The influence of traumatic intrusion of primary teeth on their permanent successors. A radiographic and histologic study in monkeys.

Intrusion of maxillary central primary incisors was performed in 18 green vervet monkeys (Cercopithecus aethiops) with the purpose of studying early and late histologic changes in odontogenesis of permanent tooth germs and the effect of treatment of primary tooth injury. Immediate changes were studied in five monkeys where both maxillary central primary incisors were intruded. The monkeys were sacrificed 10-15 min after intrusion. The immediate changes in the permanent tooth germ consisted of contusion and displacement of the reduced enamel epithelium and slight displacement of the hard dental…

Mandibular extracellular vesicles mediate morphogenesis and mineralization of tooth germs in miniature swine through the miR-206/HDAC4 signaling axis.

BACKGROUND: Reciprocal communication between odontogenic tissues underpins the complexity of tooth morphogenesis. Despite mandible serving a … the exosomal miR-206/HDAC4 signal axis redefines the mandible as an active epigenetic modulator of odontogenesis. This vesicle-mediated regulation enables long-range delivery of epigenetic effectors-revealing a paradigm shift in tooth development and a druggable target for tooth regeneration.

Ginsenoside Rb1 restores dental pulp by promoting odontogenic differentiation of dental pulp stem cells via the TGF-β/Smad signaling pathway.

BACKGROUND: Dental surgery is a common treatment for pulpitis, which can lead to decreased pulp vitality. A novel approach is needed to indu … ivity, and Alizarin Red S staining. RNA sequencing was performed to confirm the effects of G-Rb1 on odontogenesis of hDPSCs. The effects in the rat pulpotomy model were evaluated by examining reparative dentin formation using von Kossa staining. RESULTS: G-Rb1 was non-cytotoxic and significantly enhanced hDPSC migration and odontoblastic differentiation in hDP …

Double Teeth and Coexistent Anomalies: Examples From Continental Africa.

OBJECTIVES: Whether gemination or fusion, double teeth are rare worldwide, including Africa based on few published data. New cases from the … cisors are comparable to other global examples, indicative of common developmental processes during odontogenesis. Prevalence is lower than published modern rates, to suggest some exceptionality in Africans as reported earlier for other dental variants. Finally, though circumstantial, double teeth and accompanying anomalies may share an etiology. Continuing res …

Presence of dental signs of congenital syphilis in pre-modern specimens.

OBJECTIVE: Tooth morphology can vary due to genetic factors, infectious diseases and other environmental stresses. Congenital syphilis is known to interrupt tooth formation i.e. odontogenesis and amelogenesis, producing specific dental characteristics. Variation of those characteristics can occur, resulting in dental signs "not typical" of the disease, however, they are described in the 19th century literature. Past treatments of congenital syphilis with mercury also interrupted dental processes resulting in significantly different dental signs. The aim of this study is to examine the dentitio…

TBX1 protein interactions and microRNA-96-5p regulation controls cell proliferation during craniofacial and dental development: implications for 22q11.2 deletion syndrome.

T-box transcription factor TBX1 is the major candidate gene for 22q11.2 deletion syndrome (22q11.2DS, DiGeorge syndrome/Velo-cardio-facial syndrome), whose phenotypes include craniofacial malformations such as dental defects and cleft palate. In this study, Tbx1 was conditionally deleted or over-expressed in the oral and dental epithelium to establish its role in odontogenesis and craniofacial developmental. Tbx1 lineage tracing experiments demonstrated a specific region of Tbx1-positive cells in the labial cervical loop (LaCL, stem cell niche). We found that Tbx1 conditional knockout (Tbx1(cK…

Enhanced Wnt/β-catenin signalling during tooth morphogenesis impedes cell differentiation and leads to alterations in the structure and mineralisation of the adult tooth.

BACKGROUND INFORMATION: Previous studies have indicated that over-activation of the wingless interaction site (Wnt)/β-catenin signalling pat … s and led to a total lack of cell differentiation. Finally, over-activation of the Wnt route during odontogenesis resulted in adult teeth which presented altered size, morphology and mineralisation. CONCLUSIONS: Our results indicate that Wnt/β-catenin over-activation during tooth morphogenesis is sufficient to cause dramatic alterations in the adult tooth, by …

Daughters of the Enamel Organ: Development, Fate, and Function of the Stratum Intermedium, Stellate Reticulum, and Outer Enamel Epithelium.

The tooth enamel organ (EO) is a complex epithelial cell assembly involved in multiple aspects of tooth development, including amelogenesis. … Together, these studies indicate that the nonameloblast layers of the EO play multiple roles during odontogenesis, including the maintenance of several p63-positive stem cell reservoirs, a role during tooth root morphogenesis and tooth succession, a stabilizing function for the ameloblast layer, the facilitation of ion transport from the EO capillaries to the e …

Histopathological and Radiographic Analysis of Dental Follicle of Impacted Teeth Using Modified Gallego's Stain.

INTRODUCTION: In the WHO classification of odontogenic tumours, hard tissue formation has been considered as a sub-classification however, this parameter has not been much explored in dental follicle in literature. Epithelial-mesenchymal interactions play an important role in odontogenesis and its associated pathologies; therefore research on dental follicle should also include mesenchymal components along with epithelial components. Additionally, special stains to identify the nature of such depositions in dental follicle have been less explored. Modified Gallego's stain is such an example wh…

Second premolar agenesis is associated with mandibular form: a geometric morphometric analysis of mandibular cross-sections.

The aim of this study was to compare mandibular form (i.e., size and shape) between patients with agenesis of the lower second premolar (P2) … c pleiotropy (H3) as the most likely origin of the observed covariation between mandibular form and odontogenesis. Clinically, reduced dimensions and greater variability of mandibular form, as well as a pronounced submandibular fossa, should be expected during the treatment planning of patients with P2 agenesis.

Trauma-induced crown dilaceration of permanent maxillary central incisors: A case report with CBCT follow-up.

Traumatic dental injuries to primary teeth may result in developmental disturbances in the permanent successors because of the close anatomical relationship between primary tooth apices and developing permanent tooth germs. Injuries occurring during crown formation may interfere with normal odontogenesis and lead to crown dilaceration. A 7-year-old healthy boy presented with abnormal eruption of the permanent maxillary central incisors. At 2.5 years of age, he had sustained avulsion of both primary maxillary central incisors and crown fractures of the primary lateral incisors, followed by extr…

Early perturbation of Wnt signaling reveals patterning and invagination-evagination control points in molar tooth development.

Tooth formation requires complex signaling interactions both within the oral epithelium and between the epithelium and the underlying mesenchyme. Previous studies of the Wnt/β-catenin pathway have shown that tooth formation is partly inhibited in loss-of-function mutants, and gain-of-function mutants have perturbed tooth morphology. However, the stage at which Wnt signaling is first important in tooth formation remains unclear. Here, using an Fgf8-promoter-driven, and therefore early, deletion of β-catenin in mouse molar epithelium, we found that loss of Wnt/β-catenin signaling completely dele
