Androgens influence the development and growth of the mammary gland in women. Treatment of animals and cultured cells with androgens has either inhibitory or stimulatory effects on the proliferation of mammary epithelia and cancer cells; the mechanisms for these dual functions are still not very clear and are discussed in this review. Pancreatic cancer induced in hamsters offers a unique model for studying important aspects of the disease. Studies of the mechanisms of carcinogenesis show that carcinogens require specific molecular structures for pancreatotrophic action. Syrian golden hamsters were treated with N delta-(N-methyl-N-nitrosocarbamoyl)-L-ornithine (MNCO), a nitrosourea amino acid, which has induced a high incidence of breast, kidney and skin neoplasms and a low incidence of pancreatic carcinoma in rats. MNCO induced a few breast and skin carcinomas, and a high incidence of foci of atypical acinar cells and of focal ductular abnormalities in the exocrine pancreas. The latter were similar to lesions observed in rats treated with MNCO. MNCO was less toxic and less effective as a carcinogen in hamster than in the rat.

Theophyllin, an inhibitor of cAMP-degrading phosphodiesterase, stimulates melanin biosynthesis in cultures of RPMI 3460 hamster melanoma cells. The development of Vx2 carcinoma induced in the kidney of rabbits was followed by urography. First the X-ray anatomy of the renal pelvis in the rabbit is described, pointing out the differences against the human pattern. Urography is easy to carry out and to evaluate, and it revealed some sign of the turnover in 75,6% of the cases in the third week and in 100% after 21 days. The radiological symptoms are described and classified. These symptoms are not specific but call attention to the progress of carcinoma.

The effect of chronic infection with the protozoan parasite Babesia rodhaini on the subsequent development of mouse leukemia was investigated in three different murine leukemia-lymphoma model systems: congenital lymphoma (in AKR mice), irradiation-induced lymphoma (in C57BL mice), and Rauscher-virus-induced leukemia in NMRI mice. Leukemia incidence was significantly greater than in controls in Babesia-infected (NMRI strain) mice that had been infected with an adjusted dose of Rauscher leukemia virus.

Large testing programs have been set up to help assess the carcinogenic potential of chemicals introduced into the environment. Most test protocols used in these programs require extensive pathologic evaluations of all test and control animals.
