Mass population screening for celiac disease in children: the experience in Republic of San Marino from 1993 to 2009.

BACKGROUND: Prevalence of celiac disease in developed countries is assessed about 1:100-1:150. The real prevalence is unknown because mass screenings are expensive and difficult to organize. Moreover celiac disease can affect people at every age and studies on asymptomatic subjects at different ages are not comparable. In this study we wanted to know the real prevalence of celiac disease in children in the Republic of San Marino. We also analysed concordance of different tests used and costs of mass screening. METHODS: The study started in 1993. From 1993 to 1997 children aged 6, 10 and 14 we…

Highly sensitive capillary electrophoresis-mass spectrometry for rapid screening and accurate quantitation of drugs of abuse in urine.

The combination of capillary electrophoresis (CE) and mass spectrometry (MS) is particularly well adapted to bioanalysis due to its high separation efficiency, selectivity, and sensitivity; its short analytical time; and its low solvent and sample consumption. For clinical and forensic toxicology, a two-step analysis is usually performed: first, a screening step for compound identification, and second, confirmation and/or accurate quantitation in cases of presumed positive results. In this study, a fast and sensitive CE-MS workflow was developed for the screening and quantitation of drugs of a

Tools for mass screening of G6PD deficiency: validation of the WST8/1-methoxy-PMS enzymatic assay in Uganda.

BACKGROUND: The distribution of the enzymopathy glucose-6-phosphate dehydrogenase (G6PD) deficiency is linked to areas of high malaria endemicity due to its association with protection from disease. G6PD deficiency is also identified as the cause of severe haemolysis following administration of the anti-malarial drug primaquine and further use of this drug will likely require identification of G6PD deficiency on a population level. Current conventional methods for G6PD screening have various disadvantages for field use. METHODS: The WST8/1-methoxy PMS method, recently adapted for field use, w

Computer-aided detection of masses in full-field digital mammography using screen-film mammograms for training.

It would be of great value when available databases of screen-film mammography (SFM) images can be used to train full-field digital mammography (FFDM) computer-aided detection (CAD) systems, as compilation of new databases is costly. In this paper, we investigate this possibility. Firstly, we develop a method that converts an FFDM image into an SFM-like representation. In this conversion method, we establish a relation between exposure and optical density by simulation of an automatic exposure control unit. Secondly, we investigate the effects of using the SFM images as training samples compar

Development of a filter paper method potentially applicable to mass and high-risk urinary screenings for Fabry disease.

Fabry disease is an X-linked lysosomal storage disorder of glycosphingolipid catabolism resulting from a deficiency of the enzyme alpha-galactosidase A, and leading to the progressive accumulation of one biomarker, globotriaosylceramide (Gb(3)), predominantly elevated in the urine of these patients. We have developed a technique for the analysis of total Gb(3) in urine samples collected on filter paper, using liquid chromatography-tandem mass spectrometry (LC-MS/MS) with a triple quadrupole instrument. Existing Gb(3) techniques being both time- and labour-intensive, this filter paper method el

Evaluation of diagnostic ultrasound as a mass screening technique for the detection of hydatid cysts in the liver and lung of sheep and goats.

Ultrasound examination of the liver and lung followed by post-mortem examination was performed in 16 sheep and 284 goats. Thirty-one (10.3%) were positive for hydatid cysts on ultrasound examination and 46 (15.3%) were positive on post-mortem examination. Twenty-one positive on post-mortem examination were falsely identified as negative on ultrasound examination. Of the 254 animals negative on post-mortem examination, six (2.4%) were falsely identified as positive on ultrasound examination. The sensitivity and specificity of ultrasound examination for detecting hydatid cysts in sheep and goats

A silica gel plate-based qualitative assay for acetylcholinesterase activity: a mass method to screen for potential inhibitors.

A procedure for the qualitative assessment of inhibitory activity towards acetylcholinesterase for a given compound is described. Solutions of the compounds of interest are spotted on silica gel TLC plates in a matrix pattern. The silica gel plate is sprayed with a solution of acetylthiocholine iodide and 5,5-dithiobis(2-nitrobenzoic acid) followed by a solution of acetylcholinesterase. The enzyme reaction produces a yellow background color with inhibitor compounds exposed as white zones where color has failed to develop. The results for a test set of compounds were compared to those obtained

Mass spectrometry‑based chemoproteomics for modern drug screening: A critical review and the 3M framework (Match-Map-Mine).

Modern drug screening leans heavily on mass spectrometry-based chemoproteomics. However, widespread adoption has not cured the field's fundamental fragmentation: component identification and target deconvolution, not to mention safety evaluation, remain disconnected disciplines, each operating within its own literature, expert circles, and technical routines. This separation carries a real cost, as discoveries rarely travel from bench to clinic. We address this gap with the Match-Map-Mine (3 M) framework, which folds these stages into a single workflow that sharpens with every cycle. We then e

LCMS-Net: Deep Learning for Raw High Resolution Mass Spectrometry Data Applied to Forensic Cause-of-Death Screening.

Current preprocessing workflows for untargeted metabolomics using liquid chromatography-high resolution mass spectrometry (LC-HRMS) are time-consuming and require significant domain knowledge. Furthermore, they lack reproducibility or may fail to detect some metabolites entirely. We introduce LCMS-Net, an end-to-end deep learning model for the analysis of LC-HRMS data, to address these challenges. LCMS-Net mitigates the need for manual data preprocessing by operating directly on the raw LC-HRMS data and explicitly modeling its spatial properties. The effectiveness of this fully automated workf

Leveraging mass spectrometry fragmentation patterns for accelerated screening and structural elucidation of new etomidate analogues in suspect samples.

Etomidate analogues are structural modifiers of etomidate, which also have psychoactive effects, and abuse of them is gradually intensifying. Due to the existence of multiple modification sites in the structures of etomidate, the rate of derivatization is extremely rapid, which brings more difficulties for rapid analysis and accurate identification of these substances in forensic laboratories. Thus, establishing a non-targeted screening and structural identification method can effectively solve this problem. In this study, the structural evolution rules of etomidate analogues were investigated

Non-target screening of toxic contaminants in secondary effluents (from several sources) using SPE-LC-Orbitrap mass spectrometry.

Toxic contaminants are widely present in aquatic environments; however, many contaminants cannot be eliminated during wastewater treatment. The identification of these contaminants using traditional targeted methods requires considerable effort, time, and specialized reference standards. In this study, a non-target screening method was developed. Samples were analyzed using ultra-high-performance liquid chromatography-Q Exactive Orbitrap mass spectrometry (UHPLC-Q Exactive Orbitrap MS). Data were processed through the small molecule screening software Compound Discoverer 3.3, and the identifie

Rapid screening of the antithrombotic active components of Agrimonia pilosa Ledeb with thrombin binding affinity ultrafiltration mass spectroscopy.

Traditional Chinese Medicine (TCM) holds significant potential as a natural source of medication, providing effective alternatives for preventing and treating thrombotic diseases. Although research on the anticoagulant activity of Agrimonia Pilosa Ledeb (Rosaceae, A. pilosa) has been paid attention to, the monomer component and its mechanism of action have not been studied deeply. Therefore, this study aimed to screen and identify the antithrombotic active components of A. pilosa by affinity ultrafiltration (AUF) combined with ultra-high performance liquid chromatography-quadrupole time-of-fli

A high-throughput newborn screening approach for SCID, SMA, and SCD combining multiplex qPCR and tandem mass spectrometry.

Early diagnosis of severe combined immunodeficiency (SCID), spinal muscular atrophy (SMA), and sickle cell disease (SCD) improves health outcomes by providing a specific treatment before the onset of symptoms. A high-throughput nucleic acid-based method in newborn screening (NBS) has been shown to be fast and cost-effective in the early detection of these diseases. Screening for SCD has been included in Germany's NBS Program since Fall 2021 and typically requires high-throughput NBS laboratories to adopt analytical platforms that are demanding in terms of instrumentation and personnel. Thus, w
