Characterization of antilipolytic action of polyamines in isolated rat adipocytes.

The interactions of polyamines with the lipolytic system were studied in isolated rat adipocytes. Spermine, spermidine and putrescine significantly inhibited adenosine deaminase-stimulated lipolysis. An antilipolytic effect of spermine was detectable at a concentration of 0.25 mM (P less than 0.05). At a concentration of 10 mM all three polyamines inhibited the stimulated lipolysis by 50-60% (P less than 0.001). In addition, spermine enhanced the antilipolytic sensitivity of insulin. Spermine (1 mM) decreased the half-maximal inhibitory concentration of insulin from 320 +/- 70 pM to 56 +/- 20 …

Engineered polyaramides for extraction of bioamines from water.

Biogenic amines (BAs) are prevalent in fermented foods, protein-rich meats, and brewed food additives. High concentrations of BAs can induce health issues such as headaches, high blood pressure, and palpitations. Polyaramides (PAs) prepared from the reaction of trimesoyl chloride with ethylene diamine (ED), p-xylene diamine (PXD), and m-xylene diamine (MXD) and characterized using FTIR, XRD, SEM, TGA, and DLS measurements. All PAs were used for the removal of five biogenic amines (i.e., putrescine, spermidine, spermine, tryptamine, and tryptophan) from water. All PAs showed high removal effici…

Serum L-ornithine-derived polyamines as indicators of Parkinson disease progression.

Prior studies reported that biospecimen concentrations of L-ornithine-derived polyamines (ODPAs) and their metabolites differentiated Parkinson disease (PD) from healthy controls. We investigated ODPA metabolism in CSF and serum specimens from well-characterized, unmedicated PD patients to explore correlations with disease severity, progression, and PD clinical subtypes using data from Unified PD Rating Scale (UPDRS) assessments. We developed targeted assays for L-ornithine and the ODPAs putrescine, spermine, spermidine, and the polyamine metabolites N8-acetylspermidine and N-acetylputrescine.…

Reduction of spermine synthase enhances autophagy to suppress Tau accumulation.

Precise polyamine metabolism regulation is vital for cells and organisms. Mutations in spermine synthase (SMS) cause Snyder-Robinson intellectual disability syndrome (SRS), characterized by significant spermidine accumulation and autophagy blockage in the nervous system. Emerging evidence connects polyamine metabolism with other autophagy-related diseases, such as Tauopathy, however, the functional intersection between polyamine metabolism and autophagy in the context of these diseases remains unclear. Here, we altered SMS expression level to investigate the regulation of autophagy by modulate…

[Polyamines in microorganisms sensitive and resistant to antibiotics].

Intracellular contents of polyamine dancyl derivatives were determined by the method of thin-layer chromatography in Coli and typhoid fever bacteria sensitive to antibiotics and their variants resistant to streptomycin, levomycetin, chlortetracyline and neomycin. Increased levels of spermidine and putrescin in the antibiotic resistant cultures were found. The increased levels of the polyamines correlated with changes in the content of nucleic acids. It is supposed that the increase in the content of the polyamines in the antibiotic resistant bacteria was the result of accumulation of nucleic a…

Recent developments in glycosyl urea synthesis.

The area of sugar urea derivatives has received considerable attention in recent years because of the unique structural properties and activities that these compounds display. The urea-linkage at the anomeric center is a robust alternative to the naturally occurring O- and N-glycosidic linkages of oligosaccharides and glycoconjugates, and the natural products that have been identified to contain these structures show remarkable biological activity. While methods for installing the β-urea-linkage at the anomeric center have been around for decades, the first synthesis of α-urea glycosides has b

Ribosomal distribution in a polyamine auxotroph of Escherichia coli.

The distribution of ribosomal particles has been studied in a polyamine-deficient mutant of Escherichia coli by sucrose gradient centrifugation analysis. Lysates from starved cells contained less 70S monomers and 30S subunits but more 50S particles than those prepared from bacteria supplemented with putrescine. The addition of the polyamine to putrescine-depleted cells induced a rapid change of the ribosomal profile. A similar effect could be obtained in vitro by equilibrium dialysis against a polyamine-containing solution. The ribosomal pattern obtained from starved bacteria was specific for

Antitrypanosomal effects of polyamine biosynthesis inhibitors correlate with increases in Trypanosoma brucei brucei S-adenosyl-L-methionine.

We reported recently that administration of ([(Z)-4-amino-2-butenyl]methylamino)-5'-deoxyadenosine (MDL 73811), an enzyme-activated irreversible inhibitor of S-adenosyl-L-methionine decarboxylase (AdoMetDC; EC 4.1.1.50), a key enzyme in the synthesis of spermidine, cures African trypanosome infections in mice. The precise mechanism of action of MDL 73811 was not clear because a rapid disappearance of trypanosomes from the bloodstream of treated rats occurred before significant depletion of spermidine. Administration of MDL 73811 to Trypanosoma brucei brucei-infected rats resulted in a 70% decr…

Dysregulated polyamine metabolism in neurological disorders: molecular mechanisms and therapeutic opportunities.

Polyamines (PAs), principally putrescine (Put), spermidine (Spd), and spermine (Spm), are ubiquitous aliphatic polycations that regulate nucleic-acid interactions, ion-channel activity, autophagy, redox balance, proteostasis, and immune signaling. Growing genetic, multi-omics, and experimental evidence indicates that disruption of PA biosynthesis, catabolism, acetylation, and transport contributes to neurological disease. In Alzheimer's disease (AD), altered PA flux intersects with Tau and amyloid-β (Aβ) pathology, methylation imbalance, oxidative stress, and impaired autophagic clearance. In …

Effects of human growth hormone on blood polyamines in hypopituitarism.

Blood levels of polyamines were evaluated in hypopituitary patients after i.m. injection of human growth hormone (hGH). After hGH administration polyamine concentration increased significantly in the first 60 minutes. The concentration of spermidine and spermine increases more in patients previously untreated or treated intermittently with hGH than in patients under continuous treatment. The results suggest that in human as well as in experimental models, polyamines could be involved in the growth process and that hGH probably stimulates polyamine synthesis.

Ethanol and polyamine metabolism: physiologic and pathologic implications: a review.

This review summarizes the effects of ethanol on the metabolism of polyamines (putrescine, spermidine, and spermine), which are endogenous polycations required for cell physiology, growth, and differentiation. Polyamines regulate DNA, RNA, and protein syntheses; stabilize ribosomes, membranes, and nucleic acids; and protect the cell against lipid peroxidation. Polyamine biosynthesis and catabolism may change after acute and chronic ethanol treatment, thereby playing a negative or positive role in the ethanol-induced injury on liver, extrahepatic tissues, embryo, and fetus.

In vivo and in vitro effects of some plant hormones on rat erythrocyte carbonic anhydrase and glucose-6-phosphate dehydrogenase activities.

The present study was undertaken to determine in vivo and in vitro effects of some plant growth regulators on rat erythrocyte carbonic anhydrase (CA) and glucose-6-phosphate dehydrogenase (G6PD) activities. Both in vivo and in vitro, spermidine and kinetin did not affect enzymatic activities of CA and G6PD, whereas putrescine decreased these activities, and abscisic acid increased them. Since plants use such growth regulators, their effects should be considered on mammals consuming them since they may possess important biological effects.

Exogenous Melatonin Enhances the Cold Tolerance of Harvested Peach (Prunus persica Batsch) Fruit by Elevating SAMDC and Decreasing ACC Synthase Activities.

Exogenous melatonin can alleviate cold injury in post-harvested fruit during fruit refrigeration. The mechanism of melatonin-mediated cold t … tonin alleviated cold injury in peach fruit and elevated SAMDC activity and, thereby, the levels of spermidine and spermine. More importantly, ACC synthase activity, the ACC level and the ethylene emission rate decreased. These results hint that exogenous melatonin enhanced the tolerance of peach fruit to cold stress by elevating SAMDC and decreasing ACC act …

Evaluation of platinum drug toxicity resulting from polyamine catabolism.

Polyamines, spermidine (Spd) and Spermine (Spm), are polycations that serve a number of important biological functions. The tissue contents of polyamines are tightly regulated through their cellular import and export, as well as their metabolism (anabolism and catabolism). Polyamine catabolism in mediated via the spermidine/spermine N1-acetyltransferase (SAT1)/acetylpolyamine oxidase (APOX) cascade and oxidation of Spm by spermine oxidase (SMOX). The expression of SAT1 and SMOX increases in injured organs in response to trauma, ischemia/reperfusion, sepsis, and exposure to toxic compounds. Cis…
