Scientific evidence for health effects attributed to the consumption of probiotics and prebiotics: an update for current perspectives and future challenges.

Probiotics and prebiotics, mainly commercialised as food ingredients and also as supplements, are considered highly profitable niche markets. However, in recent years, the food industry has suffered from a series of health claim restrictions on probiotics and prebiotics in many parts of the world, including those made by the European Food Safety Authority. Therefore, we reviewed the core benefits of probiotic and prebiotic consumption on health. A number of studies have examined the prevention and/or management of intestinal infections, respiratory tract infections, CVD, osteoporosis, urogenit…

Probiotics and prebiotics in preventing episodes of acute otitis media in high-risk children: a randomized, double-blind, placebo-controlled study.

BACKGROUND: Several studies have suggested that probiotics (proB) and/or prebiotics (preB) could reduce the burden of infection in infants and toddlers. We aimed to determine whether follow-up formula supplemented with proB and preB could reduce the risk of acute otitis media (AOM). METHODS: In this double-blind, placebo-controlled trial from November 2007 to April 2009, 37 pediatricians in France enrolled children 7 to 13 months of age with high risk of AOM who were randomly assigned to receive follow-up formula supplemented with proB (Streptococcus thermophilus NCC 2496, Streptococcus saliv…

Gut-brain health effects of PREbiotics in older adults with suspected COgnitive DEcline: design of the PRECODE randomised placebo-controlled trial.

UNLABELLED: Global lifespan is rising, alongside increasing age-related conditions. Cognitive disorders such as dementia, are projected to triple in number by 2050. Currently, no cure is available for Alzheimer's disease (AD), the most common form of dementia, and the need for preventative strategies is paramount. Subjective cognitive decline plus (SCD+) lies on the AD continuum, with increased risk of accelerated cognitive decline, providing an ideal target group for early intervention. The microbiota-gut-brain axis (MGBA) is an emerging avenue for prevention, as dietary fibres may beneficial

Integrated ¹H-NMR Metabolomics and Growth Kinetics Uncover Three Distinct Metabolic Scenarios in Lactiplantibacillus pentosus P7 Fermentation of Plant-Derived Prebiotics.

Lactic acid bacteria (LAB) drive a broad range of food and biotechnological fermentations, the outcomes of which depend not only on the bacterial genotype but also on the chemical composition of the fermentation substrate. To resolve how a single strain reorganises chemically distinct plant matrices, we profiled fermentations of Lactiplantibacillus pentosus P7 (GenBank JBLMKZ000000000) on garlic, onion, and kiwifruit extracts prepared in water and 70% ethanol, using growth kinetics combined with solvent-suppressed 500-MHz proton nuclear magnetic resonance metabolomics over 48 h, and integrated

Engineered prebiotic microcapsules Co-Encapsulating berberine and curcumin Elicit multi-synergistic therapy for ulcerative colitis.

BACKGROUND: Ulcerative colitis (UC) is a common type of inflammatory bowel disease, where the vicious cycle of inflammation and oxidative stress poses a major challenge in its treatment, and existing therapies have limitations. Berberine (BBR) and curcumin (CUR) have the potential for synergistic treatment of UC, but this potential has not been verified in UC. Additionally, both BBR and CUR suffer from poor water solubility and low bioavailability. This study aims to construct prebiotic microcapsules (BBR/CUR@MC) for the co-delivery of BBR and CUR and explore their therapeutic mechanism in UC.

Enhancement of environmental and gastrointestinal digestive stability of probiotic-loaded liposomes: Study of inulin gelation and prebiotic effects.

This study develops a liposome-inulin gel delivery system to enhance the environmental and gastrointestinal stability of Lactobacillus rhamnosus. As the inulin gel ratio increases from 1:1 to 1:3, the encapsulation efficiency of probiotic-loaded system (L-Lip@gel) significantly increases, reaching ∼89% (P < 0.05), accompanied by an increase in particle size, reduced zeta potential (-6.2 mV) and a higher polydispersity index (0.47). Microstructural characterisation indicates that the inulin gel coating interacts with the hydrophilic core of liposomes via hydrogen bonding and steric effects, for

Evaluation of antenatal prebiotic intake in infants at risk of atopy: the double-blind PREGRALL randomized clinical trial.

BACKGROUND: New primary preventive therapeutic strategies for atopic dermatitis (AD) are needed. Atopic diseases are associated with disrupted gut microbial balance in early life, suggesting that optimizing microflora through intervention could improve health. Prebiotics, which are immunomodulatory sugars, promote gut microbiota diversity. Most clinical trials focus on improving postnatal infant gut colonization, but prenatal life is crucial for establishing tolerance mechanisms. Preclinical studies indicate that maternal intake of galacto-oligosaccharide (GOS)/inulin prebiotics decreases the …

Prebiotic modulation of FMT donor microbiota enhances MASLD-relevant taxa and functions in an in vitro gut model.

AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD, formerly non-alcoholic fatty liver disease) is a prevalent and progressive condition closely linked to gut microbiota composition. Fecal microbiota transplantation (FMT) may help restore a health-associated microbiome, but its efficacy is often limited by inconsistent engraftment of beneficial taxa. Prebiotics may selectively support keystone microbes associated with reduced MASLD risk. This study evaluated two prebiotics, inulin and xylooligosaccharides (XOS), for their ability to modulate the microbiota of healthy FMT don…

The Rich Chemistry of Ionized Methanol With Sulfur Dioxide: Pathways to Prebiotic and Elusive Species in Earth's Atmosphere and Space.

Sulfur dioxide (SO2) and methanol (CH3OH) are key components in both atmospheric and astrochemical environments, where ionizing radiation can initiate complex reaction networks. Here we investigate the ion chemistry of the CH3OH•+ + SO2 system by combining synchrotron experiments with theoretical analysis. The methanol radical cation CH3OH•+, generated at different photon energies, reacts efficiently with SO2, leading to an abundant distribution of ionic and neutral products. The reaction proceeds through rapid pathways involving the formation of the bound adduct [HOCH2O(H)SO]•+, which acts as

Impact of probiotic, prebiotic, and synbiotic supplementation on the gut microbiome in older adults with sarcopenia, obesity, and sarcopenic obesity.

Gut microbiome plays an important role in several metabolic, immune, and inflammatory pathways; however, there is limited evidence for its r … n potential in enhancing SCFA production, reducing inflammation, and improving insulin sensitivity. Prebiotics are non-digestible fibers that selectively activate the growth of beneficial gut bacteria, further supporting gut health by proliferating the growth of SCFA-producing bacteria. Synbiotics, a combination of probiotics and prebiotics, provide a synerg …

Prevention of Intestinal Inflammation and Gut Dysbiosis by Prebiotic Grape Seed Flour in Mice with DSS-Induced Colitis.

Background: Inflammatory bowel disease is a complex intestinal inflammatory disorder linked to immune dysregulation, oxidative stress, and an imbalance in the gut microbiota. Grape seed flour (GSF), a winemaking by-product rich in polyphenols and fibres, displays antioxidant and anti-inflammatory properties and may help maintain gut homeostasis. Methods: The phenolic composition and antioxidant capacity of our GSF were evaluated. We assessed whether a diet containing 10% (w/w) GSF protects against dextran sulphate sodium (DSS)-induced acute colitis in BALB/c mice. Animals received either a sta

Preoperative prebiotic supplementation alleviates colitis and improves anastomotic healing associated with modulation of the gut microbiota in rats.

BACKGROUND: Anastomotic leak is among the most feared complications in colorectal surgery. Patients with inflammatory bowel disease are particularly susceptible to impaired anastomotic healing, partially because of bacterial dysbiosis. Aiming to improve anastomotic healing, we examined the role of microbiota-targeted prehabilitation with a complex prebiotic supplement containing arabinogalactan, humic substances, and micronutrients in a rat model of experimental colitis. METHOD: Rats were subjected to standard or prebiotic-supplemented diets for 4 weeks before receiving water or dextran sulfa
