Conserved and divergent mitochondrial assemblies in kinetoplastid parasites.

The mitochondrial proteomes of Leishmania tarentolae and Trypanosoma brucei contain ∼1,700 proteins, most of which lack homologs in higher eukaryotes. Here, we integrate complexome profiling and cryo-electron microscopy to define conserved and lineage-specific macromolecular assemblies that support mitochondrial functions in these kinetoplastid protozoa. Comparative analyses reveal species- and life-stage-dependent differences in the abundance and composition of respiratory, metabolic, RNA processing, and other complexes, refining and expanding current annotations. Structures of L. tarentolae

Transgenic selection and underlying mechanisms in apicomplexan parasites.

The field of apicomplexan transgenics has been transformed by selectable markers that couple metabolism, drug action, and genetic engineering. This review consolidates three decades of progress across model parasites, Toxoplasma, Plasmodium, Cryptosporidium, Neospora, Eimeria, Babesia, and Theileria, illustrating how selection markers have enabled genome engineering. We dissect the rationale and mechanisms underlying positive and negative drug selections and emphasize marker recycling. Practical guidance is paired with failure diagnostics that determine whether transgenic selections succeed in

Shaping an image: 3D-printable models of parasites.

Parasites display many three-dimensional (3D) features that are related to their parasitic lifestyle. Using two-dimensional images alone, such 3D features are difficult to convey during teaching or public outreach. Thus, we provide parasite models for 3D printing to support education and to encourage adoption and adaptation by the parasitology community.

Calcified Neurocysticercosis: Understanding Dead (Not Necessarily Inactive) Parasites.

Neurocysticercosis (NCC) is the most common helminthic infection of the human central nervous system (CNS), and a major cause of acquired epilepsy in most of the world. Although outcomes of NCC have been improved by advanced diagnostic tests, antiparasitic drugs, and appropriate anti-inflammatory treatment, little attention has been paid to calcified NCC. Calcification is a common outcome in NCC and carries increased risk for seizures and hippocampal atrophy/sclerosis. The pathophysiological mechanisms leading to calcification instead of complete resolution are basically unknown. Understanding

Flavin-deficient erythrocytes offer protection against malaria parasites.

Studies from the 1980s and 1990s conducted in Italy, where malaria was once endemic, hypothesized that individuals with erythrocytes deficient in flavin mononucleotide and flavin adenine dinucleotide (FAD)-collectively known as flavins-are partially protected against malaria. The condition was reported to be familial, consistent with a genetic element. This hypothesis, however, has never been tested. Using an erythrocyte FAD-dependent glutathione reductase activity assay, we identified individuals with flavin-deficient erythrocytes (FDE) in Ferrara, Italy (23% of 150 individuals screened), and

Vesicular mechanisms of drug resistance in apicomplexan parasites.

SUMMARYVesicular mechanisms of drug resistance are known to exist across prokaryotes and eukaryotes. Vesicles are sacs that form when a lipid bilayer 'bends' to engulf and isolate contents from the cytoplasm or extracellular environment. They have a wide range of functions, including vehicles of communication within and across cells, trafficking of protein intermediates to their rightful organellar destinations, and carriers of substrates destined for autophagy. This review will provide an in-depth understanding of vesicular mechanisms of apicomplexan parasites, Plasmodium and Toxoplasma (that

Parasites and Microbiota: Dual Interactions and Therapeutic Perspectives.

The human gut hosts a diverse and active community of bacteria that symbiotically support the physiology, metabolism, and immunity of the intestinal lining. Nevertheless, a dynamic community of parasites (helminths and protozoa) may share a habitat with gut-dwelling microbiota. Both microbiota and parasites can significantly change the physical and immunological environment of the gut, thus generating several mechanisms of interaction. Studying this field is crucial for understanding the pathogenesis of parasitic diseases. Additionally, intestinal microbiota and gut-dwelling parasites may inte

Investigation of Parasites in Food Handlers in Turkey.

Increasing world population parallelly also brings an increase in food production and consumption. As food consumption increases, so do foodborne infections. In cases where adequate food safety and hygiene is not provided in places such as restaurants, dormitories, prisons, hospitals where mass feeding is made outside the home, many parasitic agents can be transmitted to people through food. People working in the food processing and distribution sector and who are in the position of porters play an important role in the spread of parasites, as they can transmit parasitic agents to food through

What's Eating You? Phlebotomine Sandflies and Leishmania Parasites.

Phlebotomine sandflies of the Phlebotomus and Lutzomyia genera are vectors for the spread of Leishmania parasites. Leishmaniasis is a parasitic infection most common in the Middle East and Central and South America. Few cases have been reported in the United States, with most patients presenting after returning from travel to other countries; however, the parasite has been locally acquired in Texas and Oklahoma. Clinical presentation varies depending on the species of Leishmania. There are 4 general clinical classifications of leishmaniasis: cutaneous, diffuse cutaneous, mucocutaneous, and vis

Emerging biology of noncoding RNAs in malaria parasites.

In eukaryotic organisms, noncoding RNAs (ncRNAs) have been implicated as important regulators of multifaceted biological processes, including transcriptional, posttranscriptional, and epigenetic regulation of gene expression. In recent years, it is becoming clear that protozoan parasites encode diverse ncRNA transcripts; however, little is known about their cellular functions. Recent advances in high-throughput "omic" studies identified many novel long ncRNAs (lncRNAs) in apicomplexan parasites, some of which undergo splicing, polyadenylation, and encode small proteins. To date, only a few of

The flexibility of Apicomplexa parasites in lipid metabolism.

Apicomplexa are obligate intracellular parasites responsible for major human infectious diseases such as toxoplasmosis and malaria, which pose social and economic burdens around the world. To survive and propagate, these parasites need to acquire a significant number of essential biomolecules from their hosts. Among these biomolecules, lipids are a key metabolite required for parasite membrane biogenesis, signaling events, and energy storage. Parasites can either scavenge lipids from their host or synthesize them de novo in a relict plastid, the apicoplast. During their complex life cycle (sex

Mussel memory: can bivalves learn to fear parasites?

Fear plays a crucial role in predator-prey interactions and can have cascading impacts on the structure of whole ecosystems. Comparable fear effects have recently been described for hosts and their parasites but our understanding of the underlying mechanisms remains limited by the lack of empirical examples. Here, we experimentally tested if bivalves Mytilus edulis can potentially 'learn to fear' the infective transmission stages (cercariae) of the trematode Himasthla elongata, and if experienced mussels change their parasite-avoidance behaviour accordingly. Our results show that previous expe

Nitro compounds against trypanosomatidae parasites: Heroes or villains?

Chagas disease and Human African trypanosomiasis (HAT) are caused by Trypanosoma cruzi, T. brucei rhodesiense or T. b. gambiense parasites, respectively; while Leishmania is caused by parasites from the Leishmania genus. In recent years, many efforts have been addressed to develop inhibitors against these parasites, especially nitro-containing derivatives, which can interfere with essential enzymes from the protozoa. In this review, all anti-trypanosomatidae nitrocompounds reported so far are shown herein, highlighting their activities and SAR analyses, providing all the benefits and problems

Erythroid cells and malaria parasites: it's a match!

PURPOSE OF REVIEW: The current review outlines recent discoveries on the infection of erythroid cells by Plasmodium parasites, focusing on the molecular interactions governing the tropism of parasites for their host cell and the implications of this tropism for parasite biology and erythroid cell maturation. RECENT FINDINGS: Although most studies about the interactions of Plasmodium parasites and their host cell focused on the deadliest human malaria parasite, Plasmodium falciparum, and the erythrocyte, there is increasing evidence that several Plasmodium species, including P. falciparum, als

Malaria parasites differentially sense environmental elasticity during transmission.

Transmission of malaria-causing parasites to and by the mosquito relies on active parasite migration and constitutes bottlenecks in the Plasmodium life cycle. Parasite adaption to the biochemically and physically different environments must hence be a key evolutionary driver for transmission efficiency. To probe how subtle but physiologically relevant changes in environmental elasticity impact parasite migration, we introduce 2D and 3D polyacrylamide gels to study ookinetes, the parasite forms emigrating from the mosquito blood meal and sporozoites, the forms transmitted to the vertebrate host

Diarrhoea associated with gastrointestinal parasites in grazing sheep.

Diarrhoea is a common, widespread and frustrating reality for sheep enterprises in most sheep producing regions globally and of particular concern in Australia as the major risk factor for breech flystrike. Parasitic disease has long been recognised as an important factor in diarrhoea in sheep, particularly the gastrointestinal nematodes (Trichostrongylus and Teladorsagia species). This review focuses on the role of parasitic infections in causing diarrhoea in sheep, with emphasis on the epidemiology of diarrhoea outbreaks related to worms and opportunities to manage the risk of diarrhoea outb
