Metformin Protects Cells from Mutant Huntingtin Toxicity Through Activation of AMPK and Modulation of Mitochondrial Dynamics.

Huntington's disease (HD) is a devastating neurodegenerative disease caused by the pathological elongation of the CAG repeats in the hunting … ment prevented mitochondrial membrane depolarization and excess fission and modulated the disturbed mitochondrial dynamics in HD cells. We confirmed that metformin crossed the blood-brain barrier after oral administration and activated AMPK in the mouse brain. Our results urge further evaluation of the clinical potential for use of metformin in HD treatment.

Alterations in mitochondrial dynamics induced by tebufenpyrad and pyridaben in a dopaminergic neuronal cell culture model.

Tebufenpyrad and pyridaben are two agro-chemically important acaricides that function like the known mitochondrial toxicant rotenone. Although these two compounds have been commonly used to kill populations of mites and ticks in commercial greenhouses, their neurotoxic profiles remain largely unknown. Therefore, we investigated the effects of these two pesticides on mitochondrial structure and function in an in vitro cell culture model using the Seahorse bioanalyzer and confocal fluorescence imaging. The effects were compared with rotenone. Exposing rat dopaminergic neuronal cells (N27 cells)

Mangiferin attenuates UV-B-induced premature senescence through suppression of mitochondrial ROS and restoration of mitochondrial dynamics in human dermal fibroblasts and BALB/c mice.

Human skin is continuously exposed to harmful Ultraviolet Radiations from the Sun leading to various morphological (pigmentation, sun burn, loss of elasticity and wrinkling) and physiological (ROS generation, oxidative stress, inflammation, DNA damage and cell death) alterations. All these changes make skin more susceptible to skin aging out of which cellular senescence plays a major role in UV-B driven skin aging. Senescence is the phenomenon of cell cycle arrest associated with changes at morphological and cellular levels i.e. compromised nuclear integrity, dysfunctional mitochondria produci

Myosin A and F-Actin play a critical role in mitochondrial dynamics and inheritance in Toxoplasma gondii.

The single mitochondrion of the obligate intracellular parasite Toxoplasma gondii is highly dynamic. Toxoplasma's mitochondrion changes morphology as the parasite moves from the intracellular to the extracellular environment and during division. Toxoplasma's mitochondrial dynamic is dependent on an outer mitochondrion membrane-associated protein LMF1 and its interaction with IMC10, a protein localized at the inner membrane complex (IMC). In the absence of either LMF1 or IMC10, parasites have defective mitochondrial morphology and inheritance defects. As little is known about mitochondrial inhe

Mitochondrial targeted fluorescent nitrite peroxide probe for dynamic monitoring of cellular lung injury.

Herein, a mitochondrial targeted fluorescent nitrite peroxide probe CHP for dynamic monitoring of cellular lung injury was developed. For the practical delivery and selectivity, the structural features including pyridine head and borate recognition group were selected. CHP could respond to ONOO- with the 585 nm fluorescence signal. The detecting system indicated advantages such as wide linear range (0.0-30 μM), high sensitivity (LOD = 0.18 μM), high selectivity and steadiness under different environmental conditions including pH (3.0-10.0), time (48 h) and medium. In living A549 cells, the res

Dynamic observation of 5-fluorouracil-induced myocardial injury and mitochondrial autophagy in aging rats.

Patients treated with 5-fluorouracil (5-FU) can develop rare but potentially severe cardiac effects, including cardiomyopathy, angina pectoris, heart failure and cardiogenic shock. The specific pathologies and underlying mechanisms are yet to be fully understood. The results of previous studies have indicated that mitochondrial autophagy is widely detected in many angiocardiopathies. In the present study, the dynamic changes in the homeostasis of mitochondrial injury and autophagy were observed in rats treated with 5-FU for different durations. A corresponding control group and a 5-FU model gr

Dynamic changes of mitochondrial fusion and fission proteins after transient cerebral ischemia in mice.

With fusion or fission, mitochondria alter their morphology in response to various physiological and pathological stimuli, resulting in elongated, tubular, interconnected, or fragmented forms. Immunohistochemistry and Western blot analysis were performed at 2 days, 7 days, 14 days, and 28 days after 90 min of transient middle cerebral artery occlusion (tMCAO) in mice. This study showed that mitochondrial fission protein dynamin-related protein 1 (Drp1) and fusion protein optic atrophy 1 (Opa1) were both upregulated in the ischemic penumbra, with the peak at 2 days after tMCAO, whereas phosphor

The role of MICOS in modulating mitochondrial dynamics and structural changes in vulnerable regions of Alzheimer's Disease.

Mitochondrial contact site and cristae organizing system (MICOS) complexes are critical for maintaining the mitochondrial architecture, cristae integrity, and organelle communication in neurons. MICOS disruption has been implicated in neurodegenerative disorders, including Alzheimer's disease (AD), yet the spatiotemporal dynamics of MICOS-associated neuronal alterations during aging remain unclear. Using three-dimensional reconstructions of hypothalamic and cortical neurons, we observed age-dependent fragmentation of mitochondrial cristae, reduced intermitochondrial connectivity, and compartme

Simple to Complex: The Role of Actin and Microtubules in Mitochondrial Dynamics in Amoeba, Yeast, and Mammalian Cells.

Mitochondria are complex organelles that provide energy for the cell in the form of adenosine triphosphate (ATP) and have very specific structures. For most organisms, this is a reticular or tubular mitochondrial network, while others have singular oval-shaped organelles. Nonetheless, maintenance of this structure is dependent on the mitochondrial dynamics, fission, fusion, and motility. Recently, studies have shown that the cytoskeleton has a significant role in the regulation of mitochondrial dynamics. In this review, we focus on microtubules and actin filaments and look at what is currently…

Protease OMA1 modulates mitochondrial bioenergetics and ultrastructure through dynamic association with MICOS complex.

Remodeling of mitochondrial ultrastructure is a process that is critical for organelle physiology and apoptosis. Although the key players in this process-mitochondrial contact site and cristae junction organizing system (MICOS) and Optic Atrophy 1 (OPA1)-have been characterized, the mechanisms behind its regulation remain incompletely defined. Here, we found that in addition to its role in mitochondrial division, metallopeptidase OMA1 is required for the maintenance of intermembrane connectivity through dynamic association with MICOS. This association is independent of OPA1, mediated via the M

Cell-specific energy crisis in the ageing brain: Mitochondrial dynamics drives neuron-glia metabolic uncoupling and neurodegeneration.

Ageing is the primary risk factor for neurodegeneration and age-related cognitive decline, which is increasingly recognised as a systemic collapse of metabolic crosstalk between neurons and glial cells in the brain. This narrative review elucidates that mitochondrial dynamics - encompassing biogenesis, fusion, fission, and mitophagy - acts as the core regulatory mechanism governing this multicellular interaction network, and drives the cell-specific energy crisis that underpins pathological progression in the ageing brain. We delineate that senescent astrocytes disrupt the astrocyte-neuron lac…
