Laboratory and field evaluation of neem (Azadirachta indica A. Juss) and Chinaberry (Melia azedarach L.) oils as repellents against Phlebotomus orientalis and P. bergeroti (Diptera: Psychodidae) in Ethiopia.

The study evaluated the efficacy of neem (Azadirachta indica A. Juss.) and Chinaberry (Melia azedarach L.) seed oils as repellents against laboratory and field populations of some sandflies in Ethiopia. In the laboratory, concentrations of 2% and 5% neem oil in coconut oil tested against Phlebotomus orientalis (vector of visceral leishmaniasis) provided 96.28% (95% CI=95.60-96.97) protection up to a mean time of 7h and 20 min and 98.26% (95% CI=93.46-104. 07) protection up to 9h, respectively. Similarly, M. azedarach oil at 2% concentration produced 95.13% (95% CI=90.74-99.52) protection for t

Expression pattern of ARG1 and iNOS genes in macrophages of Rhombomys opimus, BALB/c and C57BL/6 mice exposed to Leishmania major and salivary gland homogenates of Phlebotomus papatasi.

Early interactions between Leishmania-macrophages (MQ) of host and effect of sand fly saliva are central to leishmaniasis outcome. Macrophages are able to kill or act as long-term hosts of parasite depending on host immunity. It proved that immunogenic proteins in sand fly saliva mostly have an exacerbating effect on leishmaniasis by up-regulating cytokines. We have explored expression of Arginase 1 (ARG1) and inducible Nitric Oxide Synthase (iNOS) genes in macrophages of three different rodents, BALB/c (susceptible), C57BL/6 (resistant) and Rhombomys opimus (R. opimus, natural reservoir) in p

Altitudinal cross-sectional study of the sand fly fauna of the highlands of Tigray Region, northern Ethiopia.

BACKGROUND: Cutaneous leishmaniasis (CL), caused by Leishmania aethiopica and transmitted by Phlebotomus longipes, is endemic in northern Ethiopia. However, sand fly fauna is poorly documented in the northern Tigray region. High-altitude CL cases have also been observed, but their local acquisition is uncertain. To identify the faunal composition, their geographical and altitudinal distribution, cross-sectional surveys were conducted in ten villages (average altitude range: 2411 - 3186 meters) with confirmed CL cases across four endemic districts of the region. A total of 170 light trap-nights

Presence of localized elevated metabolic enzymes and kdr mutations in the voltage-gated sodium channel gene indicate early evidence of resistance of Phlebotomus argentipes to alpha-cypermethrin in Bihar, India.

BACKGROUND: Visceral leishmaniasis, caused by Leishmania donovani and transmitted by Phlebotomus argentipes, remains a major public health challenge in the Indian subcontinent. Sand fly populations are controlled by using different insecticides, particularly dichlorodiphenyltrichloroethane (DDT) and pyrethroids such as alpha-cypermethrin. Prolonged and irregular use of these insecticides raises concern about the development of resistance in sand fly populations. METHODS: This study aimed to compare metabolic detoxification and knockdown resistance (kdr) mutations in Vgsc and ace-1 between nin

Predicting the Distribution of Phlebotomus papatasi (Diptera: Psychodidae), the Primary Vector of Zoonotic Cutaneous Leishmaniasis, in Golestan Province of Iran Using Ecological Niche Modeling: Comparison of MaxEnt and GARP Models.

Zoonotic cutaneous leishmaniasis (ZCL) is a prevalent vector-borne disease in the Golestan province of Iran, with Phlebotomus papatasi (Scopoli, 1786) serving as the main vector. The aim of this study was to model the probability of presence of this species in the study area, and to determine the underlying factors affecting its distribution. Three villages were selected from each county of the province and visited monthly for investigating ZCL. Sticky paper traps were used for collecting the sand flies to determine the species present. The presence of Ph. papatasi was modeled using genetic al

Widespread transmission in diverse ecotypes challenges visceral leishmaniasis control in East Africa.

East Africa is emerging as the global hot spot of visceral leishmaniasis (VL), yet efforts to eliminate it are hindered by substantial knowledge gaps in its ecoepidemiology. Here, we report on the high prevalence of Leishmania infection in Phlebotomus orientalis in Marsabit county, Kenya (3.9%), and Gedaref state, Sudan (3.6%), where this species comprised 99.8% (n = 1185) and 100% (n = 1350) of captured Phlebotomus females, respectively. In Aba Roba, Ethiopia, Phlebotomus martini accounted for 94.4% of 184 collected Phlebotomus females and had a lower infection rate of 1.5%. Phlebotomus orien

A single oral dose of fluralaner (Bravecto®) in dogs rapidly kills 100% of blood-fed Phlebotomus perniciosus, a main visceral leishmaniasis vector, for at least 1 month after treatment.

Dogs are the reservoir host of zoonotic visceral leishmaniasis (VL) caused by Leishmania infantum (Kinetoplastida: Trypanosomatidae). Both subclinically-infected and sick animals can be infectious to competent phlebotomine vectors. The degree and duration of insecticidal efficacy of an oral dose of fluralaner (Bravecto®; Merck Animal Health) was determined in dogs exposed to bites of Phlebotomus perniciosus (Diptera: Psychodidae), a main Mediterranean vector of VL. Twelve dogs allocated to two groups of six animals each were included in a parallel-group designed, negative-controlled, randomize

Environmental factors underlying spatial patterns of sand flies (Diptera: Psychodidae) associated with leishmaniasis in southern Sinai, Egypt.

Although Leishmania major is endemic in parts of the Sinai of Egypt, the ecology and distribution of Leishmania sand fly vectors in southern Sinai has not been well characterized. Accordingly, additional sand fly samples were obtained at 41 sites in the southern Sinai region during 1996-1997, and analyzed to improve the characterization of risk of sand fly-borne pathogens. Using a Geographic Information System (GIS), species-specific spatial distributions that might suggest zoonotic cutaneous leishmaniasis (ZCL) risk areas were determined in relation to contextual environmental factors, includ

Cloning, high-level gene expression and bioinformatics analysis of SP15 and LeIF from Leishmania major and Iranian Phlebotomus papatasi saliva as single and novel fusion proteins: a potential vaccine candidate against leishmaniasis.

BACKGROUND: Early exacerbation of cutaneous leishmaniasis is mainly affected by both the salivary and Leishmania parasite components. Little is known of the vaccine combination made by immunogenic proteins of sandfly saliva (SP15) with Leishmania parasites (LeIF) as a single prophylactic vaccine, namely SaLeish. Also, there are no data available to determine the species-specific sequence of SP15 isolated from the Iranian Phlebotomus papatasi. METHODS: Integrated bioinformatics and genetic engineering methods were employed to design, optimize and obtain a vector-parasite-based vaccine formulat

Three strains of Wolbachia pipientis and high rates of infection in Iranian sandfly species.

Individual wild-caught sandflies from Iran were examined for infections of Wolbachia pipientis by targeting the major surface protein gene wsp of this intracellular α-proteobacterium. In total, 638 male and female sandflies were screened, of which 241 were found to be positive for one of three wsp haplotypes. Regardless of geographical origins and habitats, Phlebotomus (Phlebotomus) papatasi and other sandfly species were found to be infected with one common, widespread strain of A-group W. pipientis (Turk 54, GenBank accession EU780683; AY288297). In addition, a new A-group haplotype (Turk07,
