BACKGROUND: Bletilla striata is a traditional Chinese medicine used to treat hemorrhage, scald, gastric ulcer, pulmonary diseases and inflammations. In this study, we investigated bioactivity of the effective fraction of B. striata (EFB) in reducing the inflammatory cytokine production induced by water or organic extracts of PM2.5.
METHODS: PM2.5 extracts were collected and analyzed by chromatographic system and inductively coupled plasma mass spectrometer. Cell viability was measured using MTS (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium) assay, a

Autosomal dominant (AD) NFKB1 deficiency is thought to be the most common genetic etiology of common variable immunodeficiency (CVID). However, the causal link between NFKB1 variants and CVID has not been demonstrated experimentally and genetically, and there has been insufficient biochemical characterization and enrichment analysis. We show that the cotransfection of NFKB1-deficient HEK293T cells (lacking both p105 and its cleaved form p50) with a κB reporter, NFKB1/p105, and a homodimerization-defective RELA/p65 mutant results in p50:p65 heterodimer-dependent and p65:p65 homodimer-independen

BACKGROUND: Periodontitis disease infection initiates host immune response, and alveolar bone damage is a hallmark of periodontitis. Bone damage occurs due to changes in osteoclast activity in response to local inflammation. Nuclear factor κB (NF-κB) signaling is essential for inflammatory responses and plays a pivotal role in osteoclast formation and activation. Tripartite motif 14 (Trim14) is a crucial regulator of the noncanonical NF-κB signaling. Here, we investigated the role of Trim14 in chronic periodontitis.
METHODS: The development of immune cells and osteoclast formation was evaluat

Hodgkin lymphoma (HL) arises from aberrant B lymphocytes due to genetic mutations, epigenetic alterations, and persistent inflammation, leading to chronic hyperactivation of the NF-κB signaling cascade, which drives cell survival and immune evasion. The pathway regulates cell survival and immune response and is abnormally activated through an interplay between genetic mutations, epigenetic changes, and persistent inflammation. This review examines the relationship between B cell mutations and NF-κB signaling in the progression of HL. Particular focus is drawn to genetic mutations concerning Ep

Three distinct chromosomal translocations, t(11;18), t(1;14), and t(14;18), involving the API2-MALT1 fusion protein, BCL10, and MALT1 have been convincingly implicated in the pathogenesis of mucosa-associated lymphoid tissue (MALT) lymphomas. Recent genetic and biochemical studies have indicated that BCL10 and MALT1 form a physical and functional complex and are both essential for nuclear factor kappaB (NF-kappaB) activation by antigen receptor stimulation in lymphocytes. API2-MALT1 can bypass the BCL10/MALT1 pathway linking to NF-kappaB activation, thereby inducing antigen receptor-independen

Tax, a regulatory protein of human T-cell leukemia virus type 1 (HTLV-1), is an oncoprotein which immortalizes human T cells and induces tumors in transgenic mice. These effects may be due to its interaction with cellular proteins, consisting of several transcription factors including CREB, NF-kappa B and SRF, and the transcriptional inhibitor, I kappa B. Here, we found that Tax binds to a cyclin-dependent kinase inhibitor, p16INK4A, which has ankyrin motifs similar to I kappa B. p16INK4A binds to the cyclin-dependent kinases, CDK4 and CDK6, and inhibits their activity, resulting in suppressio

Nuclear factor kappa B (NF-κB) regulates inflammation and immunity and is frequently dysregulated in autoimmune diseases, inflammatory disorders, and cancers. Cytokines and pathogens activate NF-κB transcription factors, and nutrients and metabolites, including fatty acids, also modulate its activation, though the underlying mechanisms remain unclear. Here, we report that RelA (p65), a major component of the NF-κB transcription complex, is autopalmitoylated at cysteine 197 and 206. Palmitoylation of p65 enhances its nuclear localization and p65-p50 complex formation, thereby increasing NF-κB t

Recent evidence emphasized that transglutaminase 2 (TG2), a protein cross-linking enzyme, may play a role in the early phase of inflammation. High levels of TG2 have been associated with the constitutive activation of nuclear factor-kappa B (NF-κB) that is considered the main regulator of inflammation. In this context, the receptor activator of NF-kappa B ligand (RANKL) and receptor activator of NF-κB have extensive functions in the regulation of cytokine secretion associated with different pathological conditions. The human periodontal ligament (HPDL) cells, which express and secrete osteopro

The effects of methyl mercury chloride (MMC) on DNA binding activities of nuclear factor kappa B(NF-kB) in developing rat cerebella and cerebra were investigated with electrophoretic mobility shift assays(EMSAs). The bindings of NF-kB in nuclei of rat cerebra and cerebella to kB probes showed two bands on gel shift in both control and experiment groups. NF-kB I and NF-kB II DNA binding activities of nuclear protein extracts from rat cerebra exposed to MMC in uterus, was lower than control groups on postnatal day 3 and 7, while that from rat cerebella was higher than control groups. The results

Nuclear factor kappa B (NF-κB) and type 1 interferon (T1-IFN) signaling are innate immune mechanisms activated upon viral infection. However, the role of NF-κB and its interplay with T1-IFN in antiviral immunity is poorly understood. We show that NF-κB is essential for resistance to ectromelia virus (ECTV), a mouse orthopoxvirus related to the virus causing human smallpox. Additionally, an ECTV mutant lacking an NF-κB inhibitor activates NF-κB more effectively in vivo, resulting in increased proinflammatory molecule transcription in uninfected cells and organs and decreased viral replication.

Nuclear factor-kappa B (NF-kappaB) comprises a family of inducible transcription factors that serve as important regulators of the host immune and inflammatory responses. The NF-kappaB signals are activated via the canonical and/or noncanonical pathways in response to diverse stimuli. The excessive action of NF-kappaB signal-transduction pathways frequently causes self-injurious phenomena such as allergic diseases, vascular disorders, and ischemia-reperfusion neuronal damage. In the inner ear, the role of NF-kappaB has not been clarified because the activated NF-kappaB signals potentially indu

Human T-cell lymphotropic virus type I(HTLV-I) is a type C retrovirus, closely associated with adult T-cell leukemia. In the past few years, studies have revealed an association between the virus and disorders of organ systems including myelopathy, polymyositis, alveolitis, and Sjögren's syndrome. In addition, we have proposed that HTLV-I-associated proliferative changes in nonlymphocyte synovial cells are termed HTLV-I-associated arthropathy(HAAP). To clarify the pathologic association of HTLV-I with this arthropathy, we attempted to detect HTLV-I proviral DNA in synovial tissue. Proviral HTL

Osteoclast hyperactivity represents a central mechanism in pathological bone destruction, underscoring the importance of discovering novel anti-resorptive compounds. In this study, we present early-stage evidence that 8-Epixanthatin can inhibit osteoclast differentiation induced by receptor activator of nuclear factor kappa-B ligand (RANKL). 8-Epixanthatin exhibited no significant cytotoxicity at the concentrations used for osteoclast differentiation studies. The compound showed concentration-dependent reductions in TRAP-positive multinucleated osteoclasts, with an IC50 value of 2.3 μM. Our me

Incontinentia pigmenti is an X-linked dominant multisystemic disorder caused by pathogenic variants in the inhibitor of nuclear factor kappa B kinase regulatory subunit gamma (IKBKG) gene. Mutations in the IKBKG gene, encoding the NF-kappa-B essential modulator protein, underlie its pathogenesis, leading to inflammation-driven vascular ischemia and significant ocular manifestations. The authors report a case of an Asian-Indian infant diagnosed as having stage 3 incontinentia pigmenti in both eyes treated with anti-vascular endothelial growth factor and laser photocoagulation where standard who

OBJECTIVES: Our study investigated: 1) the contribution of nuclear factor kappa-B (NF-kappaB) signaling pathway to the enhanced inflammatory response observed in unstable angina (UA) patients with elevated levels of C-reactive protein (CRP); and 2) whether CRP may have direct proinflammatory effects via NF-kappaB activation.
BACKGROUND: Unstable angina patients with elevated CRP have enhanced inflammatory response and increased risk of persistent instability, myocardial infarction, and death.
METHODS: We studied 28 patients with history of UA and persistently elevated CRP (>3 mg/l) followed

Nuclear factor kappa-B (NF-κB) signaling-mediated inflammation contributes greatly to the pathogenesis of periodontitis. Neddylation, a ubiquitin-like posttranslational modification, is known to regulate NF-κB signaling. DCUN1D1 (defective in cullin neddylation 1 domain containing 1) is a critical factor in neddylation and has been shown to regulate NF-κB activation. However, the previse roles of DCUN1D1 in periodontitis are not fully elucidated. To explore the roles of DCUN1D1 in periodontitis, the expression of DCUN1D1 was measured in gingival tissues of patients with periodontitis. We inhib

The A(3) adenosine receptor (A(3)AR) is over-expressed in inflammatory cells and was defined as a target to combat inflammation. Synthetic agonists to this receptor, such as IB-MECA and Cl-IB-MECA, exert an anti-inflammatory effect in experimental animal models of adjuvant- and collagen-induced arthritis. In this study we present a novel A(3)AR agonist, CF502, with high affinity and selectivity at the human A(3)AR. CF502 induced a dose dependent inhibitory effect on the proliferation of fibroblast-like synoviocytes (FLS) via de-regulation of the nuclear factor-kappa B (NF-kappaB) signaling pat

Lithium, the lightest natural-occurring alkali metal with an atomic number of three, stabilizes the mood to prevent episodes of acute manic and depression. Multiple lines of evidence point to lithium as an anti-suicidal, anti-viral, anti-cancer, immunomodulatory, neuroprotective and osteoprotective agent. This review article provides a comprehensive review of studies investigating the bone-enhancing effects of lithium and its possible underlying molecular mechanisms. Most of the animal experimental studies reported the beneficial effects of lithium in defective bones but not in healthy bones.

Parkinson's disease (PD) is a progressive neurodegenerative disorder hallmarked by dopaminergic neuronal loss and pathological α-synuclein accumulation. Increasing evidence positions neuroinflammation as a central driver of disease initiation and progression, orchestrating maladaptive glial responses that amplify neuronal vulnerability. At the heart of these inflammatory cascades lies the NF-κB signaling pathway, a master regulator of immune and stress responses in neurons and glia. While basal NF-κB activity supports neuronal homeostasis, chronic dysregulation in PD perpetuates microglial act

The nuclear factor κB (NF-κB) signaling pathway plays a crucial role in regulating immune responses, inflammation, and cell survival. After the discovery of NF-κB by Ranjan Sen and David Baltimore as a nuclear factor that binds near the κ light-chain gene in activated B-cells [1], extensive research has been conducted to uncover key biochemical mechanisms underlying the pathway and the downstream genes regulated by NF-κB. The core regulatory module of NF-κB/IκB (inhibitor of NF-κB)/IKK (IκB kinase) has also been a leading example of cell signaling pathways that can produce oscillatory activiti
