Biosynthesis of pyomelanin from methanol with engineered Komagataella phaffii and its characterizations.

Pyomelanin has been extensively applied in various fields. However, the yield of pyomelanin isolated from natural producers is low. Engineering microbial biosynthesis is considered a sustainable and economically feasible alternative. We utilize engineered Komagataella phaffii to increase the yield of homogentisic acid by 66-fold by balancing three different biosynthetic modules and further synthesizing pyomelanin by oxidative polymerization. During this process, we establish a pyomelanin color screening system and apply this system to perform directed evolution of 3-deoxy-D-arabinoheptulosonat

Visible-light-driven conversion of methane to methanol mediated by hydroxyl radical.

Selective oxidation of methane (CH4) to value-added liquid methanol (CH3OH) drives the synthesis of chemicals and energy sustainability. It is extremely challenging to mimic in artificial reaction systems due to the inertness of CH4 molecule and overoxidation to byproducts. In this work, we developed an effective and noble-metal-free iron-based catalyst (UNFe) that delivers exceptional performance of methane oxidation to methanol with visible light at room temperature. Specifically, the hydroxyl radical (OH) generated through FeIII/FeII cycling in UNFe catalyst is essential for promoting activ…

Medicolegal evaluation of methanol poisoning from illicit alcohol: a five-case series.

Methanol poisoning resulting from illicit alcohol consumption constitutes a major public health problem and may lead to severe metabolic acidosis, visual disturbances, and significant medicolegal implications. This case series describes five male patients who ingested illicit alcohol and were managed at a center without access to serum methanol measurement. All patients presented with high-anion-gap metabolic acidosis and visual symptoms, allowing a clinical diagnosis of methanol poisoning despite the absence of toxicological confirmation. Emergency hemodialysis was performed in all cases, and…

A Monolithic Artificial Leaf for Solar Methanol Production from CO2 and H2O.

Methanol, an important liquid fuel and chemical feedstock, has yet to be produced using solar energy, H2O, and CO2 as sole inputs in a standalone device. This study directly addresses this longstanding challenge through presenting the first demonstration of unbiased solar methanol production from CO2 and H2O with a monolithic artificial leaf design, surpassing the previous best energy efficiency in solar alcohol production by at least 1 order of magnitude. We first develop a new generation of photocathodes based on Si micropillar arrays and a cobalt tetraaminophthalocyanine molecular catalyst.…

Photocatalytic CO2 reduction to methanol: from mechanistic insights to reactor design integration.

The conversion of CO2 into high-value fuels, particularly methanol, represents a promising pathway for closing the carbon cycle and storing intermittent solar energy. While significant progress has been made in photocatalyst development, the transition from laboratory scale to industrial application remains hindered by low quantum efficiency and mass transfer limitations. This review systematically examines the state-of-the-art in photocatalytic CO2 reduction, with a specific focus on methanol production. Unlike previous reviews that isolate catalyst materials from reactor engineering, this pa…

Silanol Networks Control Methanol Reactivity in Nano- and Micron-sized Silicalite-1.

The catalytic activity of silanol groups in zeolites has long been overlooked because of their much lower acidity compared to Brønsted acid sites. Here, we demonstrate their non-negligible catalytic role in methanol conversion using pure silica MFI zeolite (Silicalite-1). Two silicalite-1 samples with different crystal sizes, micron-sized (Sil1_micro) and nano-sized (Sil1_nano), were synthesized and investigated by in situ and operando FTIR spectroscopy using probe molecules (carbon monoxide, pyridine, and methanol). multivariate-curve regression by alternating least squares was applied to elu…

Theoretical Study into Water Promoted CO2 Hydrogenation to Methanol over PdMo Alloy.

CO2 hydrogenation to methanol is generally considered one of the most promising approaches in CO2 utilization. However, how to design highly efficient catalysts still remains a significant challenge. In this paper, density functional theory as well as microkinetic modeling is employed to systematically investigate the reaction mechanism and catalytic performance of methanol formation over two surface models, hcp-PdMo, i.e., st_PdMo and fl_PdMo. On the basis of our calculation results, the adsorption and activation behavior of CO2 and H2 are closely related to the catalyst surface they are bind…

Adaptive laboratory evolution enhances methanol-driven CO2 fixation in Sporomusa sphaeroides KIAC.

Acetogens are microorganisms that reduce carbon dioxide (CO2) into acetate via the Wood-Ljungdahl pathway, a process that necessitates an external electron donor. While hydrogen gas is the conventional electron donor, its low aqueous solubility and severe gas-liquid mass transfer limitations significantly restrict industrial scalability. In contrast, methanol has emerged as a promising alternative one-carbon liquid feedstock due to its complete miscibility and abundant availability. This study demonstrates the efficacy of utilizing methanol as an electron donor for CO2 reduction in the novel a…

Plug-and-play - enzymatic amino acid production from methanol and carbon dioxide.

Amino acids are the building blocks of proteins and thus among the macronutrients to feed humankind. They have extensive industrial applications as animal feed and food additives like dietary supplements, and flavor enhancers and moreover as chemical precursors. We present the design of a synthetic enzyme cascade system for synthesizing a series of amino acids from methanol. This acts as a case study on how to contribute to the sustainable, renewable energy-based supply of food and feed. The modular nature of the cascade allows for plug-and-play module swapping and fine tuning of enzyme compos…

Methanol Production from Aqueous Formaldehyde Using Ruthenium Imidazolylamine Complexes: Ligand-Tuned Selectivity.

The transformation of aqueous formaldehyde to H2 and methanol is respectively governed by competing dehydrogenation and hydrogenation pathways in water. Herein, we demonstrated the tuning of product selectively, H2 vs methanol from aqueous formaldehyde over molecular arene Ru complexes bearing furan, thiophene, imidazole-based methanol and methylamine ligands under moderate condition. A distinct ligand effect was observed, where furan- and thiophene-based ligands favored formaldehyde dehydrogenation, while imidazolyl methylamine-based ligands favored formaldehyde hydrogenation to methanol unde…

Selective methane-to-methanol conversion in a biochar-mediated photocatalytic biohybrid system.

Biological methane-to-methanol conversion is fundamentally constrained by an intrinsic trade-off between methanol accumulation and intracellular NADH regeneration, often motivating external electron supplementation, which can be inefficient. Herein, a biochar-mediated photocatalytic biohybrid system was constructed by integrating a biochar-TiO2 composite with Methylosinus trichosporium OB3b for methane-to-methanol conversion. The system achieved a peak methanol yield of 10.89 mmol·L-1 with a selectivity of 99.4%, combining relatively high methanol accumulation with high selectivity without the…

Diagnostic Challenges of Methanol Poisoning in Resource-Limited Settings: A Case Series.

Methanol poisoning is a life-threatening emergency, but diagnosis is challenging in resource-limited settings lacking toxicology testing. This case series presents three cases of methanol poisoning: two patients with neurological symptoms and unexplained high anion gap metabolic acidosis were initially misdiagnosed as stroke or diabetic ketoacidosis and died after transfer, whereas the third patient received prompt empirical hemodialysis based on clinical suspicion and survived with full recovery. These cases illustrate that early recognition and empirical hemodialysis guided by clinical judgm…

Selective production of olefins from methanol over a heteroatomic SAPO-34 zeolite.

The methanol-to-olefins (MTO) process has the potential to bridge future gaps in the supply of sustainable lower olefins. Promoting the selectivity of propylene and ethylene and revealing the catalytic role of active sites are challenging goals in MTO reactions. Here, we report a novel heteroatomic silicoaluminophosphate (SAPO) zeolite, SAPO-34-Ta, which incorporates active tantalum(V) sites within the framework to afford an optimal distribution of acidity. SAPO-34-Ta exhibits a remarkable total selectivity of 85.8% for propylene and ethylene with a high selectivity of 54.9% for propylene on f…

Platinum-copper nanowire networks with enhanced CO tolerance toward methanol oxidation electrocatalysis.

Developing platinum-based electrocatalysts with high CO tolerance for the methanol oxidation reaction (MOR) is crucial for the practical application of direct methanol fuel cells (DMFCs). Herein, we employed a straightforward one-step method to synthesize Pt x Cu y network nanowires (NWNs), which exhibit the advantages of structural stability and bimetallic ensembles. The synergistic effect of compressive strain and the ligand effect, induced by Cu incorporation, can effectively lower the d-band center of Pt, thereby weakening the adsorption strength of CO on the catalyst surface. The optimize…

Defect-Promoted Catalytic Conversion of Carbon Monoxide to Methanol on ThO2 Surfaces.

The conversion of carbon monoxide (CO) to methanol (CH3OH) is an attractive process as it converts a toxic and environmentally harmful gas into a widely used industrial chemical via hydrogenation. Traditionally, this process is catalyzed by Cu-based multicomponent materials, which require high pressures and temperatures, making the process costly and environmentally unfriendly. In this article, we show that thorium dioxide (ThO2) is an attractive alternative catalyst that is earth abundant and chemically and thermally stable. Using quantum chemistry calculations based on density functional the…
