Constitutive expression of a 34 kDa Mst1 fragment in the forebrain distinct from the caspase 3-cleaved isoform.

Mammalian sterile 20-like kinase 1 (Mst1), a ubiquitously expressed 60 kDa serine/threonine kinase, plays crucial roles in various cellular processes. Mst1 is activated by autophosphorylation at Thr183 and caspase 3-mediated cleavage, producing a catalytically active 36/37 kDa (or, in some studies, a 34 kDa) fragment under apoptotic conditions. However, whether the 34 and 36/37 kDa fragments observed in different studies are identical remains unclear, given their similar sizes and the specific detection of a 34 kDa fragment in normal mouse liver and intestinal epithelial cells. This study aime

Forebrain AR Deletion Restores PR Expression but not Reproduction in Prenatally Androgenized Female Mice.

Prenatal androgen excess (PNA), an etiologic factor for polycystic ovary syndrome (PCOS), is implicated in programming long-term reproductive deficits in females such as anovulation, subfertility, and hyperandrogenism. Impaired steroid hormone feedback is a key neuroendocrine feature suspected to underpin the development of reproductive dysfunction in both clinical PCOS and in PNA mice exposed to dihydrotestosterone during late gestation. PNA is suspected to act in the brain to program the impaired sensitivity of the GnRH neuronal network to progesterone negative feedback, centrally dysregulat

Forebrain expression of serine racemase during postnatal development.

N-methyl-D-aspartate receptors (NMDARs) are important for synaptogenesis, synaptic maturation and refinement during the early postnatal weeks after birth. Defective synapse formation or refinement underlie cognitive and emotional abnormalities in various neurodevelopmental disorders (NDDs), including schizophrenia (Sz) and autism spectrum disorder (ASD). Serine racemase (SR) is a neuronal enzyme that produces D-serine, a co-agonist required for full NMDAR activation. NMDAR hypofunction as a result of genetic SR elimination and reduced synaptic availability of D-serine reduces neuronal dendriti

Metabolomic study of altered energy metabolism during global forebrain ischemia and ischemic precoditioning in blood plasma in homocysteine treated rats.

Elevated homocysteine (Hcy) level is a well known risk factor for cardiovascular and neuropsychiatric diseases. In this study, we investigated metabolic changes in blood plasma in Hcy-treated rats. In combination with Hcy injections to induce hyperhomocysteinemia-like state, we used an animal model of global cerebral ischemia to investigate metabolic changes after 24 h reperfusion in rats. We also focused on the endogenous phenomenon known as ischemic tolerance induced by the preischemic treatment. The experiments were carried out on blood plasma samples as they are easily available and metabo

Neural progenitor fate decision defects, cortical hypoplasia and behavioral impairment in Celsr1-deficient mice.

The development of the cerebral cortex is a tightly regulated process that relies on exquisitely coordinated actions of intrinsic and extrinsic cues. Here, we show that the communication between forebrain meninges and apical neural progenitor cells (aNPC) is essential to cortical development, and that the basal compartment of aNPC is key to this communication process. We found that Celsr1, a cadherin of the adhesion G protein coupled receptor family, controls branching of aNPC basal processes abutting the meninges and thereby regulates retinoic acid (RA)-dependent neurogenesis. Dysfunction of spatacsin leads to axonal pathology in SPG11-linked hereditary spastic paraplegia.

Hereditary spastic paraplegias are a group of inherited motor neuron diseases characterized by progressive paraparesis and spasticity. Mutations in the spastic paraplegia gene SPG11, encoding spatacsin, cause an autosomal-recessive disease trait; however, the precise knowledge about the role of spatacsin in neurons is very limited. We for the first time analyzed the expression and function of spatacsin in human forebrain neurons derived from human pluripotent stem cells including lines from two SPG11 patients and two controls. SPG11 patients'-derived neurons exhibited downregulation of specifi

Rat forebrain perfusion in vivo by 1 artery like the isolated kidney model: a robust recovery model permitting ischemia without anesthesia to compare multiple brain injury states.

BACKGROUND: Rat brain perfusion models are critical to basic research, but they can be imprecise and/or not durable for extended outcome studies. OBJECTIVE: To demonstrate a rat brain perfusion model that provides a simplified reliable brain perfusion circuit, reduces variables during experiment and recovery, and therefore permits more precise, reliable, and context-independent research data. METHODS: Rat forebrain perfusion was reduced surgically to that by 1 internal carotid artery without injury to the animal. The next day, the fully awake rat was studied for brain ischemia painlessly yet

Immunohistochemical localization of the angiotensin-(1-7) receptor Mas in the murine forebrain.

Apart from the well-known biologically active angiotensin II, other biologically active angiotensins have been discovered, including angiotensin IV and angiotensin-(1-7). Some years ago, we and others discovered that the Mas proto-oncogene encodes a receptor that is essential for angiotensin-(1-7) signaling. Angiotensin-(1-7) is not only expressed in the periphery but also within the brain. Based on that, we examined the distribution of Mas within the murine brain, using an antibody directed against the 3(rd) cytoplasmic loop of the receptor protein. Strongest Mas protein expression was detect

Sleep homeostasis and depression: studies with the rat clomipramine model of depression.

Neonatal treatment of rat pups with clomipramine (CLI) has been shown to cause long-lasting and persistent depression-related behaviors and changes in sleep architecture and in brain-derived neurotrophic factor (BDNF) signaling in adult animals, producing an animal model of depression. However, the molecular mechanisms which mediate these effects of early-life CLI treatment on adult animals remain largely unknown. In order to characterize these further, we investigated in neonatally CLI-treated rats the sleep architecture as well as the extracellular and cellular levels of sleep regulators (ni

Bilateral projections from rat MI whisker cortex to the neostriatum, thalamus, and claustrum: forebrain circuits for modulating whisking behavior.

In rats, whisking behavior is characterized by high-frequency synchronous movements and other stereotyped patterns of bilateral coordination that are rarely seen in the bilateral movements of the limbs. This suggests that the motor systems controlling whisker and limb movements must have qualitative or quantitative differences in their interhemispheric connections. To test this hypothesis, anterograde tracing methods were used to characterize the bilateral distribution of projections from the whisker and forepaw regions in the primary motor (MI) cortex. Unilateral tracer injections in the MI w

Neuropathology of a 4-month-old infant born to a woman with phenylketonuria.

We evaluated the brain of a 4-month-old male infant whose mother had inadequately controlled maternal phenylketonuria (MPKU). At autopsy his brain was normally developed but underweight. We found ventriculomegaly, hypoplastic cerebral white matter, and delay of myelination in late myelinating tracts without white matter astrocytosis and without chronic lesions in any gray matter structure. We compared the development of the infant's white matter tracts with published data on infant myelination. Congenital heart disease complicated the case. Abnormalities in developmental white matter may accou

Repeated antipsychotic drug exposure in developing rats: dopamine receptor effects.

Antipsychotic drugs are often prescribed to juvenile psychiatric patients, though their cerebral effects during development are incompletely described. Accordingly, we studied the effects of repeated treatment with dissimilar antipsychotic drugs on dopamine (DA) receptors in juvenile vs. adult rats. Tissue levels of DA receptor types (D1, D2, D3, and D4) in forebrain regions of juvenile rats were quantified after 3 weeks of daily treatment with representative first- (fluphenazine) and second-generation (clozapine and olanzapine) antipsychotics, and compared with similarly treated adult rats ex

[Cyclooxygenase and prostanoids--biological implications].

Arachidonic acid is the main precursor of eikosanoids, which regulate the function of various organs and systems. It is released from cellular membrane phospholipids by phospholipase A2 or indirectly by phospholipases C and D. Prostaglandins, prostacyclin (PGI2), and thromboxane A2 (TAX2) are synthesized from arachidonic acid on a metabolic pathway dependent on prostaglandin H2 synthase activity, also known as cyclooxygenase (COX). Of the 12 prostaglandins, the most potent are PGD2, PGE2, and PGF2. The others are synthesized in a primary step of arachidonic acid transformation (PGG2, PGH2), by

Selectivity of canary HVC neurons for the bird's own song: modulation by photoperiodic conditions.

To what extent seasonal factors modify the neuronal functional properties within the nuclei of the avian song system remains an open question. In adult songbirds, neurons of the song premotor nucleus HVC (used as a proper name) exhibit selective responses for the bird's own song (BOS). Here we examine whether, outside the breeding season, when songs are less stereotyped, HVC neurons of male canaries still respond selectively to the BOS produced during this period. In an initial experiment, single-unit recordings (n = 114) revealed that the neuronal selectivity for the current BOS was attenuate

Wired for reproduction: organization and development of sexually dimorphic circuits in the mammalian forebrain.

Mammalian reproduction depends on the coordinated expression of behavior with precisely timed physiological events that are fundamentally different in males and females. An improved understanding of the neuroanatomical relationships between sexually dimorphic parts of the forebrain has contributed to a significant paradigm shift in how functional neural systems are approached experimentally. This review focuses on the organization of interconnected limbic-hypothalamic pathways that participate in the neural control of reproduction and summarizes what is known about the developmental neurobiolo

[Nature of centrogenic reactions developing during surgical interventions on the brain].

The nature of hemodynamic reactions and EEG changes developing during surgery is discussed, based on analysis of 392 interventions for removal of brain tumors. Two main types of centrogenic reactions are distinguished: (1) connected with the direct effect on brain centers engaged in cardiovascular regulation and (2) caused by stimulation of the median structures of the brain, most often caused by hyperperfusion in different compartments of the brain. Use of anesthesiological method limiting the functional relations between the brain stem and forebrain notably decreases the incidence of centrog

Progesterone treatment increases Fos-immunoreactivity within some progestin receptor-containing neurons in localized regions of female rat forebrain.

In female rats, the sequential release of estradiol and progesterone from the ovaries is required for the expression of sexual behavior during the estrous cycle. Many of the neuronal effects of estradiol and progesterone involve estrogen and progestin receptors. Treatment with a behaviorally-effective dose of estradiol increases Fos expression, suggestive of neuronal response, and subsequent treatment with a behaviorally-effective dose of progesterone further increases Fos expression within a few hours in female rat brain. In order to determine if neurons that respond to progesterone with incr

Impairment in memory function and neurodegenerative changes in the cholinergic basal forebrain system induced by chronic intake of ethanol.

Chronic intake of ethanol both in human and rat results in a substantial impairment in memory function associated with a reduction in the number of cholinergic neurons in the basal forebrain which give rise to the cholinergic afferentation of the cortical mantle. Degenerative changes in the basal forebrain are paralleled by the concomitant reduction of presynaptic cholinergic markers (synthesis, content and release of acetylcholine) in the neocortex and hippocampus. Cognitive dysfunction in rat induced by ethanol treatment can be ameliorated by the pharmacological manipulation of central choli

The organization of the thalamocortical connections of the mediodorsal thalamic nucleus in the rat, related to the ventral forebrain-prefrontal cortex topography.

The medial and central segments of the mediodorsal nucleus of the thalamus (MD) receive afferents from the ventral forebrain, including the piriform cortex, the ventral pallidum, and the amygdaloid complex. Because MD is reciprocally interconnected with prefrontal and agranular insular cortical areas, it provides a relay of ventral forebrain activity to these cortical areas. However, there are also direct projections from the piriform cortex and the amygdala to the prefrontal and agranular insular cortices. This study addresses whether this system has a "triangular" organization, such that str
