Comparative transcriptomic profiling of high-yielding Saccharopolyspora pogona mutants generated by consecutive N-methyl-N'-nitro-N-nitrosoguanidine mutagenesis.

Butenyl-spinosyn is a highly effective macrolide insecticide produced by Saccharopolyspora pogona, but its low yield remains a major bottleneck for large-scale industrial application. In this study, a strategy based on consecutive N-methyl-N'-nitro-N-nitrosoguanidine (NTG) mutagenesis was employed, leading to the isolation of a genetically stable high-yielding mutant strain, ASAGF9-86, which produced 134.8 mg/L of butenyl-spinosyn, a 2.25-fold increase compared to the parental strains. Comparative transcriptomic analysis at two key fermentation stages revealed extensive transcriptional reprogr

The Warburg effect and beyond: Glycolytic reprogramming in cancer progression and emerging therapeutic strategies.

Metabolic reprogramming, especially the upretoning and functional reshaping of glycolysis, is the cornerstone of cancer progression, driving … chymal transition (EMT) and maintain the plurity of cancer stem cells (CSC). The cyclic flux of the pentose phosphate pathway (PPP) and the truncated tricarboxylic acid (TCA) cycle flux further highlight the metabolic flexibility inherent in malignant cells. Finally, we critically evaluated emerging treatment strategies for these glycolysis vulnerabilities, including inhib …

Insights into enhanced toxic effects by the binary mixture of carbendazim and procymidone on hepatic lipid metabolism in mice.

Carbendazim (CBZ) and procymidone (PRO) are two widely used fungicides in agriculture. However, there are still gaps in knowledge regarding … ifferential metabolites were discovered by metabolomics and enriched in different pathways, such as pentose phosphate pathway and purine metabolism. A unique effect, a decrease in glucose-6-phosphate (G6P) that promoted more NADPH production, was observed in the CBZ + PRO group. These results demonstrated that exposure to CBZ + PRO caused more serious lipid metabolism diso …

Chlamydia trachomatis genes whose products are related to energy metabolism are expressed differentially in active vs. persistent infection.

The Chlamydia trachomatis genome encodes glycolysis and pentose phosphate pathway enzymes, two ATP/ADP exchange proteins, and other energy transduction-related components. We asked if and when chlamydial genes specifying products related to energy transduction are expressed during active vs. persistent infection in in vitro models and in synovia from Chlamydia-associated arthritis patients. Hep-2 cells infected with K serovar were harvested from 0-48 h post-infection (active infection). Human monocytes identically infected were harvested at 1, 2, 3, 5 days post-infection (persistent). RNA from…

Gluconeogenesis using glycerol as a substrate in bloodstream-form Trypanosoma brucei.

Bloodstream form African trypanosomes are thought to rely exclusively upon glycolysis, using glucose as a substrate, for ATP production. Expanding the clinical and genetic spectrum of G6PD deficiency: The occurrence of BCGitis and novel missense mutation.

Glucose-6-phosphate dehydrogenase (G6PD) is a key enzyme in the pentose phosphate pathway that ensures sufficient production of coenzyme nicotinamide adenine dinucleotide phosphate (NADPH) by catalyzing the reduction of NADP+ to NADPH. Noteworthy, the latter mediates the production of reactive oxygen species (ROS) by phagocytic cells such as neutrophils and monocytes. Therefore, patients with severe forms of G6PD deficiency may present impaired NADPH oxidase activity and become susceptible to recurrent infections. This fact, highlights the importance to characterize the immunopathologic mechan…

Early strain-related changes in enzyme activity in osteocytes following bone loading in vivo.

The skeleton's architecture is matched to the changing loads to which it is subjected because mechanical loading directly or indirectly influences the activity of cell populations to deposit, maintain, or remove bone tissue as appropriate. This responsiveness to load bearing presupposes that certain cells are sensitive to load itself or to its consequences within the tissue. The nature of this effect and the cells concerned have not yet been determined. In this series of experiments, bones were exposed in vivo to a single short period of dynamic loading, which if repeated daily had been shown

Repressing malic enzyme 1 redirects glucose metabolism, unbalances the redox state, and attenuates migratory and invasive abilities in nasopharyngeal carcinoma cell lines.

A large amount of nicotinamide adenine dinucleotide phosphate (NADPH) is required for fatty acid synthesis and maintenance of the redox stat … ism of nasopharyngeal carcinoma(NPC) cells. High incorporation of glucose and an enhancement of the pentose phosphate pathway were observed in ME1-repressed cells. However, there were no obvious changes in the other two pathways for glucose metabolism: glycolysis and oxidative phosphorylation. Interestingly, NADPH was decreased under low-glucose condition in ME1-repressed …

Characterization of Plasmodium falciparum 6-Phosphogluconate Dehydrogenase as an Antimalarial Drug Target.

The enzyme 6-phosphogluconate dehydrogenase (6PGD) of the malaria parasite Plasmodium falciparum catalyzes the third step of the pentose phosphate pathway converting 6-phosphogluconate (6PG) to ribulose 5-phosphate. The NADPH produced by 6PGD is crucial for antioxidant defense and redox regulation, and ribose 5-phosphate is essential for DNA and RNA synthesis in the rapidly growing parasite. Thus, 6PGD represents an attractive antimalarial drug target. In this study, we present the X-ray structures of Pf6PGD in native form as well as in complex with 6PG or nicotinamide adenine dinucleotide pho…

Comparative lifespan trajectories of brain energy metabolism in human and macaque.

The brain's high energy demands are met by a suite of metabolic pathways that selectively appropriate glucose across development, yet how this metabolic strategy changes across the lifespan and whether it is conserved across species remains incompletely understood. We previously mapped five core energy metabolism pathways across the human cortex and lifespan, revealing a fundamental dichotomy between anabolic and energy-producing pathway expression: the pentose phosphate pathway, which contributes to biomass, peaks prenatally, while glycolysis, the tricarboxylic acid cycle, and oxidative phosp…

Biallelic Variants in KMO Cause a Novel Form of Congenital NAD Deficiency.

Congenital NAD deficiency disorder (CNDD) is a gene × environment disorder caused by disruptions of the kynurenine pathway. To date, CNDD ha … erformed in Kmo -/- embryos suggested that NAD + deficiency may perturb the pyrimidine, purine, and pentose phosphate pathways. These findings establish KMO deficiency as a new cause of CNDD and highlight a critical gene × environment interaction influencing NAD metabolism and congenital anomalies.

Metabolic profiling of retinal organoids reveals conserved core metabolites and alterations in glycolytic and amino acid metabolism.

INTRODUCTION: Retinal organoids have emerged as powerful models of retinal development and disease; however, the extent to which they recapitulate the metabolic landscape of the native retina remains largely unknown. OBJECTIVES: This study aimed to compare the metabolomic profiles of mouse retina and mouse embryonic stem cell-derived retinal organoids and to evaluate metabolic changes during organoid maturation. METHODS: Untargeted gas chromatography mass spectrometry based metabolomic profiling was performed on intracellular metabolites from mouse retina at postnatal day 3 and mouse retinal
