Organic derivatives of mercury and tin as promoters of membrane lipid peroxidation.

The toxicity mechanisms of mercury and tin organic derivatives are still under debate. Generally the presence of organic moieties in their molecules makes these compounds lipophilic and membrane active species. The recent results suggest that Hg and Sn compounds deplete HS-groups in proteins, glutathione and glutathione-dependent enzymatic systems; this process also results in the production of reactive oxygen species (ROS), the enhancement of membrane lipids peroxidation and damage of the antioxidative defence system. The goal of this review is to present recent results in the studies oriente

Monitoring hydrogen peroxide in the extracellular space of the brain with amperometric microsensors.

Interest in the detection of hydrogen peroxide in living brain tissue is growing for several reasons. Peroxide and other reactive oxygen species are implicated in neurodegenerative disorders and appear to have neuromodulatory functions in the brain. Also, there is a need to measure peroxide levels as a companion to measurements with amperometric sensors that rely on enzymes to generate peroxide for the detection of glutamate, choline, and glucose. Herein, we report on measurements performed in the brain of anesthetized rats with carbon fiber amperometric sensors coated with a cross-linked redo

Lipid peroxidation and antioxidant enzyme activities in experimental maxillary sinusitis.

The aims of this study were to assess whether th e increased oxidative stress of acute maxillary sinusitis is reflected by tissue lipid peroxidation and whether the activities of selected antioxidant enzymes are altered during inflammation of the maxillary sinus mucosa. Unilateral rhinosinusitis was induced in 8 rabbits by instillation of 0.2 ml of a killed suspension of Staphylococcus aureus into the right maxillary sinus cavity; control instillation of saline solution into the left maxillary sinus cavity of the same rabbits was also performed. At 7 days post-treatment, mucosal samples were e

Lipid peroxidation and erythrocyte antioxidant enzymes in patients with Behçet's disease.

In spite of unknown etiology, it is now accepted that reactive oxygen species (ROS) produced by neutrophils may be related to the pathogenesis of Behçet's Disease (BD). The objective was to investigate whether increased production of ROS may affect erythrocyte oxidant/antioxidant system in patients with BD. The levels of malondialdehyde (MDA), one of the end products of lipid peroxidation, in plasma and erythrocyte, and the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px), antioxidant enzymes, in erythrocyte, also C-reactive protein (CRP) and erythrocyte sedimentati

Antioxidant enzymes and lipid peroxidation in the scalp of patients with alopecia areata.

Alopecia areata (AA) is an autoimmune inflammatory disease. However, little is known about the alterations in lipid peroxidation and antioxidant enzymes in the scalp of patients with AA. Therefore, the aim of this study was to investigate the status of oxidative stress in the scalp of patients with AA. We measured the levels of thiobarbituric acid reactive substances (TBARS) as lipid peroxidation status, superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) as antioxidant enzymes in the scalp of ten patients with AA and ten control subjects. The levels of TBARS in scalp of patients wi

Comparison of hydrogen peroxide instillation with Goodsall's rule for fistula-in-ano.

BACKGROUND: Identification of the internal opening is an essential step in the management of fistula-in-ano. The predictive accuracy of Goodsall's rule is compared with instillation of hydrogen peroxide for fistulas-in-ano. METHODS: Thirty-five patients (32 male, three female; median age 42 years; range: 6 months-70 years) were studied. Hydrogen peroxide solution was injected into the external opening of the fistula track and effervescence was observed at the internal opening within the anal canal. A fistula track was either excised or incised. Setons were placed within high fistulas. RESULT

Normobaric hyperoxic stress in budgerigars: enzymic antioxidants and lipid peroxidation.

The effects of acute (3 h), repeated acute (3 exposures each of 3 h) and chronic (72 h) normobaric hyperoxic exposure in budgerigars (Melopsittacus undulatus) were evaluated by monitoring the effects on pulmonary enzymic antioxidants, and indicators of lipid peroxidation. All durations of oxygen exposure resulted in significant respiratory alkalosis and elevated pulmonary and blood glutathione peroxidase concentrations. The concentrations of other pulmonary enzymic antioxidants including glutathione reductase and superoxide dismutase were not significantly altered by oxygen exposure. Pulmonary

[Lipid peroxidation and antioxidant defenses in neonates treated with exogenous surfactants].

Certain parameters were studied associated with lipid peroxidation (LPO) and antioxidant defense (AOD) in those newborn infants having been treated with the exogenous surfactant Alveofact (Boehringer Ingelheim, Germany). Kept under medical surveillance were 60 preterm infants presenting with primary pulmonary pathology and severe respiratory insufficiency aged 35 hours old. Of these, ten babies (the main group) received two dosages of the above drug, with the mean age at administration having been 17.36 +/- 6.54 hours. The time-related course was studied of the following parameters--lipid pero

Vitamin E reduces lipid peroxidation in experimental hepatotoxicity in rats.

BACKGROUND AND AIMS: Lipid peroxidation is believed to be involved in the pathophysiology of a number of diseases and in the process of aging. This study investigates the effects of dietary supplementation with vitamin E (20 g/kg diet of all-rac-alpha-tocopheryl succinate for 3 weeks) on both non-enzymatic and enzymatic lipid peroxidation in experimental rats with carbon tetrachloride (CCl4)-induced hepatotoxicity (2.5 mL/kg body). METHODS: Plasma, urine and liver samples from control rats (n = 6), CCl4-treated rats (n = 6), and rats supplemented with vitamin E prior to CCl4 treatment (n = 8)

Enhanced hepatic lipid peroxidation in patients with primary biliary cirrhosis.

OBJECTIVE: The mechanisms responsible for hepatic injury have not been fully clarified in primary biliary cirrhosis (PBC). It has recently been suggested that hepatic lipid peroxidation may be involved in the pathogenesis of PBC. The aims of the current study were to determine whether patients with PBC have evidence of enhanced hepatic lipid peroxidation and to evaluate its relationship to clinicopathological features. METHODS: Immunohistochemical detection of 4-hydroxynonenal (HNE) protein adducts was performed in the liver biopsies of 20 patients with PBC. Histological stages of PBC were ev

Copper deficiency and cardiovascular disease: role of peroxidation, glycation, and nitration.

Dietary copper deficiency causes a variety of cardiovascular deficits. Systemic effects include high blood pressure, enhancement of inflammation, anemia, reduced blood clotting, and possibly arteriosclerosis. Effects on specific organs or tissues include weakened structural integrity of the heart and blood vessels, impairment of energy use by the heart, reduced ability of the heart to contract, altered ability of blood vessels to control their diameter and grow, and altered structure and function of circulating blood cells. In some instances, the cause of a defect can be directly attributed to

Acidic pH amplifies iron-mediated lipid peroxidation in cells.

The goal of our study was to investigate the mechanism by which changes in extracellular pH influence lipid peroxidation processes. Ferrous iron can react with hydroperoxides, via a Fenton-type reaction, to initiate free radical chain processes. Iron is more soluble at lower pH values, therefore we hypothesized that decreasing the environmental pH would lead to increased iron-mediated lipid peroxidation. We used Photofrin, a photosensitizer that produces singlet oxygen, to introduce lipid hydroperoxides into leukemia cells (HL-60, K-562, and L1210). Singlet oxygen reacts with the PUFA of cells

Vanadium catalyzed reduction of dioxygen to hydrogen peroxide: an oscillating process.

In acid isopropanol/water solution and aerobic conditions, (Bu)4N+VO3- in the presence of an initial amount of H2O2, catalyzes the autoxidation of isopropanol to acetone and the contextual dioxygen reduction to hydrogen peroxide, which accumulates in solution. We have observed that, in the system under examination, the build-up of H2O2 concentration shows an oscillatory behavior. Speciation of the peroxovanadium complexes in iPrOH/H2O has been explored with the combined use of 51V NMR, UV-Vis and ESI-MS techniques.

Mechanisms of hydrogen peroxide-induced calcium dysregulation in PC12 cells.

The mechanisms of H(2)O(2)-induced elevated calcium baselines in PC12 cells were investigated in the present study by using fura-2-fluorescent image analysis. The results showed that the calcium comes from both intracellular and extracellular sources. Although the major intracellular source was mitochondria, only the extracellular calcium influx was responsible for the sustained post-H(2)O(2)-exposure increases. This calcium influx was partially blocked by calcium channel antagonists [verapamil (L-type) or mibefradil (nonselective)] and was more effectively blocked by the sodium channel antago

Expired hydrogen peroxide in breath condensate of cystic fibrosis patients.

Stimulated inflammatory cells release large amounts of hydrogen peroxide (H2O2). Breath condensate H2O2 has been shown to be elevated in stable asthmatic children, chronic obstructive pulmonary disease and intubated adult respiratory distress syndrome. In cystic fibrosis airways, where neutrophilic inflammation dominates, it is postulated that H2O2 in breath condensate would be elevated and may be used as a marker of airways inflammation. Expired breath condensate was collected from 16 clinically stable cystic fibrosis (CF) patients (mean age 25.3 yrs, mean forced expiratory volume in one seco

Contrasting roles for nitric oxide and peroxynitrite in the peroxidation of myelin lipids.

Peroxynitrite is formed by the reaction of nitric oxide (NO) and superoxide. Since widespread peroxynitrite activity was observed during experimental allergic encephalomyelitis (EAE), the effect of this strong lipid-peroxidizing agent on myelin integrity was examined. Incubation of myelin suspensions with the peroxynitrite donor 3-morpholinosydnonimine (SIN-1) resulted in the formation of the lipid peroxidation product, malondialdehyde (MDA). MDA formation was inhibited in the presence of butylated hydroxytoluene, which interrupts the progression of the lipid peroxidation chain reaction. Super

Reactivity of biologically important thiol compounds with superoxide and hydrogen peroxide.

The reactivities of glutathione, cysteine, cysteamine, penicillamine, N-acetylcysteine, dithiothreitol and captopril with superoxide generated from xanthine oxidase and hypoxanthine, and with reagent hydrogen peroxide, have been investigated. Rates of thiol loss on adding hydrogen peroxide, and superoxide-dependent thiol loss and oxygen uptake were measured. The relative reactivities of the different thiols with both oxidants were inversely related to the pK of the thiol group, such that at pH 7.4, penicillamine was the most reactive. N-acetylcysteine weakly reactive and no reaction was seen w

Effects of the location of distal histidine in the reaction of myoglobin with hydrogen peroxide.

To clarify how the location of distal histidine affects the activation process of H2O2 by heme proteins, we have characterized reactions with H2O2 for the L29H/H64L and F43H/H64L mutants of sperm whale myoglobin (Mb), designed to locate the histidine farther from the heme iron. Whereas the L29H/H64L double substitution retarded the reaction with H2O2, an 11-fold rate increase versus wild-type Mb was observed for the F43H/H64L mutant. The Vmax values for 1-electron oxidations by the myoglobins correlate well with the varied reactivities with H2O2. The functions of the distal histidine as a gene
