Mycotoxin deoxynivalenol drives antibiotic resistance evolution in Bacillus cereus via cell wall remodeling, compromising clinically critical antibiotics.

The molecular mechanism by which deoxynivalenol (DON) induces bacterial antibiotic resistance remains unexplored. As the most prevalent mycotoxin contaminant, DON poses significant public health risks due to the absence of dedicated monitoring standards in probiotic products. This study uncovers the previously unknown mechanism through which DON exposure drives antibiotic resistance evolution via bacterial cell wall remodeling in Bacillus cereus. Analysis of 30 Bacillus-containing probiotic products in China detected universal DON contamination (134.37-674.56 ppb). Following 10-day sublethal D

The mycotoxin zearalenone and its metabolite α-zearalenol impair anticancer activity of doxorubicin in estrogen receptor-positive breast cancer cells.

Our environment, especially our diet, exposes us to numerous bioactive compounds that have the potential to interact with anticancer therapies. Although understanding these exposome-drug interactions is crucial for optimizing anticancer therapy, limited knowledge exists on the interference of contaminants, especially mycotoxins, with anticancer drugs. One of the most prevalent mycotoxins in food and feed is zearalenone (ZEN), which demonstrated estrogenic activity that is even stronger in its metabolite α-zearalenol (α-ZEL). Consequently, in this study, we investigated whether these mycoestrog…

Low Doses of Deoxynivalenol and Zearalenone Alone or in Combination with a Mycotoxin Binder Affect ABCB1 mRNA and ABCC2 mRNA Expression in the Intestines of Pigs.

Mycotoxin binders, in combination with enzymes degrading some mycotoxins, contribute to feed detoxification. Their use reduces economic losses and the negative impacts of mycotoxins on animal health and productivity in farm animals. The aim of this study was to evaluate the efficacy of a mycotoxin detoxifier on the expression of the ATP-binding cassette efflux transporters ABCB1 mRNA and ABCC2 mRNA, which transport xenobiotics and thus have a barrier function, in the tissues of pigs exposed to low doses of deoxynivalenol (DON, 1 mg/kg feed) and zearalenone (ZEN, 0.4 mg/kg feed) for 37 days. Effect of Compound Probiotics and Mycotoxin Degradation Enzymes on Alleviating Cytotoxicity of Swine Jejunal Epithelial Cells Induced by Aflatoxin B₁ and Zearalenone.

Zearalenone (ZEA) and aflatoxin B₁ (AFB₁) are two main kinds of mycotoxins widely existing in grain and animal feed that cause a lot of economic loss and health problems for animals and humans. In order to alleviate the cytotoxic effects of AFB₁ and ZEA on swine jejunal epithelial cells (IPEC-J2), the combination of a cell-free supernatant of compound probiotics (CFSCP) with mycotoxin degradation enzymes (MDEs) from Aspergillus oryzae was tested. The results demonstrated that coexistence of AFB₁ and ZEA had synergetic toxic effects on cell viability. The cell viability was decreased with mycot…

A single exposure to the tremorgenic mycotoxin lolitrem B inhibits voluntary motor activity and spatial orientation but not spatial learning or memory in mice.

The indole diterpenoid toxin lolitrem B is a tremorgenic agent found in the common grass species, perennial ryegrass (Lolium perenne). The toxin is produced by a symbiotic fungus Epichloë festucae (var. lolii) and ingestion of infested grass with sufficient toxin levels causes a movement disorder in grazing herbivores known as 'ryegrass staggers'. Beside ataxia, lolitrem B intoxicated animals frequently show indicators of cognitive dysfunction or exhibition of erratic and unpredictable behaviours during handling. Evidence from field cases in livestock and controlled feeding studies in horses h

Dietary AFETOXIN-BD supplementation mitigates multi-mycotoxin effects on performance, egg quality, and health status of late-laying hens.

Mycotoxins in poultry feed remain a major challenge to laying hen productivity, mainly in the late laying period when hens are at a higher risk of oxidative stress, compromised gut integrity and reduced reproductive performance. The efficacy of a commercial mycotoxin degrading preparation (AFETOXIN-BD) in reducing the negative impact of multi-mycotoxin contaminated feeds was evaluated. Two hundred 54-week-old Hy-Line Brown laying hens were randomly allotted to four dietary treatments (5 replicates of 10 hens) and fed for 35 days: a control diet (CON), a mycotoxin-contaminated diet (MT), CON pl…

Butyrate reduces epithelial barrier dysfunction induced by the foodborne mycotoxin deoxynivalenol in cell monolayers derived from pig jejunum organoids.

The foodborne mycotoxin deoxynivalenol (DON) produced by Fusarium species threats animal and human health through disruption of the intestinal barrier. Targeting the gut microbiota and its products appears as a promising strategy to mitigate DON intestinal toxicity. In this study, we investigated whether the bacterial metabolite butyrate could alleviate epithelial barrier disruption induced by DON. We used a model of cell monolayers derived from porcine jejunum organoids allowing to reproduce the cellular complexity of the intestinal epithelium. Our results show that DON dose-dependently disru

The salicylate 1,2-dioxygenase from Pseudaminobacter salicylatoxidans DSM 6986T is a bifunctional enzyme that inactivates the mycotoxin ochratoxin A by a novel amidohydrolase activity.

The salicylate 1,2-dioxygenase from the bacterium Pseudaminobacter salicylatoxidans DSM 6986T (PsSDO) is a versatile metalloenzyme that participates in the aerobic biodegradation of aromatic compounds, such as gentisates and salicylates. Surprisingly, and unrelated to this metabolic role, it has been reported that PsSDO may transform the mycotoxin ochratoxin A (OTA), a molecule that appears in numerous food products that results in serious biotechnological concern. In this work, we show that PsSDO, together with its dioxygenase activity, behaves as an amidohydrolase with a marked specificity f

Evaluation of the mycotoxin patulin on cytotoxicity and oxidative stress in human glioblastoma cells and investigation of protective effect of the antioxidant N-acetylcysteine (NAC).

Mycotoxins are secondary metabolites produced by various kinds of fungi that can induce disease in humans. The fungal species Penicillium expansum produces patulin (C7H6O4), a polyketide lactone mycotoxin found in fruits. Patulin is classified as noncarcinogen; however, recently, it has been associated with harmful effects on the central nervous system. Patulin's toxic action has been established in various brain models; however, its effect on human glioblastoma remains elusive. This study explores whether patulin induces cytotoxicity through oxidative stress in DBTRG-05MG human glioblastoma c…
