N6-methyladenosine (m6A) maintains maternal RNA stability in oocytes. One regulator of m6A, ALKBH5, reverses m6A deposition and is essential in RNA metabolism. However, the specific role of ALKBH5 in oocyte maturation remains elusive. Here, we show that Alkbh5 depletion causes a wide range of defects in oocyte meiosis and results in female infertility. Temporal profiling of the maternal transcriptomes revealed striking RNA accumulation in Alkbh5-/- oocytes during meiotic maturation. Analysis of m6A dynamics demonstrated that ALKBH5-mediated m6A demethylation ensures the timely degradation of m

Increases in delayed childbearing worldwide have elicited the need for a better understanding of the biological underpinnings and implications of age-related infertility. In women 35 years and older the incidences of infertility, aneuploidy, and birth defects dramatically increase. These outcomes are a result of age-related declines in both ovarian reserve and oocyte quality. In addition to waning reproductive function, the decline in estrogen secretion at menopause contributes to multisystem aging and the initiation of frailty. Both reproductive and hormonal ovarian function are limited by th

All the major components of the WNT signalling pathway are expressed in female germ cells and embryos. However, their functional relevance in oocyte biology is currently unclear. We examined ovaries collected from TCFGFP mice, a well-known Wnt reporter mouse model, and found dynamic changes in the Wnt/βcatenin signalling activity during different stages of oocyte development and maturation. To understand the functional importance of Wnt signalling in oocytes, we developed a mouse model with the germ cell-specific constitutive activation of βcatenin using cre recombinase driven by the DEAD (Asp

Eukaryotic DNA replication begins from multiple origins. The origin recognition complex (ORC) binds origin DNA and scaffolds assembly of a p … at the origins before activation of the pre-RC. H3.3 was also enriched at the origins during early oogenesis, raising the possibility that H3.3 bookmarks sites for later amplification. However, flies null mutant for both of the H3.3 genes in Drosophila did not have overt defects in developmental gene amplification or genomic replication, suggesting that H3 …

Diethylhexyl phthalate (DEHP) is an estrogen-like compound widely used as a commercial plasticizer and present in medical devices, tubing, f … F1 offspring at the adult age. These results show that DEHP may have a number of adverse effects on oogenesis, especially when exposure occurs during early postnatal age, arising concerns about the exposure of human female infants and children to this compound.

The relations between nutritional state and vitellogenic growth of oocytes were studied in 87 sexually mature female toads Bufo bufo during the postbreeding season before initiation of the next annual ovarian cycle. Recruitment of small oocytes to vitellogenic growth was correlated with resumption of food uptake and the restoration of a positive energy balance in the toads that had become depleted of energy reserves during hibernation and breeding. Treatment with exogenous gonadotropin (hCG) immediately initiated vitellogenic growth, independently of the nutritional state. But the hCG-induced

PURPOSE: N6-methyladenosine (m6A) is the most prevalent base epigenetic modification within eukaryotic mRNAs. It participates in post-transcriptional regulation, including maternal RNA maintenance and decay in mouse oocytes and during maternal-to-zygotic transition. The landscape in other mammalian species remains largely unexplored. The present work analyzed m6A dynamics in sheep cumulus oocyte complexes (COCs), during in vitro maturation. To explore potential relationships with oocyte developmental competence, a previously established model consisting of oocytes derived from adult and prepub

Infertility affects couples worldwide. Premature ovarian insufficiency (POI) refers to loss of ovarian function before 40 years of age and is a contributing factor to infertility. Several case studies have reported dominant-inherited POI symptoms in families with heterozygous EIF4ENIF1 (4E-T) mutations. However, the effects of EIF4ENIF1 haploinsufficiency have rarely been studied in animal models to reveal the underlying molecular changes related to infertility. Here, we demonstrate that Eif4enif1 haploinsufficiency causes mouse subfertility, impairs oocyte maturation and partially arrests ear

vasa gene encodes a highly conserved DEAD-box RNA helicase, required for germ cell development across animal kingdom. Vasa mutations cause male infertility in mammals. It has been widely used as a biomarker for studying animal fertility or manipulating germ cells in organisms. However, in reptilians, the functions of vasa gene involved in germ cell differentiation are largely unclear; this hampers the development of biological techniques and the improvement of the productivity in these species. Here a vasa cDNA was isolated in Chinese soft-shell turtle and it predicts a protein of 691 amino ac

Polycystic ovary syndrome (PCOS) is the most common cause of anovulatory infertility in women. Folliculogenesis in the PCOS ovary is often disrupted, leading to suboptimal oocyte competence for fertilization. This alteration in oocyte development is likely due to intrinsic molecular defects in the oocyte along with the state of androgen excess in PCOS patients. Many therapeutic options are available to infertile couples with PCOS, including controlled ovarian hyperstimulation and in vitro fertilization. However, because of immature oocytes, many patients are not able to conceive using standard

Human follicle development requires the recruitment of primordial follicles into a cohort of growing follicles from which one follicle is selected to ovulate a mature oocyte. During this developmental process, complex endocrine and intraovarian paracrine signals create a changing intrafollicular hormonal milieu. With this microenvironment, appropriate cumulus cell-oocyte signaling governs oocyte developmental competence, defined as the ability of the oocyte to complete meiosis and undergo fertilization, embryogenesis, and term development. Many of these mechanisms are perturbed in polycystic o

Heterochromatin and its core effectors from the Heterochromatin Protein 1 (HP1) family are essential for epigenetic silencing of repetitive regions and genome integrity from yeast to humans. However, how HP1s and associated factors regulate heterochromatin properties in vivo to sustain silencing across development, aging, and environmental factors, remains incompletely understood. Here, we identify Clump/CG30403 - a previously uncharacterized MADF-BESS domain protein - as a novel heterochromatin factor required for robust transposon silencing during Drosophila oogenesis and sustained fertility…

PURPOSE: Mitochondria are essential for oocyte maturation and early embryonic development, supplying ATP and maintaining mitochondrial DNA (mtDNA) integrity. During oogenesis, mtDNA undergoes dramatic amplification, but the mechanisms and functional significance of this process remain unclear. The purpose of this study was to elucidate the role of mitochondrial transcription factor A (TFAM) in mouse oocytes using an in vitro growth (IVG) system.
METHODS: Oocytes at different growth stages were analyzed for mtDNA copy number and expression of mitochondrial biogenesis genes. To assess TFAM func…

Pulsatile actomyosin networks emerge as a widespread mechanism driving tissue morphogenesis. Compared with myosin-II pulses, F-actin pulses are largely unexplored. By studying Drosophila oogenesis, we report that basal pulsatile actomyosin networks consist of F-actin bundle and meshwork architectures, implicating the existence of different F-actin microstructures and corresponding nucleators. Here, Rac1 and the Scar/WAVE-Arp2/3 complex (branched F-actin nucleator), exhibiting constant levels, are necessary to support F-actin pulsation, whereas pulses of Dia (unbranched F-actin nucleator) trigg…
