The Loeffler's methylene blue stain: an inexpensive and rapid method for detection of Helicobacter pylori.

Loeffler's methylene blue, commonly used as a counter-stain for acid fast bacilli, was used to detect Helicobacter pylori in paraffin sections and touch smears of gastric mucosal biopsies from 15 patients with duodenal ulcer and 35 with non-ulcer dyspepsia, and the results were compared with the modified Giemsa stain. The time taken to stain smears by Loeffler's methylene blue was approximately 10 min and the results correlated well with those stained by the modified Giemsa stain. However, the Loeffler's methylene blue method was found to be simpler, quicker and cheaper than the modified Giems

The innermost cell layer of the outer root sheath is positive with Ki-67.

The expression of a cell proliferation-associated human nuclear antigen was immunohistochemically studied in human anagen hair and hair follicles using the monoclonal antibody Ki-67. The reaction of Ki-67 in mature anagen hair follicles was observed in the hair matrix cells and outer root sheath (ORS) cells. Nuclear staining was seen in a small number of matrix cells and in some ORS cells; this finding corresponded to the thymidine or bromodeoxyuridine labeling studies previously reported. In addition, there were two different patterns of cytoplasmic staining in the ORS: strong staining of the

Recent Advancements in Phospholipid Metabolic Labeling Using Clickable Probes.

Herein, we present a perspective on recent advancements in the field of metabolic labeling as a method for interrogating phospholipids in terms of their biosynthesis, trafficking, and localization. Phospholipids are crucial for establishing cell morphology and signaling processes mediated by interactions between nearly all relevant cellular substrates. Bioorthogonal chemistry has provided a convenient platform in which clickable probes that mimic biologically active precursors are deployed to generate labeled versions of phospholipids in cells, enabling the study of downstream lipid activities

Standardized Workflow for High-Content Imaging and Data Processing.

To address the challenges of low reproducibility due to operational variations in scientific research imaging and the complexity of analyzing a large number of samples using conventional methods, we introduce a protocol for staining HT22 cells with PI and Hoechst 33342 and then imaging and analyzing the data using the High-Content Imaging System. This workflow aims to establish robust imaging setting parameters and stringent quality control standards. It demonstrates the instrument's imaging settings and the workflow for three different image processing methods provided by the instrument: Mult

Histopathological findings and staining performance in cutaneous nodules from a cat with Leishmania amazonensis infection.

Cutaneous leishmaniasis (CL) is a zoonotic disease with wide geographic distribution and significant public health impact. In Brazil, the main etiological agents are Leishmania braziliensis and Leishmania amazonensis. In animals, the disease usually presents as a chronic condition characterized by poorly healing skin lesions, oral and ocular involvement, and cutaneous nodules, most commonly affecting the head and neck regions. Diagnosis may be achieved using molecular techniques, immunohistochemistry, cytology, histopathology, and culture. Despite the endemic nature of CL in several regions, h

Assessment of Pollen Viability and Germination Through Histochemical, In Vivo and In Vitro Methods.

Pollen viability is a key parameter for understanding the functioning of plant reproductive systems and for supporting strategies aimed at genetic resource conservation and plant breeding. Accurate assessments of this attribute are essential for identifying genotypes with a higher reproductive rate, monitoring prezygotic barriers, testing compatibility between individuals, and planning controlled crosses, especially in cases of floral asynchrony or spatial isolation. Accordingly, this chapter compiles the main methods used for this purpose, providing detailed and up-to-date protocols. It cover

Near-Freezing-Temperature Golgi Neuronal Staining for X-ray Imaging of Human Brain.

Achieving detailed neuronal structural information in large-volume brain tissue has been a longstanding challenge in human brain imaging. A key obstacle arises from the trade-off between staining efficiency and tissue autolysis. Traditional Golgi staining, typically conducted at room temperature or 37 °C to optimize staining efficiency, leads to rapid autolysis of brain tissue, resulting in the loss of fine structural details. Here, a near-freezing temperature (NFT) staining strategy in post-mortem frozen (PMF) human brain samples are presented, using a mercury chloride-based method under ice-

An Optimized Positive Staining Protocol Method for Clear Visualization of Extracellular Vesicles by TEM Using Uranyless and Lead Citrate.

Transmission electron microscopy (TEM) is widely used for visualizing extracellular vesicles (EVs) due to its nanometer-scale resolution. However, existing sample preparation protocols often lack sufficient methodological detail, requiring researchers to rely on trial-and-error approaches to achieve high-quality images. Here, we report an optimized positive-staining method that utilizes the sequential application of Uranyless and lead citrate with an intermediate buffered wash. This approach yields high image contrast, preserves EV morphology, and significantly reduces background artifacts. St

Unusual Icteric Discoloration of Single Donor Platelets in Platelet Additive Solution With Container Staining: A Rare Observation.

This report describes an unusual case of an apheresis platelet (AP) unit in platelet additive solution (PAS) exhibiting marked icteric discoloration and bag staining, despite sterile cultures and initially preserved platelet morphology. Donor evaluation revealed mild unconjugated hyperbilirubinemia, implicating donor-derived pigments as the source of discoloration. By Day 5, reduced platelet function was observed. This case underscores that icteric appearance in PAS-suspended platelets, though rare, may signal donor-related pigment transfer and warrants reevaluation of current visual inspectio

Live-cell painting: Image-based profiling in live cells using acridine orange.

Image-based profiling has been used to analyze cell health, drug mechanism of action, CRISPR-edited cells, and overall cytotoxicity. Cell Painting is a broadly used image-based assay that uses morphological features to capture how cells respond to treatments. However, this method requires cell fixation for staining, which prevents examining live cells. To address this limitation, here we present Live-Cell Painting (LCP), a high-content method based on acridine orange, a metachromatic dye that labels different organelles and cellular structures. We began by showing that LCP can be applied to fo

Supported Immunohistochemical Staining of Mucins.

Immunohistochemistry (IHC) staining is the most common method to detect the distribution and localization of biomarkers in different parts of a tissue. Antibodies for tandem repeat peptide of mucins are very popular, but antibodies for glycosylation or others are also used. IHC for mucin is the same protocol as IHC for others. This description includes IHC according to ABC method for processed formalin-fixed paraffin-embedded (FFPE) tissues. Protocol of in situ hybridization is also shown.

An algorithm-based technique for counting mitochondria in cells using immunohistochemical staining of formalin-fixed and paraffin-embedded sections.

PURPOSE: Visualizing mitochondria in cancer cells from human pathological specimens may improve our understanding of cancer biology. However, using immunohistochemistry to evaluate mitochondria remains difficult because almost all cells contain mitochondria and the number of mitochondria per cell may have important effects on mitochondrial function. Herein, we established an objective system (Mito-score) for evaluating mitochondria using machine-based processing of hue, saturation, and value color spaces. METHODS: The Mito-score was defined as the number of COX4 (mitochondrial inner membrane)

A Suggested Mechanism for Green Discoloration of the Postmortem Brain.

In the putrefied brain, the cortex and basal ganglia show dark-grayish to green discoloration due to sulfhemoglobin formed from hydrogen sulfide (H 2 S) produced by endogenous bacteria and hemoglobin. In this study, we propose and demonstrate another mechanism of green discoloration in the brain. The formalin-fixed brain of a cadaver donated for medical education with no putrefaction was used. Half of the brain was immersed in sodium hydrosulfide solution, to imitate the H 2 S produced by bacteria. This half showed greenish discoloration, mainly in the basal ganglia and cortex. The other half

Techniques in Staining of Rodent and Human Olfactory Tissue.

Staining of olfactory tissue allows for evaluation of the organization of specific cell types within the specimen and allows for assessment in abnormalities of function that may be related to changes in normal tissue architecture. Histochemical staining and immunohistochemical (IHC) techniques are the most common methods for visualizing olfactory epithelium and olfactory bulbs. Here we describe the method of IHC for olfactory tissue utilizing both fluorescent and peroxidase-based methods of visualization.

Case Report: Giant Oral Ulcers Attributed to Cytomegalovirus Infection in a Patient with AIDS.

Oral ulcers are often neglected in patients with AIDS. However, giant oral ulcers are uncommon and are usually suspected to be malignant lesions. Our study presents a case of giant ulcers in an AIDS patient that were initially suspected to be oral cancer. To assist with diagnosis, conventional microbiological tests, metagenomic next-generation sequencing, and a pathological examination were conducted on oral lesion biopsy specimens. The case was finally confirmed via hematoxylin-eosin staining and immunohistochemical staining to be a cytomegalovirus infection.
