Boron Neutron Capture Therapy Combined with Early Successive Bevacizumab Treatments for Recurrent Malignant Gliomas - A Pilot Study.

Recurrent malignant gliomas (RMGs) are difficult to control, and no standard protocol has been established for their treatment. At our institute, we have often treated RMGs by tumor-selective particle radiation called boron neutron capture therapy (BNCT). However, despite the cell-selectivity of BNCT, brain radiation necrosis (BRN) may develop and cause severe neurological complications and sometimes death. This is partly due to the full-dose X-ray treatments usually given earlier in the treatment course. To overcome BRN following BNCT, recent studies have used bevacizumab (BV). We herein used

Arc-induced Long Period Gratings in standard and speciality optical fibers under mixed neutron-gamma irradiation.

In this paper, for the first time, the effects of mixed neutron and gamma flux on the spectral and sensing responses of Long Period Gratings (LPGs) are thoroughly analyzed. Six LPGs written by means of Electric Arc Discharge (EAD) technique in standard and speciality fibers, including radiation-hardened ones, were tested. The EAD technique was chosen because it enables the writing of gratings both in standard and not photosensitive fibers. The experiments have been carried out in a "TRIGA" pulsed nuclear reactor and the LPGs were irradiated by a gamma-ray dose-rate of 9 Gy/s and a mean 1.2∙101

Magnetic excitations in metalloporphyrins by inelastic neutron scattering: determination of zero-field splittings in iron, manganese, and chromium complexes.

Zero field splitting (ZFS) parameters of several nondeuterated metalloporphyrins [M(TPP)Cl] and [Mn(TPP)] (H2TPP = tetraphenylporphyrin) have been directly determined by inelastic neutron scattering (INS). The ZFS values are the following: D = 6.33(8) cm(-1) for [Fe(TPP)Cl], -2.24(3) cm(-1) for [Mn(TPP)Cl], 0.79(2) cm(-1) for [Mn(TPP)], and |D|= 0.234(12) cm(-1) for [Cr(TPP)Cl]. The work shows that compounds with magnetic excitations below ∼30 cm(-1) could be determined using nondeuterated samples.

Paternal monoenergetic neutron exposure results in abnormal sperm, and embryonal lethality and transgenerational tumorigenesis in mouse F1 offspring.

Experiments were conducted to assay whether monoenergetic neutron-induced genetic damage in parental germline cells can give rise to development of cancer in the offspring. Seven-week-old C3H male mice were irradiated with monoenergetic neutrons with energy levels of 0.2 or 0.6 MeV at doses of 0, 50, 100 or 200 cGy. Two weeks after irradiation, when the male mice showed an increased incidence of sperm abnormalities, they were mated with virgin 9-week-old C57BL females. Litter size was decreased and embryo lethalities were increased in a dose-dependent manner. Furthermore, tumor incidence in ma…

Synthesis and biological evaluation of folate receptor-targeted boronated PAMAM dendrimers as potential agents for neutron capture therapy.

Successful treatment of cancer by boron neutron capture therapy (BNCT) requires the selective delivery of (10)B to constituent cells within a tumor. The expression of the folate receptor is amplified in a variety of human tumors and potentially might serve as a molecular target for BNCT. In the present study we have investigated the possibility of targeting the folate receptor on cancer cells using folic acid conjugates of boronated poly(ethylene glycol) (PEG) containing 3rd generation polyamidoamine dendrimers to obtain (10)B concentrations necessary for BNCT by reducing the uptake of these c

First attempt of boron neutron capture therapy for adenoid cystic carcinoma liver metastasis: a case report and literature review.

This report details a 55-year-old male with liver metastases from Adenoid Cystic Carcinoma (ACC), enrolled in a study on Boron Neutron Capture Therapy (BNCT) utilizing Boronophenylalanine (BPA) as a carrier. The patient underwent two BNCT sessions. Within three months of the first BNCT, the tumor area remained stable, and pre-existing telangiectasia completely disappeared. No irreversible adverse effects of BNCT were observed. Despite ongoing dose exploration, this study highlights BNCT's therapeutic potential for ACC-related liver lesions.

Systemic tumor eradication by single-modality immunogenic neutron radiotherapy via size-controlled boron-10-enriched nanoparticles.

Photon-based radiotherapy is a mainstay of cancer treatment but rarely induces systemic tumor rejection alone. Boron neutron capture therapy (BNCT) is a promising alternative consisting of neutron radiation and boron drug administration. However, systemic antitumor response by BNCT typically requires additional immunomodulator and high boron drug dosages, raising safety concerns. Herein, we develop a low-dose nanoparticle (NP)-mediated immunogenic neutron radiotherapy to eradicate tumor systemically without external immunomodulator. To realize this, we synthesize poly(glycerol)-functionalized

First estimation of the clinical utility of boronotyrosine as a boron delivery compound for boron neutron capture therapy in head and neck cancer.

Boronotyrosine (BTS) is a candidate boron delivery compound for Boron Neutron Capture Therapy (BNCT). In comparison to the current standard, boronophenylalanine (BPA), BTS provides higher solubility and improved specificity, retention, and tumor control in small animal models. The purpose of this work is to estimate the advantages of BTS for BNCT of human head and neck cancer. Biodistributions of BTS and BPA were measured after bolus injection using a human hypopharyngeal cancer xenograft model in CB17 SCID mice. Pharmaco-kinetic (P-K) models were used to allow bolus response to be generalized

Maximizing nuclear capture and fission reactions: The innovative potential of boron delivery agents for neutron capture therapy in cancer treatment.

Neutron capture therapy (NCT) is a binary radiotherapeutic modality that relies on the nuclear capture and fission reactions of boron-10 for the selective destruction of cancer cells. Boron delivery agents (BDAs) are used to selectively deliver boron-10 to cancer cells, where it can be activated by neutron irradiation to produce highly cytotoxic alpha particles and recoiling lithium-7 nuclei that cause localized DNA damage and cell death. The development of BDAs for NCT has been an area of active research for several decades, with significant progress made in recent years. Polyhedral boranes,

Prolonged low-dose neutron irradiation induces late cellular and molecular impairments in the mice hippocampus and causes cognitive deficit.

Late effects of prolonged low-dose neutron exposure on the brain function remain poorly studied. We analyzed the content of neural progenitor cells, microglia and astrocytes, the number of PSD-95 puncta, the expression level of pro- and anti-inflammatory cytokines in the hippocampus, and long-term spatial and recognition memory in conventional (CV) and specific pathogen-free (SPF) mice 2 months after prolonged neutron irradiation at doses of 0.05 (1 day) and 0.5 Gy (10 day). Neurogenesis was intact in all mice. The level of PSD-95 was increased only in CV mice irradiated at 0.05 Gy. In CV mice

In silico assessments of the small molecular boron agents to pave the way for artificial intelligence-based boron neutron capture therapy.

Boron neutron capture therapy (BNCT) is a highly targeted, selective and effective technique to cure various types of cancers, with less harm to the healthy cells. In principle, BNCT treatment needs to distribute the 10boron (10B) atoms inside the tumor tissues, selectively and homogeneously, as well as to initiate a nuclear fission reaction by capturing sufficient neutrons which releases high linear energy particles to kill the tumor cells. In BNCT, it is crucial to have high quality boron agents with acceptable bio-selectivity, homogeneous distribution and deliver in required quantity, simil…
