Human xeroderma pigmentosum (XP) or Fanconi anemia (FA) fibroblasts displayed shouldered 45 degrees C heat survival curves not significantly different from normal fibroblasts, a result similar to that previously found for ataxia telangiectasia (AT) cells, indicating heat resistance is not linked to either uv or low-LET ionizing radiation resistance. Hyperthermia (45 degrees C) sensitized normal and XP fibroblasts to killing by gamma radiation but failed to sensitize the cells to the lethal effects of 254 nm uv radiation. Thermal inhibition of repair of ionizing radiation lesions but not uv-ind

PURPOSE: Environment and lifestyle factors are being attributed toward increased instances of male infertility. Rapid technological advancement, results in emission of electromagnetic radiations of different frequency which impacts human both biologically as well as genetically. Devices like cell phone, power line and monitors emit electromagnetic radiation and are a major source of the exposure. Numerous studies describe the detrimental consequence of radiation on physiological parameters of male reproductive system including sperm parameters (morphology, motility, and viability), metabolism

After large releases of radionuclides, exposure of the embryo or fetus can take place by external irradiation or uptake of radionuclides. The embryo and fetus are radiosensitive throughout prenatal development. The quality and extent of radiation effects depend on the developmental stage. During the preimplantation period (one to 10 days postconception, p.c.), a radiation exposure of at least 0.2 Gy can cause the death of the embryo. Malformations are only observed in rare cases when genetic predispositions exist. Macroscopic, anatomical malformations are induced only after irradiation during

We used differential display, a method designed to amplify partial cDNA sequences from subsets of mRNAs, to identify mRNAs induced by ionizing radiation in human Epstein Barr Virus (EBV)-transformed lymphoblastoid cells. Increased expression of a cDNA corresponding to the inositol 1,4,5 trisphosphate receptor (InsP3R) type 1 was observed after exposure of cells to 3Gy gamma-rays. This was confirmed by Northern blot analysis. The increase in mRNA for InsP3R type 1 was accompanied by a corresponding increase in the level of InsP3R type 1 protein as determined by Western blotting. Exposure of cel

The fibroblast culture 180BR, established from a patient showing an adverse response to radiotherapy, has been shown previously to be hypersensitive to ionizing radiation and to be defective in the repair of DNA double-strand breaks. We demonstrate here that the products of the catalytic subunit of DNA-dependent protein kinase (DNA-PKcs) and its regulatory subunits (Ku 70 and Ku 80) are present at normal levels and possess functional activity. The product of the gene mutated in the human genetic disorder ataxia-telangiectasia was also detected in these cells. Apoptosis was detected after high-

Classical radiation biology as we understand it clearly identifies genomic DNA as the primary target of ionizing radiation. The evidence appears rock-solid: ionizing radiation typically induces DSBs with a yield of ~30 per cell per Gy, and unrepaired DSBs are a very cytotoxic lesion. We know very well the kinetics of induction and repair of different types of DNA damage in different organisms and cell lines. And yet, higher organisms differ in their radiation sensitivity-humans can be unpredictably radiosensitive during radiotherapy; this can be due to genetic defects (e.g., ataxia telangiecta

The findings from both animal and human studies on the radiation risk at low doses and low dose rates are far from conclusive, primarily due to statistical limitations. However, to arrive at some estimate of radiation risk, a conservative approach has been taken, and a linear extrapolation of radiation effects from high doses to low doses has been made. Thus, it is assumed that any exposure to radiation carries some risk of somatic or genetic damage and that there is no threshold or safe dose. In medical practice, diagnostic radiological procedures should be performed only if useful clinical i

Chromosomal instability is characterized by an increased rate of chromatid and chromosome rearrangements occurring multiple generations after exposure to ionizing radiation. Although chromosomal instability has been observed in many different cell types after exposure to ionizing radiation, the mechanisms perpetuating the manifestation of this instability phenotype are unknown. This review explores the current evidence concerning the role of dicentric-mediated bridge-breakage-refusion cycles and the role of recombination at interstitial telomere bands in mediating chromosomal instability. Cytogenetics is the gold-standard in biological dosimetry for assessing a received dose of ionizing radiation. More modern techniques have recently emerged, but none are as specific as cytogenetic approaches, particularly the dicentric assay. Here, we will focus on the principal cytogenetic techniques used for biological dosimetry: the dicentric assay in metaphase cells, the micronuclei assay in binucleated cells, and the premature condensed chromosome (PCC) assay in interphase cells. New fluorescence in situ hybridization (FISH) techniques (such as telomere-centromere hybridization) have faci

Radiation exposures from tritium make up a substantial fraction of the occupational and accidental radiation exposures associated with the nuclear power industry. Tritiated water, the most abundant form of tritium, is of particular interest because it is readily taken up by human cells and its irradiation of the cells is spread over a period of days. To approximate the prolonged exposure and the conditions that the cells of an individual would experience in vivo, we irradiated human lymphocytes with tritiated water for 48 h in a 1:1 blood:medium mix. For estimation of the tritium beta-ray dose

The occurrence of chromosomal aberrations was studied at 1-14 days post-exposure in female BALB/c mice exposed to various doses of gamma radiation. The frequency of abnormal cells, chromatid and chromosome breaks, dicentrics, centric rings, acentric fragments and total aberrations increased with exposure dose, and it was highest at 7 Gy. A peak was recorded on day 1 post-exposure with a gradual decline thereafter. The chromosomal aberration yield reached a nadir on day 14 post-irradiation, without restoration to the control level. The best fit for the present data was by a linear-quadratic rel

Sequences of 38 independent X-ray-induced mutations in a cDNA of the human hypoxanthine phosphoribosyltransferase (hprt) gene were analyzed after it was integrated into mammalian chromosomes as part of a shuttle vector. There were 15 base substitutions (39%), 9 frameshifts (24%), 11 deletions (29%), and 3 other mutations (8%) of two kinds. Radiation-induced mutations were found throughout the coding region of the gene. Base substitution mutations occurred more often at A:T sites than at G:C sites, and there were more transversions than transitions. Most of the frameshifts and deletions had sho

The present study was aimed to evaluate the radioprotective efficacy of hesperidin (HN), a flavonone glycoside against gamma-radiation-induced cellular damage in cultured human peripheral blood lymphocytes. Different concentrations of HN (3.27, 6.55, 9.83, 13.10, 16.38 and 19.65 microM) were pre-incubated with lymphocytes for 30 min prior to gamma-irradiation [4 Gy] and the micronuclei (MN) scoring, dicentric aberration and comet assay were performed to fix the effective dose of HN against gamma-irradiation induced cellular damage. The results indicated that among all the concentrations, 16.38

Recovery of yeast cells after exposure to ionizing radiation was discovered in 1957. During the following decade, i.e., in the 'premolecular period' of studying this phenomenon, its basic features including dependence on cell ploidy, cellular energy exchange and radiation linear energy transfer (LET) were determined and a mathematical model of recovery was proposed. In addition, DNA double-strand breaks were shown to be the damage causing death of irradiated haploid and diploid cells. The results of these investigations led to the formulation of the concept that the ability of a cell to repair

Two sisters had breast cancer at four years and 11 years after diagnosis of Hodgkin's disease. The affected breast tissues had received several hundred rads of scatter radiation during treatment of the lymphoma. Family history revealed breast cancer in a third sister and five other women in the paternal line. Cytogenetic and HLA studies showed no markers of susceptibility to neoplasia. Development of second primary neoplasms of the breast in the two sisters may have resulted from interactions between genetic factors and carcinogenic effects of radiation exposure.

To analyse a role of the factor of a genetic fundamentals of cells in formation of radiation-induced genome instability (RIGI) we investigated a condition of DNA pattern, content of superoxide anion-radical O2*- and a sum of reactive oxigen species (ROS) (O2*-, OH*, H2O2), and also catalase activity in bone marrow cells of male and female mice of 101/H strain in the norm and at once after chronic (10 day) exposure to 200 mGy gamma-radiation. Thus we based on conception about a significance of mechanisms of DNA repair and production of reactive oxygen species (ROS) in development of radiation-i

This paper reviews current knowledge on the deterministic and stochastic risks (the latter including the risk of cancer and of hereditary disease) associated with exposure to ionizing radiation. Particular attention is paid to cancer risks following exposure to man-made low linear energy transfer radiation. Excess cancer risks have been observed in the Japanese atomic bomb survivors and in many medically and occupationally exposed groups. In general, the relative risks among Japanese survivors of atomic-bomb explosions are greater than those among comparable subsets in studies of medically exp

This review examines the environmental and genetic contributions to the anti-neutrophil cytoplasmic antibody-associated systemic vasculitides. The dominant environmental risk factors appear to be silica exposure for all three syndromes, and vitamin D deficiency is strongly suggested by the latitude and ultraviolet radiation gradient observed for Wegener's granulomatosis and Churg-Strauss syndrome. Genetic factors are generally not very strong, consistent with the rarity of these conditions in children. However, multiple genetic factors, each with a relatively small effect, may combine to creat
