Recent studies have highlighted palmitoylation, a novel protein post-translational modification, as a key player in various signaling pathways that contribute to tumorigenesis and drug resistance. Despite this, its role in bladder cancer (BCa) development remains inadequately understood. In this study, ZDHHC9 emerged as a significantly upregulated oncogene in BCa. Functionally, ZDHHC9 knockdown markedly inhibited tumor proliferation, promoted tumor cell apoptosis, and enhanced the efficacy of gemcitabine (GEM) and cisplatin (CDDP). Mechanistically, SP1 was found to transcriptionally activate Z

Translational control has emerged as a critical layer of gene expression reprogramming in cancer, with the ribosome functioning as a central regulatory node rather than a passive protein synthesis machinery. This review provides an integrated overview of how defects in ribosomes and translational regulation contribute to the pathogenesis of lymphoid leukemias and lymphomas. We focus on 3 major axes: mutations in ribosomal proteins, alterations in ribosomal RNA, and dysregulation of translation initiation mediated by the eukaryotic initiation factor 4F complex. Recurrent mutations in ribosomal

Horticultural plants play a vital role in agricultural production and daily life. Post-translational modifications are essential for regulating various cellular processes, but the functions of SUMOylation, an increasingly significant post-translational modification, remain to be thoroughly explored in horticultural plants. In this review, we summarize the current research advancements regarding protein SUMOylation in horticultural plants and offer perspectives for future investigations. We begin by introducing the SUMOylation system in horticultural plants. Subsequently, we focus on the roles

Methacrylyl-CoA is a key metabolic intermediate in the valine catabolic pathway. Its accumulation has been found to be cytotoxic and associated with pathological conditions. Nevertheless, detailed biological effects of methacrylyl-CoA and methacrylate in human physiology and pathology are poorly understood. We propose that the electrophilicity of the alkene bond in the methacrylyl group can react with the cysteine residues in proteins resulting in an unexplored protein post-translational modification (PTM), cysteine S-2-carboxypropylation (C2cp). To test and validate this mechanistic hypothesi

N6-methyladenosine (m6A) is the most abundant internal modification in eukaryotic mRNAs, yet how plants recognize this chemical modification to swiftly adjust developmental plasticity under environmental stresses remains unclear. Here we show that m6A mRNA modification and its reader protein EVOLUTIONARILY CONSERVED C-TERMINAL REGION 8 (ECT8) act together as a key checkpoint for negative feedback regulation of abscisic acid (ABA) signalling by sequestering the m6A-modified ABA receptor gene PYRABACTIN RESISTANCE 1-LIKE 7 (PYL7) via phase-separated ECT8 condensates in stress granules in respons

Controlled modifications of amino acids are an indispensable tool for advancing fundamental and translational research based on peptides and proteins. Yet, we still lack methods to chemically modify each naturally occurring amino acid sidechain. To help address this gap, we show that N,α-diaryl oxaziridines expand the scope of bioconjugation methods to chemically modify cysteine, methionine, and tryptophan residues with evidence for additional tyrosine labelling in a proteomic context. Conjugation primarily at tryptophan sites can be accessed by selective cleavage of modifications at other sid

Protein lysine modifications (PLMs) are hotspots of post-translational modifications and are involved in many diseases; however, their role in human sperm remains obscure. This study examined the presence and functional roles of a classical PLM (lysine acetylation, Kac) and three novel PLMs (lysine malonylation, Kmal; lysine succinylation, Ksucc; lysine crotonylation, Kcr) in human sperm. Immunoblotting and immunofluorescence assays revealed modified proteins (15-150 kDa) in the tails of human sperm. An immunoaffinity approach coupled with liquid chromatography/tandem mass spectrometry reveale

Precise, context-sensitive control of protein synthesis is critical in eukaryotic cells, and mRNA structure has emerged as a central mechanism for achieving this control. mRNA structures influence every stage of translation. During initiation, structures within 5' untranslated regions modulate ribosome association with mRNA and the balance between translation of the main open reading frame and use of upstream or alternative start sites. During elongation, structures in the coding sequence govern ribosome pausing in ways that can either protect mRNA stability or trigger decay. During terminatio

Environmental changes put stress on living organisms. We find that nutrient starvation induces dynamic protein aggregations in yeast cells, and many chaperones are involved in this process. Among them, Sti1/HOP, the co-chaperone of Hsp70 and Hsp90, plays roles in the formation of protein quality control (PQC) compartments and protein stasis (or proteostasis) maintenance, and it co-localizes to insoluble protein deposits (IPOD) by liquid-liquid phase separation (LLPS). Notably, the subcellular localization and cytoplasmic aggregation of Sti1 are rigorously regulated by the PQC machinery, includ

Palmitoylation is a key post-translational modification regulating viral replication, yet its regulatory mechanism in Dengue virus (DENV) infection remains elusive. This study aimed to elucidate the underlying regulatory function of palmitoylation and zinc finger Asp-His-His-Cys (ZDHHC) proteins in DENV replication and identify palmitoylated DENV proteins. We explored the function of palmitoylation in DENV replication using the palmitoylation inhibitor 2-bromopalmitate (2BP) and enhancer palmitic acid (PA), combined with qRT-PCR, western blot, confocal immunofluorescence microscopy, co-immunop

Post-translational modification(PTM) is the terminal regulatory step of gene expression, directly regulating protein function through reversible or irreversible chemical modifications, enabling cells to rapidly respond to internal and external environmental stimuli. Among various post-translational modifications (PTMs), the dynamic balance between phosphorylation and dephosphorylation constitutes the molecular switch for inflammatory signal transduction, and disruption of this balance is an important cause of the persistent non-healing of inflammatory bowel disease (IBD). The catalytic subunit

We have studied the effect of free radical generating agents on the mono-(ADP)-ribosylation of rat liver cytosolic proteins. Our results show that this post-translational modification, whose physiological significance is still unclear, is activated by lipid peroxidant agents via activation of cytoplasmatic mono-(ADP)-ribosyltransferases. The implication of free radicals in this process is demonstrated by the fact that mono-(ADP)-ribosylation can be prevented by melatonin, N-tert-butyl-alpha-phenylnitrone and dithiothreitol. On the basis of our results, we discuss the modification of proteins c

RNA modifications have emerged as crucial regulators of cellular processes linked to aging. However, the dynamic changes in global RNA modification patterns during aging and their functional implications remain underexplored. Here, we comprehensively deciphered the RNA modification landscapes of total RNA, poly(A)-enriched mRNA, and tRNA-enriched fragments across six tissues in both young and aged mice, as well as in multiple cellular senescence models. Aged tissues exhibit overall decline of RNA modification abundance in tRNA, rather than in total RNA or mRNA, when compared to young tissues.

Activation of plant immunity is associated with dramatic changes in the cellular redox status. Both oxidative and reductive bursts have been described that trigger a set of down stream responses resulting in reprogramming of the transcriptome and establishment of disease resistance. Nonetheless, how these redox changes are sensed and signal to downstream regulators remained a missing link in studies of plant immunity. Emerging evidence now indicates that pathogen-induced changes in the cellular redox environment are sensed by reactive cysteine residues of key regulatory proteins. Varying degre

SUMOylation is characterized as a ubiquitin-like post-translational modification that exhibits mechanistic similarities with ubiquitination. Importantly, it modulates the ubiquitination levels of some target proteins, thereby influencing protein stability. Dysregulation of this modification pathway plays a pivotal role in the pathogenesis and progression of various tumors. While recent studies have demonstrated that protein SUMOylation regulates tumor progression, the specific function and mechanism of Ran-binding protein 2 (RanBP2), an E3 SUMO ligase, remain to be elucidated in lung adenocarc

Neuroglobin protects neurons from hypoxia in vitro and in vivo; however, the underlying mechanisms for this effect remain poorly understood. Most of the neuroglobin is present in a hexacoordinate state with proximal and distal histidines in the heme pocket directly bound to the heme iron. At equilibrium, the concentration of the five-coordinate neuroglobin remains very low (0.1-5%). Recent studies have shown that post-translational redox regulation of neuroglobin surface thiol disulfide formation increases the open probability of the heme pocket and allows nitrite binding and reaction to form

Sickle cell disease (SCD) is an inherited hemoglobinopathy characterized by sickle-shaped red blood cells (RBCs). Primary cilia are mechanosensory organelles and are projected in the lumen of blood vessels to detect blood flow. We previously reported that interaction between microvasculature endothelial cells and sickled RBCs resulted in altered blood flow that can elevate reactive oxygen species, leading to increased deciliation in SCD patients. However, the impact of deciliation mediated by sickled RBCs in the context of the ciliary protein profiles remains unclear. Here, we investigated cel

The intracellular target of diphtheria toxin is a modified histidine residue, diphthamide, in the translation elongation factor, eEF2 (also known as EFT1). This enigmatic modification occurs in all eukaryotes and is produced in yeast by the action of five gene products, DPH1 to DPH5. Sequence homologues of these genes are present in all sequenced eukaryotic genomes and, in higher eukaryotes, there is functional evidence for DPH1, DPH2, DPH3 and DPH5 acting in diphthamide biosynthesis. We identified a mouse that was mutant for the remaining gene, Dph4. Cells derived from homozygous mutant embry

Poly(ADP-ribose) polymerase 1 (PARP1) is a Zinc Finger (ZF) protein that is involved in DNA damage response. PARP1 contains three zinc finger (ZF) domains, two with a Cys2HisCys (CCHC) ligand set and one with a Cys4 (CCCC) ligand set that coordinate zinc. Persulfidation of cysteine residues in proteins is a new type of post-translational modification (PTM), for which ZFs are emerging as frequently modified. PARP1 has been identified as persulfidated in several cellular based persulfide-specific proteomics studies across several different cell types. Here, we examine PARP1 persulfidation by H2S

Succinylation is a post-translational modification that occurs on lysine residues and is mediated by succinyl-CoA, an intermediate of the tricarboxylic acid (TCA) cycle. It can significantly affect protein conformation, activity, stability, and interactions with other molecules, thereby participating in the regulation of important biological processes such as cellular metabolism, energy homeostasis, and signaling pathways. In recent years, increasing evidence has shown that succinylation plays a key role in central nervous system (CNS) diseases, particularly in Alzheimer's disease, stroke, sub
