Epigenetic Legacy: The Role of Sperm miRNAs in the Paternal Inheritance of Diabetes and Obesity Development.

In recent decades, obesity and diabetes have reached pandemic levels, with obesity now recognised as a major health risk factor. Evidence shows that metabolic diseases are more pronounced in the offspring of malnourished parents, suggesting that predisposition can be inherited via epigenetic information in gametes. This has sparked growing interest in small regulatory RNAs in sperm as carriers of epigenetic inheritance. However, the functional annotation of dysregulated sperm microRNAs (miRNAs) in obesity and diabetes remains limited. This work addresses this gap by analysing publicly availabl

Preconception paternal mental health history as predictor of antenatal depression in pregnant women.

BACKGROUND: Depression occurring during the perinatal period (PND) could affect both future mother and father. PND may lead to several adverse physical and mental health outcomes for the whole family. Several psychopathological determinants have been identified, even though few studies investigated the role of paternal mental health in the onset of maternal perinatal depression (MPND). Hence, a retrospective cohort study was carried out in order to investigate the relationship between paternal mental health and the occurrence of antenatal maternal depression as well as identifying potential so

The impact of childhood maltreatment on paternal antenatal bonding: the mediating role of antenatal depression.

BACKGROUND: As fathers become more involved in pregnancy and early parenting, more research attention has been given to their emotional connection with their unborn child. The transition to fatherhood is a critical developmental phase involving psychological and relational changes that shape paternal identity and bonding with the fetus. However, the factors influencing prenatal father-infant bonding remain unclear. This study examines whether emotional abuse and neglect experienced in childhood impact paternal bonding during pregnancy, and whether antenatal depressive symptoms mediate this rel

Advanced Paternal Age Impacts Common Loci in the Sperm and Placenta DNA Methylomes.

BACKGROUND: Epidemiological studies have reported an association between advanced paternal age at conception and an increased risk of neurodevelopmental disorders in offspring, such as autism spectrum disorder. Evidence suggests that DNA methylation alterations in spermatozoa of older men may be transmitted to the feto-placental unit and associated with offspring brain development and behavioral differences later in childhood. OBJECTIVE: We aimed to assess the association of advanced paternal age with DNA methylation alterations in the human placenta and compare the results to previous findin

Effects of anti-TNF agents on fertilization and pregnancy outcomes from a paternal perspective.

Anti-TNF drugs are effective in the treatment of inflammatory arthritis. However, concerns about the impact of anti-TNF therapy on fertility, particularly in male patients, have been insufficiently explored. This study aimed to investigate the effects of anti-TNFs on male fertility and examine the pregnancy outcomes of their partners. This study was conducted on male patients with inflammatory arthritis undergoing anti-TNF therapy. Male patients whose female partners became pregnant during the anti-TNF therapy were evaluated regarding their partners' pregnancy outcomes. The characteristics of

Paternal Cocaine Exposure and Its Testicular Legacy: Epigenetic, Physiological, and Intergenerational Consequences.

Cocaine use remains a major public health concern, with rising global prevalence and a well-established profile of neurotoxicity and addictive potential. While the central nervous system has been the primary focus of cocaine research, emerging evidence indicates that cocaine also disrupts male reproductive physiology. In the testis, cocaine alters the endocrine microenvironment, induces cell-specific damage, and disrupts spermatogenesis. Cocaine also interferes with epigenetic programming in germ cells and mature sperm, potentially leading to heritable epimutations. Epidemiology data reveal th

Struggles of Fatherhood: A Prospective Study on the Incidence of Paternal Postpartum Depression and Associated Factors.

AIM: To investigate the incidence and the factors associated with paternal postpartum depression amongst new fathers in Singapore. DESIGN: A prospective repeated measures study was conducted from September 2023 to March 2024. METHODS: A total of 200 fathers aged 21 years and above who had a neonate born at full term and no serious health events for both mother and neonate were recruited from a local hospital via convenience sampling. Fathers completed questionnaires at the third trimester of pregnancy (baseline) and 1 month postpartum. Multiple regression analyses were used to identify signi

Intergenerational influences of paternal combat-related trauma on offspring behavioral and brain function.

Post-traumatic stress (PTS) is a debilitating mental health condition that is highly prevalent in Veteran populations owing to their increased exposure to combat-related trauma. PTS is associated with numerous comorbid conditions including major depressive disorder, anxiety, and chronic pain. Although the causal mechanisms are currently unknown, paternal trauma has been linked to an increased risk for pathology in their offspring. Therefore, using a preclinical model of combat trauma, we examined the relationship between paternal PTS and offspring socio-emotional functioning, pain perception,

MicroRNA and Gut Microbiota Alter Intergenerational Effects of Paternal Exposure to Polyethylene Nanoplastics.

Nanoplastics (NPs), as emerging contaminants, have been shown to cause testicular disorders in mammals. However, whether paternal inheritance effects on offspring health are involved in NP-induced reproductive toxicity remains unclear. In this study, we developed a mouse model where male mice were administered 200 nm polyethylene nanoparticles (PE-NPs) at a concentration of 2 mg/L through daily gavage for 35 days to evaluate the intergenerational effects of PE-NPs in an exclusive male-lineage transmission paradigm. We observed that paternal exposure to PE-NPs significantly affected growth phen

Effect of paternal age on clinical outcomes of in vitro fertilization-embryo transfer cycles.

PURPOSE: This study aimed to investigate the impact of paternal age > 40 years on clinical pregnancy and perinatal outcomes among patients undergoing in vitro fertilization treatment. METHODS: We selected 75 male patients (aged > 40 years) based on predefined inclusion and exclusion criteria. Propensity score matching was performed in a 1:3 ratio, resulting in a control group (aged ≤ 40 years) of 225 individuals. Various statistical tests, including the Mann-Whitney U test, Chi-square test, Fisher's exact test, and binary logistic regression, were used to analyze the association between pater

Risk factors of paternal perinatal depression during the COVID-19 pandemic in Japan.

PURPOSE: The study aims to investigate risk factors of paternal perinatal depression during the COVID-19 pandemic. METHODS: We conducted an online cross-sectional study of 473 prenatal fathers and 1246 postnatal fathers in August 2021. We applied a modified Poisson regression to estimate relative risk ratios of possible factors for paternal perinatal depression (measured by Edinburgh Postnatal Depression Scale), sequentially introducing the following factors into the model: individual factors, interpersonal factors, obstetric/pediatric factors, and service utilization factors. RESULTS: Prena

Characterisation of the Paternal Influence on Intergenerational Offspring Cardiac and Brain Lipid Homeostasis in Mice.

There is growing evidence that poor paternal diet at the time of conception increase the risk of offspring developing a range of non-communicable metabolic diseases, such as obesity, diabetes and cardiovascular disease, in adulthood. We hypothesise that a paternal low protein-high carbohydrate diet perturbs offspring tissue lipid abundance through both sperm and seminal plasma-mediated mechanisms. To test our hypothesis, we fed male C57BL/6 mice either a control normal protein diet (NPD; 18% protein) or an isocaloric low protein diet (LPD; 9% protein) for a minimum of 8 weeks. We generated off

A posteriori parameters from paternity tests of a Mexican laboratory with the powerplex fusion system.

Population studies regarding Human identification (HID) systems report a priori forensic parameters, but rarely they describe a posteriori parameters from concluded paternity tests. We analyzed data from the PowerPlex® Fusion System in 1503 paternity tests from a Mexican laboratory for five years (2016-2020). The motherless duo paternity tests (89.8%) were more frequent than the standard trio tests (10.2%). A notable increase in motherless tests was noted regarding our previous report (89.8% vs 77.3%), probably explained by the COVID-19 pandemic. The estimated exclusion frequency in Mexico ran

Multiple paternity is related to adult sex ratio and sex determination system in reptiles.

The adult sex ratio (ASR, the proportion of males in the adult population) is an emerging predictor of reproductive behaviour, and recent studies in birds and humans suggest it is a major driver of social mating systems and parental care. ASR may also influence genetic mating systems. For instance male-skewed ASRs are expected to increase the frequency of multiple paternity (defined here as a clutch or litter sired by two or more males) due to higher rates of coercive copulations by males, and/or due to females exploiting the opportunity of copulation with multiple males to increase genetic di

Sex differences in paternal arsenic-induced intergenerational metabolic effects are mediated by estrogen.

BACKGROUND: Gene-environment interactions contribute to metabolic disorders such as diabetes and dyslipidemia. In addition to affecting metabolic homeostasis directly, drugs and environmental chemicals can cause persistent alterations in metabolic portfolios across generations in a sex-specific manner. Here, we use inorganic arsenic (iAs) as a prototype drug and chemical to dissect such sex differences. METHODS: After weaning, C57BL/6 WT male mice were treated with 250 ppb iAs in drinking water (iAsF0) or normal water (conF0) for 6 weeks and then bred with 15-week-old, non-exposed females for

Association of maternal and paternal perinatal depression and anxiety with infant development: A longitudinal study.

BACKGROUND: Maternal and paternal perinatal depression and anxiety are theorised to adversely impact infant development. Yet, few studies have assessed both mental health symptoms and clinical diagnoses within the one study. Moreover, research on fathers is limited. This study therefore aimed to examine the association between symptoms and diagnoses of maternal and paternal perinatal depression and anxiety with infant development. METHOD: Data were from the Triple B Pregnancy Cohort Study. Participants included 1539 mothers and 793 partners. Depressive and anxiety symptoms were assessed using

Maternal, but not paternal, ratings of child preinjury functioning predict child posttraumatic stress.

Pretrauma internalizing and externalizing behaviors have been found to predict posttraumatic stress disorder (PTSD) symptoms (PTSS) in children following injury. However, child in-hospital self-report of pretrauma symptoms may be impacted by their injury and associated medical care (e.g., sedation/pain medications). Parental report of child pretrauma risk factors may be easier to capture; however, mothers and fathers differ in the extent to which they report, and agree on, internalizing versus externalizing behaviors in their child. The present study examined the differential utility of matern
