Abnormalities of the ear associated with exencephaly in mouse fetuses induced by maternal exposure to cadmium.

Exencephaly was induced in mouse fetuses by maternal injection of cadmium chloride (CdCl2) on day 7 of gestation. The heads of exencephalic, nonexencephalic experimental, and control fetuses were embedded in paraffin and sections were stained with hematoxylin and eosin. Compared to those of controls, the ears of the exencephalic fetuses were smaller (microtia) and low set. The meatal plug representing the external auditory canal was thick, variously branched, and often directed inferiorly. Usually, there were just two ossicles. The stapedial artery, facial nerve, and stapedius muscle were hypo

Maternal ethanol consumption and fetal development in the rat: a comparison of ethanol exposure techniques.

Comparison of the polydipsia and liquid diet procedures for ethanol exposure were used in female rats, either before and/or during gestation. Autopsy of the fetuses on day 20 of gestation found a significant weight reduction in the polydipsia-exposed animals that could be attributed to ethanol. No such effect was found in the liquid diet-exposed animals. The data are discussed in relation to the human fetal alcohol syndrome.

Chronic prenatal corticosterone exposure alters postpartum maternal behavior and shapes sex-specific offspring outcomes.

Stress during the prenatal period increases the risk in the next generation for negative health outcomes, including cardio-metabolic dysfunction and psychiatric diseases. To standardize prenatal exposure to stress hormone levels, we administered corticosterone (20 mg/kg s.c.) chronically (C-CORT) to pregnant mouse dams daily from gestational day 12-18. We monitored physiological, neural, and behavioral endpoints in dams and their offspring at adolescence and adulthood. Dams treated with C-CORT displayed increased anxiety-like behavior and altered maternal behavior, including increased time to

Targeting aryl hydrocarbon receptor signaling attenuates hypertension programmed by maternal Di-2-ethylhexylphthalate exposure.

Adverse early-life conditions can predispose offspring to long-term health risks. Phthalate exposure, particularly to di-2-ethylhexylphthalate (DEHP), during pregnancy and lactation has been implicated in programming offspring hypertension via aryl hydrocarbon receptor (AHR) activation, renin-angiotensin system (RAS) dysregulation, nitric oxide (NO) deficiency, and gut microbiota alterations. Using a maternal DEHP exposure rat model, we investigated whether blockade of AHR signaling-directly with the AHR inhibitor CH223191 or indirectly with the indoleamine 2,3-dioxygenase (IDO) inhibitor INCN

Maternal Analgesic Exposure and Fetal Ductal Constriction: A Prospective Cohort Study in Late Pregnancy.

OBJECTIVE: To evaluate the association between maternal analgesic exposure in late pregnancy and fetal ductal and pulmonary hemodynamics using serial echocardiographic assessment, and to explore potential differences between metamizole and acetaminophen. METHODS: In this prospective cohort study, 67 third-trimester pregnancies were evaluated: 47 exposed to analgesics (27 metamizole and 20 acetaminophen) and 20 unexposed controls. Two standardized fetal echocardiograms were performed: during exposure (T1) and after a 5-7 day drug-free interval (T2). Ductal Doppler parameters, including systoli

Maternal Lead Exposure Induces Glandular Alterations in the Salivary Glands of Rat Male Offspring.

Lead (Pb) is a widely distributed environmental pollutant that is highly noxious due to its toxicokinetic characteristics. Pb exposure can damage vital organ systems, including the central nervous and stomatognathic systems. As salivary glands play an important role in oral homeostasis, this study aimed to investigate the effects of maternal Pb exposure during pregnancy and lactation on the salivary glands of rat male offspring. Six 90-day-old pregnant Wistar rats were randomly divided into two groups of three animals each: a control group and a lead acetate group (treated with 50 mg/kg lead a

Association analysis of maternal exposure to air pollution during pregnancy and offspring asthma incidence.

BACKGROUND: Air pollution has a significant negative impact on human health. Pregnant mothers and children are typical susceptible groups, and environmental exposure has a crucial impact on children's health. We established a childhood asthma cohort to analyze the factors influencing the development of asthma in offspring, with a focus on prenatal exposure to air pollutants. The goal was to explore potential early preventive measures to reduce the incidence of childhood asthma. METHODS: This nested case-control study included mothers who were registered and delivered at Lianyungang Maternal a

Maternal exposure factors in pregnancy that affect fetal CHD risk: a case-control study.

AIM: We sought to assess the degree to which environmental risk factors affect CHD prevalence using a case-control study. METHODS: A hospital-based study was conducted by collecting data from outpatients between January 2016 and January 2021, which included 31 CHD cases and 72 controls from eastern China. Risk ratios were estimated using univariate and multivariate logistic regression models and mediating effect analysis. RESULTS: Residential characteristics (usage of cement flooring, odds ratio = 17.04[1.954-148.574], P = 0.01; musty smell, odds ratio = 3.105[1.198-8.051], P = 0.02) and ind

Maternal gut microbiota-derived daidzein prevents osteoporosis in female offspring following prenatal prednisone exposure.

Prenatal exposure to glucocorticoids is linked to long-term health risks in offspring, but the role of maternal gut microbiota in mediating these effects remains unclear. Here, we demonstrate that prenatal prednisone therapy (PPT) in humans and prenatal prednisone exposure (PPE) in rats result in sex-specific long bone dysplasia in offspring, including reduced peak bone mass (PBM) and heightened osteoporosis risk in female offspring. Multi-omics profiling and fecal microbiota transplantation show that PPE alters maternal gut microbiota composition and depletes the microbial metabolite daidzein

Single pulmonary nanopolystyrene exposure in late-stage pregnancy dysregulates maternal and fetal cardiovascular function.

Large-scale production and waste of plastic materials have resulted in widespread environmental contamination by the breakdown product of bulk plastic materials to micro- and nanoplastics (MNPs). The small size of these particles enables their suspension in the air, making pulmonary exposure inevitable. Previous work has demonstrated that xenobiotic pulmonary exposure to nanoparticles during gestation leads to maternal vascular impairments, as well as cardiovascular dysfunction within the fetus. Few studies have assessed the toxicological consequences of maternal nanoplastic (NP) exposure; the

DNA methylation signatures of youth-onset type 2 diabetes and exposure to maternal diabetes.

OBJECTIVE: Youth-onset type 2 diabetes (T2D) is physiologically distinct from adult-onset, but it is not clear how the two diseases differ at a molecular level. In utero exposure to maternal type 2 diabetes (T2D) is known to be a specific risk factor for youth-onset T2D. DNA methylation (DNAm) changes associated with T2D but which differ between youth- and adult-onset might delineate the impacts of T2D development at different ages and could also determine the contribution of exposure to in utero diabetes. METHODS: We performed an epigenome-wide analysis of DNAm on whole blood from 218 youth

Impacts of Maternal Preeclampsia Exposure on Offspring Neuronal Development: Recent Insights and Interventional Approaches.

Preeclampsia, a hypertensive disorder during pregnancy, frequently correlates with adverse neurological outcomes in offspring, including cognitive impairments, autism spectrum disorder, depressive disorder, attention deficit hyperactivity disorder, and cerebral palsy. Despite these known consequences, the understanding of neuronal damage in the offspring of preeclamptic mothers remains insufficient. Here, we review the neuronal abnormalities resulting from maternal preeclampsia exposure, which include disrupted neurogenesis, loss of neuronal cell integrity, accumulation of cellular debris, dec

Maternal exposure to dietary uranium causes oxidative stress and thyroid disruption in zebrafish offspring.

The contamination of uranium in aquatic ecosystems has raised growing global concern. However, the understanding of its chronic effects on aquatic organisms is limited, particularly with regards to transgenerational toxicity. In this study, we evaluated the maternal transfer risk of uranium using zebrafish. Sexually mature female zebrafish were exposed to 2 and 20 ng/g of uranium-spiked food for 28 days. The induced bioconcentration, thyroid disruption, and oxidative stress in both the adults (F0) and their embryos (F1) were further investigated. Element analysis showed that uranium was presen

Can maternal exposure to tamoxifen compromise sperm and behavioural parameters of male rat offspring?

Tamoxifen, a selective non-steroidal estrogen receptor modulator, is the standard adjuvant endocrine treatment for breast cancer. Since information on the risk of using tamoxifen during pregnancy is still scarce, this study evaluated whether the in utero and lactational treatment with this drug could compromise reproductive and behavioural parameters in male offspring. Pregnant Wistar rats were exposed to three doses of tamoxifen (0.12; 0.6; 3 μg/kg), by gavage, from gestational day 15 to lactational day 20. Tamoxifen exposure did not alter the anogenital distance in the male offspring; howeve

Maternal high-fat diet during pregnancy with concurrent phthalate exposure leads to abnormal placentation.

Di(2-ethylhexyl) phthalate (DEHP) is a synthetic chemical commonly used for its plasticizing capabilities. Because of the extensive production and use of DEHP, humans are exposed to this chemical daily. Diet is a significant exposure pathway and fatty food contain the highest level of phthalates. The impact on pregnancy following DEHP exposure and the associated interaction of high fat (HF) diet remains unknown. Here we report that exposure of pregnant mice to an environmentally relevant level of DEHP did not affect pregnancy. In contrast, mice fed a HF diet during gestation and exposed to the
