Therapeutic effects of human adipose stem cell-conditioned medium on stroke.

Stem cell therapy is a promising approach for stroke. However, low survival rates and potential tumorigenicity of implanted cells could undermine the efficacy of the cell-based treatment. The use of stem cell-conditioned medium (CM) may be a feasible approach to overcome these limitations. Especially, specific stem cell culture condition and continuous infusion of CM into ischemic brains would have better therapeutic results. The CM was prepared by culturing human adipose-derived stem cells in a three-dimensional spheroid form to increase the secretion of angiogenic/neuroprotective factors. Is

The aqueduct of Sylvius: a sonographic landmark for neural tube defects in the first trimester.

OBJECTIVES: To describe a new first-trimester sonographic landmark, posterior displacement of the midbrain and aqueduct of Sylvius, which may be useful in early screening for neural tube defects. METHODS: This was a prospective study of 457 normal fetuses at 11 + 0 to 13 + 6 weeks' gestation. We measured the distance from the posterior border of the aqueduct of Sylvius to the anterior border of the occiput (AOS-to-occiput distance) in the axial plane and created a reference range. In the nine fetuses with abnormal midbrain position identified in the first trimester and with neural tube defect

Physics-informed neural networks and neural operators for PDEs: A review of architectures, training pathologies, and method selection.

Physics-informed neural networks (PINNs) and neural operators are increasingly used to learn solutions and solution operators for partial differential equations, but the literature remains fragmented across architectures, training objectives, and benchmark protocols. This review organizes these developments for a neural-network audience. We separate two design axes that are often conflated: representation level (solution-level vs. operator-level) and training paradigm (data-driven vs. physics-informed). The resulting taxonomy positions classical PINNs, domain-decomposition and weak-form varian

Folate deficiency disrupts key metabolic transitions within the developing neural ectoderm.

UNLABELLED: Despite long-standing epidemiological associations, the mechanism linking folate availability to gestational neural tube defects remains unclear, partly because measuring and interpreting metabolic activity in dynamic biological systems remains challenging. Here, we apply a deep-learning-based graph-guided variational autoencoder (MeRN; Metabolic Representation Network) to infer single-cell metabolic activity and states from scRNA-seq data of mouse embryogenesis. By analyzing folate-deficient embryogenesis from E7.0-E9.0, we identify a transient state within the nascent neural line

Neural and psychosocial signatures of the comorbidity between pain and affective symptoms.

INTRODUCTION: The bidirectional relationship between pain and affective symptoms suggests shared underlying mechanisms. Neurophysiological changes, psychosocial factors, and specifically alterations in instrumental learning likely contribute significantly to their co-occurrence. OBJECTIVES: In this study, we aimed to uncover factors associated with the comorbidity between pain and the severity of depressive symptoms and between pain and the severity of anxiety symptoms. METHODS: We analyzed psychosocial (n = 624) and neuroimaging (n = 689) data from a European sample of young adults with pai

Toward an intelligent computing system for the early diagnosis of Alzheimer's disease based on the modular hybrid growing neural gas.

OBJECTIVE: The proportion of older people will soon include nearly a quarter of the world population. This leads to an increased prevalence of non-communicable diseases such as Alzheimer's disease (AD), a progressive neurodegenerative disorder and the most common dementia. mild cognitive impairment (MCI) can be considered its prodromal stage. The early diagnosis of AD is a huge issue. We face it by solving these classification tasks: MCI-AD and cognitively normal (CN)-MCI-AD. METHODS: An intelligent computing system has been developed and implemented to face both challenges. A non-neural prep

Maximal Analgesic Effect Attained by the Use of Objective Neurophysiological Measurements With Closed-Loop Spinal Cord Stimulation.

OBJECTIVES: Spinal cord stimulation (SCS) has been challenged by the lack of neurophysiologic data to guide therapy optimization. Current SCS programming by trial-and-error results in suboptimal and variable therapeutic effects. A novel system with a physiologic closed-loop feedback mechanism using evoked-compound action potentials enables the optimization of physiologic neural dose by consistently and accurately activating spinal cord fibers. We aimed to identify neurophysiologic dose metrics and their ranges that resulted in clinically meaningful treatment responses. MATERIALS AND METHODS:

Detrusor nerve radiofrequency ablation (DENERA): Experimental study of safety and efficacy in an ovine model.

PURPOSE: The subtrigonal perivesical nerve plexus contains a large proportion of the bladder's innervation. A transurethral radiofrequency ablation approach has successfully denervated this region to alleviate overactive bladder symptoms, with some urothelial heat injury. We report a novel transvaginal RFA device (DENERA) and assess its feasibility and efficacy in denervating the perivesical nerve plexus of in vivo sheep. METHODS: In 14 adult female in vivo sheep, pulsed radiofrequency energy was applied transvaginally for three cycles of 4 min, maintaining the tissue temperature at 45°C, wit

Behavioral and neural measures of infant responsivity increase with maternal multisensory input in non-irritable infants.

INTRODUCTION: Parents often use sensory stimulation during early-life interactions with infants. These interactions, including gazing, rocking, or singing, scaffold child development. Previous studies have examined infant neural processing during highly controlled sensory stimulus presentation paradigms. OBJECTIVE: In this study, we investigated infant behavioral and neural responsiveness during a mother-child social interaction during which the mother provided infant stimulation with a progressive increase in the number of sensory modalities. METHODS: We prospectively collected and analyzed

Double-Sided Sapphire Optrodes with Conductive Shielding Layers to Reduce Optogenetic Stimulation Artifacts.

Optrodes, which are single shaft neural probes integrated with microelectrodes and optical light sources, offer a remarkable opportunity to simultaneously record and modulate neural activities using light within an animal's brain; however, a common problem with optrodes is that stimulation artifacts can be observed in the neural recordings of microelectrodes when the light source on the optrode is activated. These stimulation artifacts are undesirable contaminants, and they cause interpretation complexity when analyzing the recorded neural activities. In this paper, we tried to mitigate the ef

A gene expression atlas of early craniofacial development.

We present a gene expression atlas of early mouse craniofacial development. Laser capture microdissection (LCM) was used to isolate cells from the principal critical microregions, whose development, differentiation and signaling interactions are responsible for the construction of the mammalian face. At E8.5, as migrating neural crest cells begin to exit the neural fold/epidermal ectoderm boundary, we examined the cranial mesenchyme, composed of mixed neural crest and paraxial mesoderm cells, as well as cells from adjacent neuroepithelium. At E9.5 cells from the cranial mesenchyme, overlying o

The role of voltage-gated calcium channels in neurotransmitter phenotype specification: Coexpression and functional analysis in Xenopus laevis.

Calcium activity has been implicated in many neurodevelopmental events, including the specification of neurotransmitter phenotypes. Higher levels of calcium activity lead to an increased number of inhibitory neural phenotypes, whereas lower levels of calcium activity lead to excitatory neural phenotypes. Voltage-gated calcium channels (VGCCs) allow for rapid calcium entry and are expressed during early neural stages, making them likely regulators of activity-dependent neurotransmitter phenotype specification. To test this hypothesis, multiplex fluorescent in situ hybridization was used to char

Neutrophil responses to injury or inflammation impair peripheral gustatory function.

The adult peripheral taste system is capable of extensive functional plasticity after injury. Sectioning the chorda tympani (CT), a primary sensory afferent nerve, elicits transient changes in the uninjured, contralateral population of taste receptor cells. Remarkably, the deficits are specific to the sodium transduction pathway. Normal function is quickly restored in the intact nerve, in parallel with an influx of macrophages to both the denervated and uninjured sides of the tongue. However, changing the dietary environment by restricting sodium blocks the macrophage response and prolongs fun

Cell cycle kinetics of expanding populations of neural stem and progenitor cells in vitro.

Neural stem cells (NSCs) are undifferentiated, primitive cells with important potential applications including the replacement of neural tissue lost due to neurodegenerative diseases, including Parkinson's disease, as well as brain and spinal cord injuries, including stroke. We have developed methods to rapidly expand populations of mammalian stem and progenitor cells in neurosphere cultures. In the present study, flow cytometry was used in order to understand cell cycle activation and proliferation of neural stem and progenitor cells in suspension bioreactors. First, a protocol was developed

Pharmacologically altered androgens, brain activation and response inhibition in a stop-signal task in male heavy drinkers.

UNLABELLED: Heavy episodic drinking (HED) is highly prevalent in men and a major risk factor for Alcohol Use Disorder (AUD). Previous studies indicated an influence of testosterone (T) on AUD, and dihydrotestosterone (DHT) was linked to alcohol consumption, impulsivity, and response inhibition. As the role of androgens in AUD and the modulation thereof is not fully understood, this study investigated the effect of a pharmacological reduction in DHT by inhibition of the 5-alpha reductase II with finasteride on brain activation and response inhibition during a stop signal task. Fifty males with

Disentangling sensory contributions to postural control regulation through sample entropy and neural modeling: A preliminary study.

Postural control relies on the integration of visual, vestibular, proprioceptive, and auditory inputs to maintain stability. While previous studies have explored the effects of individual sensory modalities, the combined influence of multisensory disruptions on postural predictability remains unclear. We preliminary assessed postural behavior during quiet standing under manipulated sensory conditions in healthy participants. Eight adults stood barefoot on a Wii Balance Board, wearing a mixed reality headset with headphones and receiving vibrotactile stimulation to the Achilles tendons. Experim

Longitudinal mapping of a randomized controlled trial of EEG neurofeedback in PTSD: Early symptom stabilization and implications for adjunctive therapeutic potential.

INTRODUCTION: PTSD is characterized by reduced alpha (8-12 Hz) rhythms, reflecting cortical hyperarousal and disrupted large-scale network dynamics. Across single- and multi-session studies, alpha-desynchronizing EEG neurofeedback (EEG-NFB) restores alpha rhythms toward normative levels ("alpha rebound") and reduces PTSD symptoms. However, prior research has focused mainly on pre-to-post intervention changes, leaving longitudinal trajectories of therapeutic improvement and neural modulation unclear. METHODS: We conducted longitudinal mapping of clinical and neural outcomes during a 20-week ra

One is not like the other: Examining the neural response to repetitive low-level blast exposure in experienced military personnel.

BACKGROUND: Military members often report concussion-like symptoms from repetitive low-level blast (LLB) exposure, defined as overpressure from outgoing munitions like rifles and explosive breaching. Typically, the early stages of concussion and other neurological conditions (e.g., Alzheimer's Disease) lead to hyperconnectivity which is a transient and adaptive brain response to strengthen and establish neural connections to restore brain function. Over time, however, chronic hyperconnectivity can contribute to neurodegeneration. To determine whether LLB exposure also exhibits this connectivit

Exploring the spiking neural autoencoder: from hyperexcitability to noise-driven compensation.

INTRODUCTION: Understanding how artificial neural networks (ANNs) can capture biologically meaningful dynamics is a central challenge in systems neuroscience. In this work, we investigate whether spiking neural networks (SNNs) can function not only as machine-learning tools but also as biologically inspired computational analogs and tractable testbeds for studying pathological neural dynamics. METHODS: We implemented a spiking autoencoder composed of Leaky Integrate-and-Fire and Synaptic neuron models to create a controlled framework for analyzing how biologically related parametric changes t
