and BAT25 loci was examined in 117 leukemia/lymphoma cell lines consisting of 44 B-lymphoid lineage cell lines, 30 T-lymphoid cell lines and 43 myeloid cell lines to define the lineage specificity of microsatellite instability (MSI) in hematological malignancies. Seventeen (15%) cell lines were defined as having MSI. The incidence of MSI was significantly (P < 0.01) higher in cell lines of lymphoid lineage (15/74; 20%) than in those of myeloid lineage (2/43; 5%). In the cell lines of lymphoid lineage, the incidence of MSI in T cell acute lymphoblastic leukemia (T-AL

Analysis of peripheral blood normal and malignant cells with the novel murine monoclonal antibody UN2.

The monoclonal antibody (mAb) UN2 was generated upon immunization of a Balb/c mouse with human thymocytes. mAb UN2 recognized an antigen expressed by a subpopulation of human thymocytes and peripheral blood lymphocytes. In thymus, mAb UN2 recognized cortical cells; its expression was higher on CD3bright than on CD3dim thymocytes. This antigen was also detected on peripheral blood granulocytes, monocytes, platelets and on cell lines MOLT4, U937 and KG1. mAb UN2 was submitted to the 5th International Workshop and Conference on Human Leukocyte Differentiation Antigens, Boston, MA, 1993, and was a

T-cell prolymphocytic leukemia (T-PLL) with overlapping cytomorphological features with T-CLL and T-ALL: a case initially diagnosed by fine-needle aspiration cytology and immunocytochemistry.

T-cell prolymphocytic leukemia (T-PLL) is a very unusual form of chronic lymphoproliferative disorder, which has rarely been diagnosed by fine needle aspiration (FNA) cytology. We report one such case with some overlapping cytomorphological features with chronic lymphocytic leukemia and acute lymphoblastic leukemia. A 91-year-old man presented with generalized lymphadenopathy, pleural effusion, ascites, and an ulcerated growth in rectum. FNA smears from the left cervical lymph node showed a monotonous population of small lymphoid cells having small but distinct nucleoli that was initially diag

The morphology, immunohistochemistry, and incidence of hematopoietic neoplasms in mice and rats.

Hematopoietic neoplasms in the rodent may be classified into lymphoid or nonlymphoid neoplasms. Lymphoid neoplasms include the following morphologic types: follicular center cell, lymphoblast (lymphocytic), immunoblast, plasma cell, and large granular lymphocyte (LGL). Nonlymphoid hematopoietic neoplasms include histiocytic sarcoma, granulocytic leukemia, erythroid leukemia, and mast cell tumors. Most types of hematopoietic neoplasms, exclusive of LGL lymphoma (leukemia), are more common in mice than in rats. Specific strains of mice have a hematopoietic tumor incidence of more than 50% in age

[Evaluation of the proliferation and line of lymphoid cells in various lymphoproliferative diseases using a double immunocytochemical method].

The increased range of the diagnostic criteria and the introduction of the classification scheme "Practical Formulation of Non-Hodgkin's Lymphomas for Clinical Application" made the authors to study proliferative potential of lymphoid cells with identification of the cell line affiliation. The investigation was carried out with the use of the double immunocytochemical test employing anti-nuclear (Ki-67) and line-specific monoclonal antibodies on peripheral blood lymphocytes, those of lymph nodes and bone marrow obtained from 40 patients with lymphoproliferative diseases and on the cells of lym
