Updates on medical and surgical managements of diabetic retinopathy and maculopathy.

Diabetic retinopathy (DR) and diabetic macular edema (DME) are leading causes of vision loss globally. This is a comprehensive review focused on both medical and surgical management strategies for DR and DME. This review highlights the epidemiology of DR and DME, with a particular emphasis on the Asia-Pacific region, urban-rural disparities, ethnic variations, and grading methodologies. We examine various risk factors for DR, including glycemic control, hypertension, hyperlipidemia, obesity, chronic kidney disease, sex, myopia, pregnancy, and cataract surgery. Furthermore, we explore potential

Mitochondrial Energy Transformation Capacity Influences Brain Activation During Sensory, Affective, and Cognitive Tasks.

Brain function relies on energy supplied by mitochondrial energy transformation, but how cellular energetics constrains neurological function and cognition remains poorly understood. Genetic defects in mitochondrial DNA cause rare mitochondrial diseases (MitoD) that offer a unique window to examine how mitochondria affects the brain and cognition, and the possibility to identify neural processes that are most energetically constrained. In this study, we assessed functional magnetic resonance imaging (fMRI) on 29 participants with MitoD and 62 matched controls during resting state and tasks pro

Divergent Brain Network Activity in Asymptomatic C9orf72 and SOD1 Variant Carriers Compared With Established Amyotrophic Lateral Sclerosis.

Understanding the presymptomatic biology in those at high risk of developing amyotrophic lateral sclerosis (ALS) is essential for the development of preventative therapeutic interventions. Approximately 10% of ALS is associated with a C9orf72 expansion or pathogenic variants in SOD1. Magnetoencephalography (MEG), combined with machine learning algorithms, can model brain network dynamics in such at-risk populations to develop pathogenic biomarkers. Individuals with symptomatic ALS (symALS, n = 61), asymptomatic C9orf72 carriers (aC9, n = 16), or pathological SOD1 carriers (aSOD, n = 12), and h

Identification and Analysis of ZIC-Related Genes in Cerebellum of Autism Spectrum Disorders.

OBJECTIVE: Autism spectrum disorder (ASD) is a complex neurodevelopmental disorder with significant genetic heterogeneity. The ZIC gene family can regulate neurodevelopment, especially in the cerebellum, and has been implicated in ASD-like behaviors in mice. We performed bioinformatic analysis to identify the ZIC gene family in the ASD cerebellum. METHODS: We explored the roles of ZIC family genes in ASD by investigating (i) the association of ZIC genes with ASD risk genes from the Simons Foundation Autism Research Initiative (SFARI) database and ZIC genes in the brain regions of the Human Pr

Diagnostic accuracy of the point-of-care standard G6PD test™ (SD Biosensor) for glucose-6-phosphate dehydrogenase deficiency: a systematic review and meta-analysis.

BACKGROUND: Glucose-6-Phosphate Dehydrogenase deficiency (G6PDd) is a common genetic enzymopathy that can induce haemolysis triggered by various factors, including some anti-malarial drugs. Although many Point-of-Care (PoC) tests, such as Standard G6PD™ are available to detect G6PDd, its pooled diagnostic test accuracy (DTA) remains unknown. METHODS: To estimate the DTA of StandG6PD-BS at various thresholds of G6PDd, a systematic review with a DTA meta-analysis were conducted, searching EMBASE, MEDLINE, and SciELO databases up to April 4, 2024.The included studies were those that measured G6P

Severe cases of osteogenesis imperfecta type VIII due to a homozygous mutation in P3H1 (LEPRE1) and review of the literature.

BACKGROUND: Osteogenesis imperfecta (OI) is a genetic disorder that causes skeletal fragility, multiple fractures and several extraskeletal disorders. Most cases of OI are caused by mutations in COL1A1/A2. Osteogenesis imperfecta type VIII typically causes a severe and fatal phenotype that presents at birth with severe osteopenia, congenital fractures and other clinical manifestations. OBJECTIVES: We describe the cases of an 11-year-old female and a 9-year-old male with homozygous truncating mutations in P3H1. Both cases were born with intrauterine fractures and suffered multiple fractures sh

Optimal Regression Method for Near-Infrared Spectroscopic Evaluation of Articular Cartilage.

Near-infrared (NIR) spectroscopy has been successful in nondestructive assessment of biological tissue properties, such as stiffness of articular cartilage, and is proposed to be used in clinical arthroscopies. Near-infrared spectroscopic data include absorbance values from a broad wavelength region resulting in a large number of contributing factors. This broad spectrum includes information from potentially noisy variables, which may contribute to errors during regression analysis. We hypothesized that partial least squares regression (PLSR) is an optimal multivariate regression technique and

Isolation of camptothecin-sensitive chinese hamster cell mutants: phenotypic heterogeneity within the ataxia telangiectasia-like XRCC8 (irs2) complementation group.

Using a replica microwell method, four Chinese hamster lines which exhibit hypersensitivity to the topoisomerase I inhibitor camptothecin, designated CM1, CM2, CM3 and CM6, have been isolated. Their sensitivity towards camptothecin varied from 3.5- to 8.2-fold with relative sensitivity as follows: CM2 < CM3 < CM6 < CM1. Genetic analysis of the CM mutants has established that CM1, CM3 and CM6 fail to complement each other and can each be assigned to the irs2 (XRCC8) complementation group. The mutant CM2 could not be definitively assigned to a complementation group because it presented a semi-do

[Anesthesia and Angelman syndrome].

Angelman syndrome (AS) is a rare neurodevelopmental disorder with an incidence of 1:10,000-1:40,000 caused by deficient genetic imprinting in the chromosomal segment 15q11-q13. Experimental data suggest that the gamma-aminobutyric acid A (GABA(A)) receptor as well as the N-methyl-D-aspartate (NMDA) or α-amino-3-hydroxy-5-methyl-4-isoxazole proprionic acid (AMPA) receptors may be affected by this condition. The first description of the syndrome goes back to 1965 when the British pediatrician Harry Angelman (1915-1996) recognized similar clinical features in three children. Angelman's descriptio

High-Throughput Genomics Identify Novel FBN1/2 Variants in Severe Neonatal Marfan Syndrome and Congenital Heart Defects.

Fibrillin-1 and fibrillin-2, encoded by FBN1 and FBN2, respectively, play significant roles in elastic fiber assembly, with pathogenic variants causing a diverse group of connective tissue disorders such as Marfan syndrome (MFS) and congenital contractural arachnodactyly (CCD). Different genomic variations may lead to heterogeneous phenotypic features and functional consequences. Recent high-throughput sequencing modalities have allowed detection of novel variants that may guide the care for patients and inform the genetic counseling for their families. We performed clinical phenotyping for tw

Malaria Molecular Surveillance in the Peruvian Amazon with a Novel Highly Multiplexed Plasmodium falciparum AmpliSeq Assay.

Molecular surveillance for malaria has great potential to support national malaria control programs (NMCPs). To bridge the gap between research and implementation, several applications (use cases) have been identified to align research, technology development, and public health efforts. For implementation at NMCPs, there is an urgent need for feasible and cost-effective tools. We designed a new highly multiplexed deep sequencing assay (Pf AmpliSeq), which is compatible with benchtop sequencers, that allows high-accuracy sequencing with higher coverage and lower cost than whole-genome sequencin

Integrated Trajectory Planning, MEC Offloading, and Safety Coordination for Multi-UAV Disaster Response.

Rapid, reliable, and energy-efficient data collection is essential for disaster response, where terrestrial communication networks may be disrupted or unavailable. Unmanned Aerial Vehicles (UAVs) provide a flexible means of collecting critical sensing data, but their operation is constrained by limited onboard energy, stochastic wireless conditions, complex three-dimensional environments, and stringent latency requirements. This paper presents a structured multi-UAV framework that separates mission optimisation into spatial, temporal, and safety layers. In the spatial layer, a three-dimensiona

Do white matter microstructural alterations associated with childhood maltreatment explain psychopathology in early adulthood?-a longitudinal neuroimaging study.

The current study aims to identify tracts in which white-matter changes at age 19 interweave with different experiences of childhood abuse and neglect and whether these changes can explain psychopathology at ages 19 and 22. IMAGEN is a longitudinal, genetic, neuroimaging database in which community adolescents were followed from 14 to 22 years-old across eight sites in Europe. This study includes participants (n = 769, 55% females; Mage = 18.41 and 21.85) with high-quality DTI data who completed the Childhood Maltreatment Questionnaire and self-reported psychopathology symptoms at ages 19 and
