Bioactive diphenyl ether derivatives from a gorgonian-derived fungus Talaromyces sp.

Three new diphenyl ether derivatives, talaromycins A-C (1-3, resp.), together with six known analogs, 4-9, were isolated from a gorgonian-derived fungus, Talaromyces sp. The structures of the new compounds were determined by analysis of extensive NMR spectroscopic data. All of the isolated metabolites, 1-9, were evaluated for their cytotoxic and antifouling activities. Compound 4 exhibited pronounced cytotoxicity against the tested human cell lines with the IC50 values ranging from 4.3 to 9.8 μM. Compounds 3, 5, 8, and 9 showed potent antifouling activities against the larval settlement of the

Micafungin exposure drives multidrug resistance in Clavispora lusitaniae.

Fungal infections are an escalating global health concern, with rare Candida species posing an urgent threat due to emerging multidrug resistance. Clavispora (Candida) lusitaniae is an uncommon pathogen in which multidrug resistance has been documented during antifungal therapy, yet the selective forces driving this phenotype remain unclear. Here, we show that exposure to the echinocandin micafungin (MCF) alone can select for multidrug resistance in C. lusitaniae. Through controlled evolution experiments we identified individual point mutations in genes encoding ergosterol biosynthesis enzymes

Overcoming Antifungal Resistance: A Histatin 5-Gated Nanodevice for On-Demand Surfactant Delivery against C. albicans Infections.

The escalating incidence of fungal infections, coupled with the emergence of antifungal resistance, represents a formidable global public health challenge. Candida albicans is a common opportunistic fungus that causes life-threatening, widespread candidiasis, especially in immunocompromised patients. Conventional antifungals, including azoles, echinocandins, and polyenes, have some drawbacks, such as host toxicity and the rapid appearance of resistant fungal strains. To overcome these limitations, we engineered a stimuli-responsive gated antimicrobial nanoparticle using mesoporous silica nanop

Antifungal repurposing of clinical chelator DMSA against Cryptococcus neoformans via laccase and histidine biosynthesis inhibition.

As a globally distributed opportunistic fungus, Cryptococcus neoformans poses a significant threat to human health by causing fatal meningitis, which is associated with high mortality and morbidity rates. Current antifungals-polyenes, flucytosine, and azoles-are limited by toxicity and resistance, underscoring the need for new agents. This study evaluated dimercaptosuccinic acid (DMSA), a chelating agent used in the treatment of heavy metal poisoning, beginning with its effect on the virulence factor laccase. DMSA showed broad-spectrum activity against standard (H99, B3501) and clinical isolat

Identification, purification and functional characterization of a thermostable marine chitinase for potential fungal control via chitin degradation mechanism.

The growing prevalence of treatment-resistant Candida species highlights an urgent need for innovative antifungal therapies. The current range of antifungals, limited to polyenes, azoles, and echinocandins, are becoming insufficient due to the rise of resistance, including cross-resistance among fungal strains. Marine environment is an underexplored reservoir of unique enzymes which can be extremophilic. This study presents the cloning and expression of a chitinase gene from the bacterium Bacillus thuringiensis (BtChi), expressed in an E. coli system, yielding a protein with a molecular weight

Innovative antifungal strategies to combat drug-resistant Candida auris: recent advances and clinical implications.

Candida auris is an invasive fungal pathogen recognized globally as a significant health threat due to its marked resistance to multiple classes of antifungal agents, including azoles, echinocandins, and polyenes. The associated high morbidity and mortality rates present considerable public health challenges. Research efforts have largely focused on understanding the molecular mechanisms of antifungal resistance and developing alternative therapies to counteract this issue. This review summarizes current advances in the identification of natural antifungal compounds, the development of novel s

Skin's 'nano-shield' hydrogel: Co-delivery of myricetin-laurate nanoagents eliminates fungus and promotes skin regeneration.

Cutaneous fungal infections, particularly those caused by Candida albicans, pose significant clinical challenges due to high recurrence rates and rising antifungal resistance. Conventional therapies, including azoles and polyenes, are limited by toxicity, resistance, and cost. This study addresses these limitations by developing a multifunctional hydrogel integrating lauric acid (LA)-a natural antifungal anionic surfactant-with the antioxidant plant polyphenol myricetin (Myr). The synergistic combination of LA and Myr disrupts fungal membrane integrity through lipid bilayer destabilization. To

Antifungal pipeline: New tools for the treatment of mycoses.

Fungal infections are becoming an escalating public health challenge, particularly among immunocompromised individuals. The partially limited efficacy of current antifungal treatments, their potential adverse effects, and the increasing problem of resistance emphasize the need for new treatment options. Existing antifungal classes-allylamines, azoles, echinocandins, polyenes, and pyrimidine analogs-face challenges due to their similarity with human cells and rising resistance. New antifungal agents, such as ibrexafungerp, rezafungin, oteseconazole, and miltefosine, offer novel mechanisms of ac

Investigating inhibitory effect of sterol targeting compounds against B. anthracis: Membrane microdomain a probable target.

PURPOSE: This study aimed to evaluate the role of membrane microdomains (MMd) in virulence of Bacillus anthracis by assessing the effects of known antifungal compounds, referred to as raft-associated lipid biosynthesis inhibitors (RALBIs), on its pathophysiology. MATERIALS AND METHODS: FDA-approved antifungal compounds representing three distinct classes-azoles (ketoconazole), allylamines (terbinafine), and polyenes (nystatin), were tested against B. anthracis. These compounds target different enzymes involved in MMd-associated lipid biosynthesis. Their impact on sporulation, toxin secretion,

Aspergillus hiratsukae as a causative agent of endophthalmitis: A case report.

BACKGROUND: Aspergillus flavus and Aspergillus fumigatus are commonly involved in fungal endophthalmitis. However, other less pathogenic species of the genus are also causal agents of opportunistic infections. Aspergillus hiratsukae, a species within Aspergillus section Fumigati, is an uncommon fungus responsible for a few infectious cases worldwide. CASE REPORT: We present a case of post-surgical endophthalmitis in a man caused by A. hiratsukae after phacoemulsification surgery and intraocular lens implantation. The patient underwent an urgent vitrectomy in which infectious foci were observe

Sources of Antifungal Drugs.

Due to their eukaryotic heritage, the differences between a fungal pathogen's molecular makeup and its human host are small. Therefore, the discovery and subsequent development of novel antifungal drugs are extremely challenging. Nevertheless, since the 1940s, researchers have successfully uncovered potent candidates from natural or synthetic sources. Analogs and novel formulations of these drugs enhanced the pharmacological parameters and improved overall drug efficiency. These compounds ultimately became the founding members of novel drug classes and were successfully applied in clinical set

Metal Nanoparticles to Combat Candida albicans Infections: An Update.

Candidiasis is an opportunistic mycosis with high annual incidence worldwide. In these infections, Candida albicans is the chief pathogen owing to its multiple virulence factors. C. albicans infections are usually treated with azoles, polyenes and echinocandins. However, these antifungals may have limitations regarding toxicity, relapse of infections, high cost, and emergence of antifungal resistance. Thus, the development of nanocarrier systems, such as metal nanoparticles, has been widely investigated. Metal nanoparticles are particulate dispersions or solid particles 10-100 nm in size, with

Existing and emerging therapies for the treatment of invasive candidiasis and candidemia.

INTRODUCTION: Invasive candidiasis or candidemia is a severe infection affecting more than 250,000 people worldwide every year. It is present in up to 16% of ICU patients. The prognosis of these infections is unfavorable, with global death estimated around 50,000 per year, which corresponds to up to 40% depending on patient severity and comorbidities. Therapeutic failure is not rare due to the emergence of multiresistant strains and of new species poorly responsive to current therapies like Candida auris. AREAS COVERED: We first review the positioning of antifungal drugs used to treat candidi

Conventional therapy and new antifungal drugs against Malassezia infections.

Malassezia yeasts are commensal microorganisms occurring on the skin of humans and animals causing dermatological disorders or systemic infections in severely immunocompromised hosts. Despite attempts to control such yeast infections with topical and systemic antifungals, recurrence of clinical signs of skin infections as well as treatment failure in preventing or treating Malassezia furfur fungemia have been reported most likely due to wrong management of these infections (e.g., due to early termination of treatment) or due to the occurrence of resistant phenomena. Standardized methods for in

New antifungal agents and preparations.

In neutropenic patients amphotericin B remains the drug of choice for the treatment of systemic fungal infections. On the basis of a superior efficacy in combination with a lower toxicity, the triazoles have superseded the older azoles. Regularly, amphotericin B and a triazole are used simultaneously without any evidence from clinical trials that such a strategy is safe and efficacious. Liposomal preparation, lipid complex or colloidal dispersion of amphotericin B have been produced successfully to reduce toxicity. However, there is only one small randomised study that hints at the superiority

Liposomal amphotericin B-the past.

The discovery of amphotericin B, a polyene antifungal compound, in the 1950s, and the formulation of this compound in a liposomal drug delivery system, has resulted in decades of use in systemic fungal infections. The use of liposomal amphotericin B formulation is referenced in many international guidelines for the treatment of fungal infections such as Aspergillus and cryptococcal disease and Candida infections, as well as other less common infections such as visceral leishmaniasis. With the development of liposomal amphotericin B, an improved therapeutic index could be achieved that allowed
