Female albino mice were fed sublethal doses of KCN (approx. 10 micrograms/mouse/day) for 7 days, injected intraperitoneally with phenobarbitone (50 mg/kg body wt/day) in the subsequent 3 days, and sacrificed 24 hr after the last injection. Phenobarbitone sleeping time was increasingly shortened (16-27%) daily in cyanide-fed mice in comparison with cyanide-free controls. An efficient copper catalyzed strategy for the synthesis of a variety of benzothiazolethione derivatives has been developed. In the presence of CuCl, the three-component reaction of o-iodoanilines and K2S with p-toluenesulfonylmethyl isocyanide proceeded smoothly to obtain the corresponding benzothiazolethiones in good to excellent isolated yields. Notably, isocyanide functioned as a carbon source and K2S functioned as a sulfur source in this reaction.

Chemical shift anisotropy (CSA) has been an invaluable probe of structure and dynamics for a variety of systems in NMR spectroscopy. Unfortunately, the presence of strong quadrupolar couplings has severely limited the ability to measure CSA in nuclei with spins I > 1/2. Here we show that these two interactions can be refocused at different times in a 2D multiple-quantum NMR experiment on polycrystalline samples. Sodium transport and oxygen consumption have been simultaneously studied in the short-circuited toad skin. The response of dorsal root ganglion (DRG) neurons to metabolic inhibition is known to involve calcium-activated K+ channels; in most neuronal types ATP-sensitive K+ channels (K(ATP)) also contribute, but this is not yet established in the DRG. Isolated rat ventricular myocytes were investigated with the whole-cell patch-clamp technique. Chlorpromazine inhibited inward-rectifying K+ currents (IC50 = 6.1 microM), time-independent outward currents (IC50 = 16 microM) and transient outward K+ currents. The effect of coenzyme Q10 (CoQ10) on the cyanide (CN-)-induced ATP-sensitive K+ channel current (KATP) was examined in single atrial myocytes, using the patch clamp technique. Effects of the change of external ionic composition and the addition of metabolic inhibitors on rabbit cornea were studied by recording the epithelial intracellular potential. High K and Li Ringer's solutions, applied to the corneal endothelial side, caused a marked depolarization of the epithelial cells, but no potential change was seen when applied to the epithelial side. Direct measurements of intracellular K(+) and Cl(-)activities before and after blockage of cellular metabolic processes indicate that K(+) is actively transported inwardly and Cl(-)is actively transported outwardly from the giant cell of the abdominal ganglion of Aplysia. The rewarming of cells that have been cooled to 1 degrees +/- 1 degrees C causes K(+) to be taken up and Cl(-) to be extruded against electrochemical gradients.
