Insights into the Biocontrol Function of a Burkholderia gladioli Strain against Botrytis cinerea.

Pathogenic fungi are the main cause of yield loss and postharvest loss of crops. In recent years, some antifungal microorganisms have been e … table grapes against gray mold disease caused by Botrytis cinerea. Furthermore, KRS027 can trigger plant immunity by inducing systemic resistance (ISR) activated by salicylic acid- (SA), jasmonic acid- (JA), and ethylene (ET)-dependent signaling pathways. The extracellular metabolites and volatile organic compounds (VOCs) of KRS027 affected the colony extension …

Medicinal Plants and Lead Phytomolecules as Immunomodulators: An Updated Review.

All therapeutic interventions aimed at modulating the immune response to pathogens, self-antigens, carcinogens, or xenogeneic antigens are referred to as immunomodulation, which either prevents hyperactivation or restores the appropriate response of the immune system. Since antiquity, medicinal plants have been used as a source of immune-boosting medicines with fewer side effects. These medicinal plants, for example, Curcuma longa, Camellia sinensis, Artemisia annua, and Andrographis paniculata, have been used to treat immune system disorders such as multiple sclerosis, psoriasis, lupus, and o

The cloak, dagger, and shield: proteases in plant-pathogen interactions.

Plants sense the presence of pathogens or pests through the recognition of evolutionarily conserved microbe- or herbivore-associated molecular patterns or specific pathogen effectors, as well as plant endogenous danger-associated molecular patterns. This sensory capacity is largely mediated through plasma membrane and cytosol-localized receptors which trigger complex downstream immune signaling cascades. As immune signaling outputs are often associated with a high fitness cost, precise regulation of this signaling is critical. Protease-mediated proteolysis represents an important form of pathw

Blue Light Receptor PHR2-Mediated Spectrum Antiviral Immunity.

Plant viruses have evolved sophisticated virulence strategies to counteract host antiviral defenses. Blue light, as an important environmental signal, can regulate plant growth and tolerance to biological stress. Nevertheless, how blue light regulates plant responses to antiviral immunity remains largely obscure. Here, we demonstrate that blue light suppresses rice black-streaked dwarf virus (RBSDV) replication. The blue light receptor ZmPHR2 functions as a key immune component, positively regulating maize resistance to RBSDV. Mechanistically, ZmPHR2 interacts with ZmISP in chloroplasts to tri

PIF4 enhances temperature-independent tolerance to TSWV in Nicotiana benthamiana.

Tomato spotted wilt virus (TSWV) significantly threatens agricultural productivity, causing significant production and yield losses. Higher temperature have been reported to further intensify the TSWV accumulation and disease severity in several crops. PHYTOCHROME-INTERACTING FACTOR 4 (PIF4) has emerged as a key regulator of plant responses to multiple biotic and abiotic stresses; however, its precise role in plant immunity against TSWV has not been defined. To investigate this, we generated PIF4 knockout mutants and PIF4-overexpression lines of Nicotiana benthamiana by gene-editing and transg…

Apoplast multi-omics profiling during fungal infection uncovers new players of basal and early induced immunity in Arabidopsis thaliana.

The extracellular space in plant tissues, known as the apoplast, remains one of the least characterized cellular compartments. The apoplastic fluid, akin to mammalian extracellular fluid, serves as the primary interface between pathogens and their host. It allows the exchange of signalling molecules, coordinates host cell responses and enables the circulation of pathogen effectors that modulate the immune response. We describe here a comprehensive multi-omics analysis of the apoplastic washing fluids content just 6 h after the onset of Arabidopsis thaliana infection with Botrytis cinerea, a fa

DIMBOA and melatonin mediate a recruitment feedback loop between wheat and Bacillus pseudomycoides to alleviate herbicide stress.

Herbicide residues disrupt soil-plant-microbe interactions and threaten agricultural sustainability, yet how plants actively recruit beneficial microbes to mitigate stress remains unclear. Here, we uncover a metabolite-mediated recruitment model in which wheat secretes DIMBOA and melatonin in response to bensulfuron-methyl (BM) stress to recruit Bacillus pseudomycoides A3, with cross-sectional evidence supporting a self-reinforcing feedback architecture. Chemotaxis assays confirmed that DIMBOA and melatonin act as potent chemoattractant for strain A3 at environmentally relevant concentrations.

Immunogenicity of Brugia malayi Abundant Larval Transcript-2, a potential filarial vaccine candidate expressed in tobacco.

Transgenic tobacco plants with Bm ALT-2, a filarial vaccine candidate, were developed. The plant-produced antigen showed immunogenicity on par with the E.coli product. Transgenic tobacco plants were developed using Brugia malayi Abundant Larval Transcript-2 (Bm ALT-2), a major antigen produced from recombinant E.coli found to be experimentally successful as potential vaccine candidate against lymphatic filariasis. Results of experiments on the transformation and expression of the Bm ALT-2 in tobacco plant to produce plant-based vaccine are presented here. We have successfully transformed the t

Volatile Organic Compounds Emitted by Fungi Have Substantial Effects on Growth and Disease Resistance in Arabidopsis thaliana.

Volatile organic compounds (VOCs) emitted by certain fungi have previously been suggested to modulate plant development and their response to stresses. While some fungal species, such as Trichoderma harzianum, are known to produce VOCs, the production of VOCs in other phylogenetically divergent species, such as Mucor mucedo, was previously unknown. This study aimed to analyse the VOCs profiles emitted by these fungal species and examine their effects on the growth and defensive responses of Arabidopsis thaliana. Volatilome analysis revealed that the most abundant VOCs for both species were alc

High air humidity dampens salicylic acid pathway and NPR1 function to promote plant disease.

The occurrence of plant disease is determined by interactions among host, pathogen, and environment. Air humidity shapes various aspects of plant physiology and high humidity has long been known to promote numerous phyllosphere diseases. However, the molecular basis of how high humidity interferes with plant immunity to favor disease has remained elusive. Here we show that high humidity is associated with an "immuno-compromised" status in Arabidopsis plants. Furthermore, accumulation and signaling of salicylic acid (SA), an important defense hormone, are significantly inhibited under high humi…

Impairment of the Zn/Cd detoxification systems affects the ability of Salmonella to colonize Arabidopsis thaliana.

Salmonella capacity to colonize different environments depends on its ability to respond efficiently to fluctuations in micronutrient availability. Among micronutrients, Zn, besides playing an essential role in bacterial physiology, is a key element whose concentration can influence bacterial survival in a particular niche. Plant colonization by Salmonella enterica was described for several years, and some molecular determinants involved in this host-pathogen interaction have started to be characterized. However, it is still unclear if Zn plays a role in the outcome of this interaction, as wel

Botrytis cinerea-induced F-box protein 1 enhances disease resistance by inhibiting JAO/JOX-mediated jasmonic acid catabolism in Arabidopsis.

Jasmonic acid (JA) is a crucial phytohormone that regulates plant immunity. The endogenous JA level is determined by the rates of its biosynthesis and catabolism in plants. The activation of JA biosynthesis has been well documented; however, how plants repress JA catabolism upon pathogen infection remains elusive. In this study, we identified and characterized Botrytis cinerea-induced F-box protein 1 (BFP1) in Arabidopsis. The expression of BFP1 was induced by B. cinerea in a JA signaling-dependent manner, and BFP1 protein was critical for plant defense against B. cinerea and plant response to…

Plant secondary metabolites against biotic stresses for sustainable crop protection.

Sustainable agriculture practices are indispensable for achieving a hunger-free world, especially as the global population continues to expand. Biotic stresses, such as pathogens, insects, and pests, severely threaten global food security and crop productivity. Traditional chemical pesticides, while effective, can lead to environmental degradation and increase pest resistance over time. Plant-derived natural products such as secondary metabolites like alkaloids, terpenoids, phenolics, and phytoalexins offer promising alternatives due to their ability to enhance plant immunity and inhibit pest …

The Effector RipAW Enhances Ralstonia solanacearum Invasion in Arabidopsis via CBP60g/SARD1-Dependent and -Independent Pathways.

CaM-binding Protein 60-like G (CBP60g) and Systemic Acquired Resistance Deficient 1 (SARD1) are key immune signalling regulators that redundantly promote salicylic acid (SA) biosynthesis and plant immunity. Pathogen effectors often target these immune nodes to suppress plant defence. However, the role of bacterial effectors in disabling CBP60g and SARD1 to increase plant susceptibility remains unclear. In this study, we show that RipAW, an E3 ligase effector from Ralstonia solanacearum, induces root architecture changes and enhances plant susceptibility to R. solanacearum in Est::RipAW transge…

Plant Defense Networks against Insect-Borne Pathogens.

Upon infection with insect-borne microbial pathogens, plants are exposed to two types of damage simultaneously. Over the past decade, numerous molecular studies have been conducted to understand how plants respond to pathogens or herbivores. However, investigations of host responses typically focus on a single stress and are performed under static laboratory conditions. In this review, we highlight research that sheds light on how plants deploy broad-spectrum mechanisms against both vector-borne pathogens and insect vectors. Among the host genes involved in multistress resistance, many are inv

Salicylic Acid Steers the Growth-Immunity Tradeoff.

Plants possess an effective immune system to combat most microbial attackers. The activation of immune responses to biotrophic pathogens requires the hormone salicylic acid (SA). Accumulation of SA triggers a plethora of immune responses (like massive transcriptional reprogramming, cell wall strengthening, and production of secondary metabolites and antimicrobial proteins). A tradeoff of strong immune responses is the active suppression of plant growth and development. The tradeoff also works the opposite way, where active growth and developmental processes suppress SA production and immune re
