Effects of environmental toxicants on the efferent ducts, epididymis and fertility.

Many of the reproductive toxicants have primary effects on the testis, which potentially overshadow effects downstream on the efferent ducts and epididymis. The specific target of these effects depends upon the dosage and time response. It is often necessary to design experiments that separate testosterone-dependent responses arising in the testis from direct effects on epididymal tissues and spermatozoa, to uncover the mechanisms of toxicity in excurrent ducts. Recent studies have confirmed that chemicals can also alter the time required for sperm transport through the epididymis. Currently t…

Hyper- and hypoosmotic water permeability of bovine sperm as a function of age, breed, and ejaculate number: Implications for cryopreservation.

The tolerance to cryoprotectant equilibration and the necessary cellular dehydration during equilibrium or quasi-equilibrium cooling that facilitates avoidance of intracellular ice is fundamentally connected to the ability of cells to transport water across the plasma membrane. Because of this, the relationship between the rate of water transport and post-thaw motility in bovine sperm has long been hypothesized, but has not been definitively established. Previous studies suggested that variability in bull sperm water permeability might explain post-thaw motility. Here we used a stopped-flow te

Loss of the importin Kpna2 causes infertility in male mice by disrupting the translocation of testis-specific transcription factors.

Karyopherins mediate the movement between the nucleus and cytoplasm of specific proteins in diverse cellular processes. Through a loss-of-function approach, we here examine the role of Karyopherin Subunit Alpha 2 (Kpna2) in spermatogenesis. Knockout male mice exhibited reduced body size and sperm motility, increased sperm abnormalities, and led to the dysregulation of testis gene expression and ultimately to infertility. Impaired mRNA expression mainly affected clusters of genes expressed in spermatids and spermatocytes. Downregulated genes included a set of genes that participate in cell adhe

The Clitoris-An Appraisal of its Reproductive Function During the Fertile Years: Why Was It, and Still Is, Overlooked in Accounts of Female Sexual Arousal.

Stimulating the clitoris activates the brain to instigate changes in the female genital tract, namely, the enhancement of vaginal blood flow that increases vaginal luminal pO2 , vaginal transudate (lubrication) facilitating painless penile penetration and partial neutralization of the basal luminal acidic pH, vaginal tenting, and ballooning delaying sperm transport and allowing semen de-coagulation and capacitation (sperm activation) factors to act until arousal ends (often by orgasm induction). All these genital changes taken together are of major importance in facilitating the possibility of…

Effects of putative epididymal osmolytes on sperm volume regulation of fertile and infertile c-ros transgenic Mice.

Volume regulation by spermatozoa has been demonstrated to be crucial in both mice and men for transport in the female tract. In order to determine the nature of osmolytes used by spermatozoa, they were released from the cauda epididymis of fertile c-ros heterozygous mice into incubation medium of uterine osmolality (representing an osmotic challenge), containing increasing concentrations of compounds that are major epididymal fluid components and known osmolytes in somatic cells. This should nullify the concentration gradients for osmolytes that mediate volume regulation, prevent osmolyte effl

Changes in the quality of rabbit semen in 14 consecutive ejaculates obtained every 15 minutes.

Modifications of semen quality related to ejaculation frequency is one of the most important and neglected factors from the standpoint of artificial insemination or sperm competition. New Zealand white rabbits (Oryctolagus cuniculus) offer an advantageous experimental model because they have characteristic sexual behavior, they present rapid ejaculation after a single intromission, they have a very short interval between successive ejaculations, and semen can be easily collected. The authors studied the modifications on sperm quality (semen volume, sperm concentration, sperm motility) produced

Vitamin K2-Dependent GGCX and MGP Are Required for Homeostatic Calcium Regulation of Sperm Maturation.

A low-calcium microenvironment is essential for spermatozoa to mature in the epididymis; however, it remains unclear how dysregulation of epididymal luminal calcium is associated with male infertility. Using a warfarin-induced vitamin K2 deficiency rat model, we found that vitamin-K-dependent γ-glutamyl carboxylase (GGCX) and matrix Gla protein (MGP) were essential in extracellular calcium signaling of the intercellular communication required for epididymal sperm maturation. We found that GGCX and MGP co-localized in the vesicular structures of epididymal cells and spermatozoa. Calcium-regulat

Cervico-utero-tubal factors in infertility.

Infertility is a common medical problem resulting from one or more defects in the reproductive tract. Of the known causes of infertility, cervico-utero-tubal factors are important because they are generally manageable. Cervical factors include changes in cervical architecture, mucus, or both, resulting in a hostile environment through which sperm must pass. Assessment of this factor includes an evaluation of the cervical mucus both as a transport medium for sperm and as a diagnostic indicator of pending ovulation. Uterine factors include congenital defects and benign tumors resulting in a dist

Pregnancy after brompheniramine treatment of a diabetic with incomplete emission failure.

Ejaculation disorders in diabetic men are most frequently caused by retrograde ejaculation resulting from a damage of the sympathetic nerves at the level of the bladder neck. This results in incomplete or complete emission failure because of insufficient sperm transport. Diabetic ejaculatory sterility can be treated with anticholinergic drugs. The reported case shows that as a result of its antihistaminic and anticholinergic properties, brompheniramine can be successfully used in the treatment of men with diabetic ejaculatory sterility, particularly in patients with incomplete emission failure…

Importance of SLC26 Transmembrane Anion Exchangers in Sperm Post-testicular Maturation and Fertilization Potential.

In mammals, sperm cells produced within the testis are structurally differentiated but remain immotile and are unable to fertilize the oocyte unless they undergo a series of maturation events during their transit in the male and female genital tracts. This post-testicular functional maturation is known to rely on the micro-environment of both male and female genital tracts, and is tightly controlled by the pH of their luminal milieus. In particular, within the epididymis, the establishment of a low bicarbonate (HCO3 -) concentration contributes to luminal acidification, which is necessary for

In vitro effect of granulocyte-macrophage colony-stimulating factor (GM-CSF) on the expression of genes related to sperm motility and energy metabolism and intracytoplasmic sperm injection outcomes in obstructive azoospermic patients.

BACKGROUND: The presence of granulocyte-macrophage colony-stimulating factor (GM-CSF) and its receptor in various testicular cells and spermatozoa suggests a potential role in enhancing spermatogonial and postmeiotic cell development. Moreover, GM-CSF activates the pivotal pathways implicated in sperm motility regulation and glucose metabolism. However, the impact of GM-CSF on testicular biopsies from patients with obstructive azoospermia (OA) remains unexplored. Therefore, this study aimed to investigate the in vitro effects of GM-CSF on the expression of genes related to glucose transporters

Endocrine-metabolic modulators in sperm biotechnology: cross-species mechanisms and translational priorities.

Endocrine-metabolic dysregulation is increasingly recognized as a determinant of male reproductive competence. Its implications for sperm biotechnology remain under-integrated. This review advances a systems framework in which insulin resistance (IR), adipokine imbalance, and vitamin D-calcium signaling converge on core sperm functional modules, i.e., membrane lipid architecture, mitochondrial efficiency, redox stability, and capacitation kinetics. Across species, metabolic strain is associated with oxidative vulnerability and altered substrate handling - amplifying damage during cooling, cryo

GSK3α-mediated regulation of the RNA demethylase fat mass and obesity-associated protein (FTO) during murine spermatogenesis.

Idiopathic male infertility is a rising global concern characterised by ejaculatory defects, absence or low sperm count with abnormal morphology, and poor sperm motility. Deciphering the aetiology of male infertility requires a fundamental understanding of multiple spermatogenic events, including sperm maturation. Glycogen synthase kinase 3 paralog-α (GSK3α) plays a critical role in sperm maturation, specifically during epididymal motility, capacitation and hyperactivation. Methylation modulates mRNA transport, stability, turnover, and translational efficiency to meet cellular requirements. Th

Mutations in TTC29, Encoding an Evolutionarily Conserved Axonemal Protein, Result in Asthenozoospermia and Male Infertility.

In humans, structural or functional defects of the sperm flagellum induce asthenozoospermia, which accounts for the main sperm defect encountered in infertile men. Herein we focused on morphological abnormalities of the sperm flagellum (MMAF), a phenotype also termed "short tails," which constitutes one of the most severe sperm morphological defects resulting in asthenozoospermia. In previous work based on whole-exome sequencing of a cohort of 167 MMAF-affected individuals, we identified bi-allelic loss-of-function mutations in more than 30% of the tested subjects. In this study, we further an

Role of the Ca2+-ATPase Pump (SERCA) in Capacitation and the Acrosome Reaction of Cryopreserved Bull Spermatozoa.

Cryopreservation of sperm is a resource used for artificial insemination. In the case of bovines, it ensures the reproduction of animals through cells from males with a highlighted genetic background. The fertilisation capacity of sperm cells is achieved after capacitation, a process that includes subcellular modifications that could be altered by cryopreservation. Intracellular calcium handling plays a crucial role in the development of capacitation, culminating in the acrosomal reaction. SERCA protein is responsible for calcium reuptake into the acrosome, which is one of the main calcium res

"Omics" of human sperm: profiling protein phosphatases.

Phosphorylation is a major regulatory mechanism in eukaryotic cells performed by the concerted actions of kinases and phosphatases (PPs). Protein phosphorylation has long been relevant to sperm physiology, from acquisition of motility in the epididymis to capacitation in the female reproductive tract. While the precise kinases involved in the regulation of sperm phosphorylation have been studied for decades, the PPs have only recently received research interest. Tyrosine phosphorylation was first implicated in the regulation of several sperm-related functions, from capacitation to oocyte bindi
