20-HETE in the regulation of vascular and cardiac function.

20-HETE, the ω-hydroxylation product of arachidonic acid catalyzed by enzymes of the cytochrome P450 (CYP) 4A and 4F gene families, is a bioactive lipid mediator with potent effects on the vasculature including stimulation of smooth muscle cell contractility, migration and proliferation as well as activation of endothelial cell dysfunction and inflammation. Clinical studies have shown elevated levels of plasma and urinary 20-HETE in human diseases and conditions such as hypertension, obesity and metabolic syndrome, myocardial infarction, stroke, and chronic kidney diseases. Studies of polymorp

Evidence from a leukemia model for maintenance of vascular endothelium by bone-marrow-derived endothelial cells.

The maintenance of tissues of virtually all organs depends on a sufficient blood supply. During embryogenesis, primitive blood vessels are formed de novo by the aggregation of angioblasts, a process that is termed vasculogenesis. In postnatal life, the development of new blood vessels is restricted to the female reproductive tract (during the ovulatory cycle) and to sites of wound healing, and occurs through a process called angiogenesis, i.e. the sprouting of new vessels from the preexisting vasculature. However, neovascularization can also occur under pathological conditions, e.g. tumor cell

A Novel Eye Drop Formulation for Potential Treatment of Neovascular Age-Related Macular Degeneration.

PURPOSE: Drug delivery to posterior ocular tissues via topical eye drop administration is arduous due to the unique anatomy and physiology of the eye. Therefore, treatments for posterior eye disease have to be administered via intravitreal injection or systemic route, both of which have their drawbacks. Herein, the objective of this work was to demonstrate that a specially designed eye drop formulation could effectively deliver small-molecule vascular endothelial growth factor (VEGF) inhibitor to posterior ocular tissues for antiangiogenic therapy. METHODS: The unique eye drop formulation, te

Inhibition of angiogenesis, fibrosis and thrombosis by tetramethylpyrazine: mechanisms contributing to the SDF-1/CXCR4 axis.

BACKGROUND: Tetramethylpyrazine (TMP) is one of the active ingredients extracted from the Chinese herb Chuanxiong, which has been used to treat cerebrovascular and cardiovascular diseases, pulmonary diseases and cancer. However, the molecular mechanisms underlying the actions of TMP have not been fully elucidated. In a previous study we showed that TMP-mediated glioma suppression and neural protection involves the inhibition of CXCR4 expression. The SDF-1/CXCR4 axis plays a fundamental role in many physiological and pathological processes. In this study, we further investigated whether the reg

Experimental preretinal neovascularization by laser-induced venous thrombosis in rats.

PURPOSE: Retinal ischemia and neovascularization (NV) are important components of many retinal disorders. To facilitate further investigation of retinal ischemia and neovascularization, we sought to develop a reproducible in vivo experimental model of venous occlusion by photodynamic thrombosis in rats. METHODS: After anesthesia, 27 eyes of pigmented rats received an intraperitoneal injection of 0.2 ml of, 10% sodium fluorescein 15 minutes prior to laser treatment. With a blue-green argon laser, selected venous sites next to the optic nerve head were photocoagulated indirectly with a 78 diopt

Molecular and functional diversity of vascular endothelial growth factors.

Members of the vascular endothelial growth factor (VEGF) family are crucial regulators of neovascularization and are classified as cystine knot growth factors that specifically bind cellular receptor tyrosine kinases VEGFR-1, VEGFR-2, and VEGFR-3 with high but variable affinity and selectivity. The VEGF family has recently been expanded and currently comprises seven members: VEGF-A, VEGF-B, placenta growth factor (PlGF), VEGF-C, VEGF-D, viral VEGF (also known as VEGF-E), and snake venom VEGF (also known as VEGF-F). Although all members are structurally homologous, there is molecular diversity

Teriparatide attenuates scarring around murine cranial bone allograft via modulation of angiogenesis.

Nearly all bone fractures in humans can deteriorate into a non-union fracture, often due to formation of fibrotic tissue. Cranial allogeneic bone grafts present a striking example: although seemingly attractive for craniofacial reconstructions, they often fail due to fibrosis at the host-graft junction, which physically prevents the desired bridging of bone between the host and graft and revitalization of the latter. In the present study we show that intermittent treatment with recombinant parathyroid hormone-analogue (teriparatide) modulates neovascularization feeding in the graft surrounding

Fetoplacental growth and vascular development in overnourished adolescent sheep at day 50, 90 and 130 of gestation.

To establish the basis for altered placental development and function previously observed at late gestation, fetoplacental growth and placental vascular development were measured at three stages of gestation in a nutritional paradigm of compromised pregnancy. Singleton pregnancies to a single sire were established and thereafter adolescent ewes were offered an optimal control (C) or a high (H) dietary intake. At day 50, the H group had elevated maternal insulin and amniotic glucose, whereas mass of the fetus and placenta were unaltered. At day 90, the H group exhibited elevated maternal insuli

Mechanobiological regulation of endothelial vascularization after myocardial infarction: matrix mechanics, hydrogel strategies and applications.

Myocardial infarction (MI) triggers a wound-healing cascade that restores structural integrity but reshapes the cardiac mechanical microenvironment. The transition from compliant, healthy myocardium to stiff, fibrotic scar tissue creates biomechanical barriers that can impede neovascularization and contribute to heart failure progression. These biomechanical cues regulate endothelial fate decisions, including migration, proliferation, branching, and lumen formation, and thus influence neovascularization and perfusion recovery. This review addresses the mechanobiological principles governing en

Effect of gut bacterial extracellular vesicles on angiogenic potential and vascular integrity: positive and negative aspects.

Extracellular vesicles (EVs) are intercellular mediators in prokaryotic and eukaryotic systems that have the potential to regulate various physiological and pathological processes in recipient cells. Among them, bacterial extracellular vesicles (BEVs), including Gram-negative outer membrane vesicles (OMVs) and Gram-positive membrane vesicles (MVs) produced by the gut microbiota, have special roles in humans and animals. Due to their nanoscale dimensions and distinct molecular payloads, BEVs can cross biological barriers, including the vascular endothelium, to reach distant target tissues and a

Preliminary study on the effect of compressive stress on the angiogenic ability of dental pulp cells.

PURPOSE: Stem cell-based dental pulp regeneration is a promising treatment for pulp necrosis, wherein vascularization is critical. Dental pulp cells (DPCs) are mechanosensitive; however, the mechanism by which compressive stress regulates their angiogenic activity remains unclear. This study investigated the underlying mechanotransductive mechanisms and optimal loading parameters. METHODS: An in vitro compressive stress model for DPCs was established. Cell proliferation and migration under varying stress magnitudes and durations were evaluated using Cell Counting Kit-8 (CCK-8) and scratch wou

Tie1 ectodomain cleavage governs vascular responses to inflammatory stimulation but is dispensable for angiogenic signaling.

BACKGROUND: The coronavirus disease 2019 (COVID-19) is spreading around the world. The COVID-19 vaccines may improve concerns about the pandemic. However, the roles of inactivated vaccines in older patients (aged ≥60 years) with infection of Delta variant were less studied. METHODS: We classified the older patients with infection of Delta variant into three groups based on the vaccination status: no vaccination (group A, n = 113), one dose of vaccination (group B, n = 46), and two doses of vaccination (group C, n = 22). Two inactivated COVID-19 vaccines (BBIBP-CorV or CoronaVac) were evaluate

Emerging functionalized artificial periostea: Diverse origins, multifaceted roles and therapeutic frontiers.

The periosteum, a specialized connective tissue layer enveloping bone surfaces, serves as a critical regulator of bone healing through its integrated neurovascular architecture, stem cell niche properties, and mechanical barrier function. Clinically, traumatic injuries or pathological conditions frequently result in periosteal tearing, avulsion, or complete loss, fundamentally disrupting its biological integrity. In practice, repairing a torn or absent periosteum is difficult, because existing options rarely recreate its coordinated biological cues and protective function. The limited availabi

Associations between physical activity, sedentary behavior, body mass index and changes in placental structure and vascularization - data from the FitMum randomized controlled trial.

OBJECTIVE: Maternal physical activity (PA), sedentary time (ST) and body-mass index (BMI) might influence placental development. We examined associations between PA, ST during pregnancy and pre-pregnancy BMI, and placental structural outcomes as well as mRNA expression levels of vascular growth factors. METHODS: The FitMum randomized controlled trial included 220 healthy, pregnant women. PA and ST were objectively measured 24/7 from inclusion before gestational week 15 + 0 until delivery. This secondary analysis of FitMum includes all participants with an available placenta sample (n = 118).

RNA-Seq Analysis of Human Cumulus Cells Identifies Angiogenic Pathways Associated with Infertility.

Non-invasive assessment of oocyte quality remains a challenge in assisted reproductive technology (ART). Through their bidirectional communication with the gamete, cumulus cells (CCs) act as a functional mirror of oocyte competence; however, the specific angiogenic signature within this microenvironment is still poorly understood. In the present study, we performed RNA-seq on CCs from healthy oocyte donors and infertile patients, utilizing a multi-pipeline bioinformatic approach (STAR-Cufflinks, TopHat-HTSeq, and HISAT2-StringTie) to establish a high-confidence, exploratory transcriptomic prof

Targeting endothelial cytoskeletal remodeling to restore angiogenesis in ischemic heart disease.

Ischemic heart disease (IHD) remains a leading cause of morbidity and mortality worldwide. Effective restoration of myocardial perfusion relies on angiogenesis, a process frequently impaired in patients due to endothelial dysfunction. Endothelial cells orchestrate angiogenesis through dynamic cytoskeletal remodeling, which regulates migration, polarity, proliferation, and lumen formation. This review summarizes the molecular mechanisms underlying endothelial cytoskeletal remodeling and how their dysregulation contributes to angiogenesis failure in IHD. We focus on alterations in actin filament

Cascade-Responsive MXene@Cu-MOF Heterostructure Integrates Antioxidant Activity, Infection Control, and Vascularization for Tracheal Repair.

Extensive tracheal repair is limited by oxidative injury, infection, and insufficient vascularization with delayed cartilage maturation. We developed cascade-responsive MXene@Cu-MOF heterostructures within gelatin methacryloyl (GelMA) hydrogels to enable staged tracheal repair. Copper-based metal-organic framework (Cu-MOF) nanocrystals grown in situ on ultrathin Ti3C2Tx MXene form an integrated 2D/3D heterointerface, preventing restacking and ensuring uniform "backpack" distribution on a photothermal skeleton. The platform acts in three stages: 1) MXene scavenges radicals, reducing oxidative s

Immortalized smooth muscle cells enhance in vitro vasculogenesis.

De novo vessel formation (vasculogenesis) in vitro is a key step in tissue engineering to preserve tissue viability for long-term assays and testing therapeutic agents. However, in vitro vasculogenesis is often unreliable due to differences in vascular-supporting cells, including endothelial cells and stromal cells such as smooth muscle cells (SMCs) and fibroblasts. Here, we developed a robust co-culture system of HUVECs and SMCs to generate stable vascular networks capable of maintaining tissue viability over extended periods. Given that SMC plasticity is a major limitation in supporting endo
