3D self-locking granular metamaterials.

Mechanical metamaterials derive unique properties from microarchitectures, yet most designs remain static once fabricated. Here, we propose a three-dimensional (3D) self-locking granular metamaterial composed of rigid ellipsoidal particles with flexible hook-like appendages inspired by Xanthium seed burs. The hooks interlock to form self-confined structures that fail via sequential stepwise unhooking under tension, greatly enhancing ductility. Under shear, the material transitions from an initially low-resistance, fluid-like state to a rigid, solid-like state once hooks engage, exhibiting a sl

Smart Secondary Metabolites in Marine Environments: Exploring the Oxasqualenoid Dehydrothyrsiferol.

Dehydrothyrsiferol (DT), a brominated oxasqualenoid from the red alga Laurencia viridis, represents a compelling example of this framework. This review establishes DT as a model Smart Secondary Metabolite based on the convergence of a unique molecular architecture of rigid stereogroups connected by flexible bonds; a high metabolic yield (0.42% w/w of crude extract); potent selective bioactivity against kinetoplastids and drug-resistant tumors; multi-target modulation of protein phosphatase 2A (PP2A) and cell-surface integrins; and distinctive chemotaxonomic relevance within Macaronesian commun

Holocene ocean-atmosphere coupling and Mediterranean sensitivity to Atlantic circulation: Lessons from the Late Bronze Age collapse.

Climate variability has profoundly influenced societies in the semiarid Eastern Mediterranean. Proxy records indicate progressive mid-Holocene drying culminating in severe droughts ∼1250 BCE, coincident with the Late Bronze Age collapse (1300 to 900 BCE). Using the EC-Earth3-Tr8K transient Holocene simulation, we reconstruct regional hydroclimate evolution and its underlying mechanisms. The model reproduces a gradual orbitally driven aridification, spatially heterogeneous across the region, with persistent drying in the Levant but enhanced effective moisture in parts of the Balkans and Anatoli

Intragenic viral silencer element regulates HTLV-1 latency via RUNX complex recruitment.

Retroviruses integrate their genetic material into the host genome, enabling persistent infection. Human T cell leukaemia virus type 1 (HTLV-1) and human immunodeficiency virus type 1 (HIV-1) share similarities in genome structure and target cells, yet their infection dynamics differ drastically. While HIV-1 leads to high viral replication and immune system collapse, HTLV-1 establishes latency, promoting the survival of infected cells and, in some cases, leading to leukaemia. The mechanisms underlying this latency preference remain unclear. Here we analyse blood samples from people with HTLV-1

Progress of Programmed Cell Death in Steroid-Induced Osteonecrosis of the Femoral Head.

Steroid-induced osteonecrosis of the femoral head is a severe osteoarticular condition resulting from glucocorticoid overuse, characterized by femoral head bone structure collapse and cell death, now predominant among nontraumatic femoral head necroses. The increasing clinical use of glucocorticoids has led to a rise in the incidence of steroid-induced osteonecrosis of the femoral head, yet its precise molecular mechanisms remain incompletely understood, posing challenges for clinical management. This review proposes that the "GC-induced metabolic-inflammatory-oxidative stress vicious cycle" s…

Integrated bioprinting of trachea-like structures based on tissue-specific bioink.

Segmental tracheal reconstruction remains clinically challenging due to insufficient blood supply and mechanical instability in current graft materials. Inspired by the natural trachea's alternating cartilage-fiber structure, we developed a 3D-bioprinted chondro-fibrous integrated trachea (CFT) using customized tissue-specific hydrogels modified with glucomannan-peptide copolymer (GM-P). This design mimics the native tissue's layered organization, combining cartilage-like rings (from iPSC-derived chondrocytes) and fibrous connective layers (from fibroblasts) through precise bioprinting. The GM

FAP-α is an effective tool to evaluate stroma invasion of lung adenocarcinoma.

The main difficulty in the diagnosis of atypical in situ adenocarcinoma lies in the distinction between true and false stromal invasion. Moreover, how to identify local alveolar wall collapse in situ lung adenocarcinoma and how to identify whether the trapped adenoid structure around scar is an invasion component have become the key points for accurate diagnosis of lung adenocarcinoma. In the present study, we detected 40 cases of lung adenocarcinoma in situ and 40 cases of invasive adenocarcinoma by using immunohistochemical techniques. We found FAP-α had not immunreactivity in the stroma of

Direct Single Crystal to Amorphous Transformation and Memory Effect in AlPO4-17.

Pressure induced amorphization provides a distinct route to prepare novel amorphous materials. Single crystals of the porous aluminophosphate AlPO4-17 directly transform to an amorphous state beginning at 0.6 GPa, without fragmentation into polycrystalline material. Apart from a reduction in dimensions, the amorphous material retains the form of the initial single crystal. Remnant crystalline domains in the amorphous material also preserve the initial orientation of the single crystal. X-ray diffraction indicates the compression of the structure around the empty pores in the xy plane and such

Kienböck's disease: preventing disease progression in early-stage disease.

Currently Kienböck's disease remains an 'unsolved' problem in hand surgery. Different factors have been associated with the avascular necrosis of the lunate. Mechanical, vascular and biological factors, alone or in combination, may have an influence in the aetiopathogenesis and determine the progress of the disease and even the results of the treatment. This is especially relevant in the early stages, in which conservative or surgical treatment may modify the natural history of the disease, maintaining the lunate structure and thus preserving the joint surfaces. There are multiple surgical tre

Semaphorin3F Drives Dendritic Spine Pruning Through Rho-GTPase Signaling.

Dendritic spines of cortical pyramidal neurons are initially overproduced then remodeled substantially in the adolescent brain to achieve appropriate excitatory balance in mature circuits. Here we investigated the molecular mechanism of developmental spine pruning by Semaphorin 3F (Sema3F) and its holoreceptor complex, which consists of immunoglobulin-class adhesion molecule NrCAM, Neuropilin-2 (Npn2), and PlexinA3 (PlexA3) signaling subunits. Structure-function studies of the NrCAM-Npn2 interface showed that NrCAM stabilizes binding between Npn2 and PlexA3 necessary for Sema3F-induced spine p

Running on the verge of collapse in lyophilization: What is the impact of the edge vial effect on colloidal protein stability and particle morphology?

Lyophilization is frequently employed to stabilize sensitive biopharmaceuticals. The potential importance of the edge vial effect, defined as the discrepancy in temperature and drying behavior between vials located in different positions of the batch, attracts considerable attention from the lyophilization community. Here, we lyophilized two model fusion-protein formulations by applying a design space representation to select conservative as well as aggressive lyophilization protocols on the verge of structural collapse of the cake. The effect of vial position and number of neighbor vials on c

Cooperative effects of natural acetylenic acetogenins fromPorcelia macrocarpaon lipid packing and mechanical stability of Langmuir monolayers as protozoan membrane models.

This study investigates the synergistic interfacial behavior of two natural bioactive acetylenic acetogenins, (2S,3R,4R)-3 hydroxy-4-methyl-2-(n-eicos-11'-yn-19'-enyl)butanolide (1) and (2S,3R,4R)-3-hydroxy-4 methyl-2-(n-eicos-11'-ynyl)butanolide (2), obtained from Brazilian plant Porcelia macrocarpa (Annonaceae) and their interactions with membrane models using Langmuir films. Previous biological studies demonstrated that mixtures of 1 and 2 exhibit enhanced antiparasitic activity compared to the isolated compounds, although the physicochemical basis for this synergism remained unclear. Free-vascularized medial femoral condyle bone transfer in the treatment of scaphoid nonunions.

BACKGROUND: Scaphoid nonunions with associated avascular necrosis and carpal collapse have proven difficult to treat reliably. This study outlines the rationale and approach to the use of a free-vascularized medial femoral condyle bone graft and the authors' experience with it in the treatment of scaphoid nonunions. METHODS: A retrospective review was conducted to identify all patients with scaphoid nonunions with avascular necrosis and carpal collapse treated with a medial femoral condyle bone graft. Between July of 2004 and August of 2007, 12 such patients (12 men and no women), with a mean

Self-Cross-Linked Collapsible Starch Aerogels Loaded with Ag Nanoparticles: A Platform for Multiphase SERS Detection of Trace Pollutants in Complex Matrices.

Regulating "hotspots" in surface-enhanced Raman scattering (SERS) constitutes a key research focus, whereas achieving highly sensitive and full-coverage detection of multiphase analytes remains a major obstacle to the widespread application of SERS technology. To address the drawbacks that most current SERS substrates are confined to single-phase detection and that multifunctional integrated substrates are in short supply, constructed three-dimensional Ag nanoparticle (AgNPs)-decorated cassava starch aerogels (ACA) as SERS substrates through a green and facile gelatinization, retrogradation, a

CD36-Pocket Targeting Induces Metabolic Collapse and Disulfidoptosis for Sonodynamic Immunometabolic Therapy of Liver Cancer.

Obesity-driven lipid accumulation in hepatocellular carcinoma (HCC) establishes a coupled metabolic-immunosuppressive niche that fuels tumor progression and undermines anti-tumor immunity. Through multiplexed immunofluorescence profiling of clinical HCC tissues stratified by visceral adiposity, we explicitly map this lipid-immunosuppressive landscape and demonstrate that fatty acid translocase CD36 overexpression directly correlates with FoxP3+ Treg infiltration and CD8+ T cell exclusion, defining a clinically actionable immunometabolic vulnerability. To therapeutically exploit this vulnerabil

Anatomical Basis of Obstructive Sleep Apnoea: A Review of Randomized Controlled Trials.

Repeated obstruction and closure of the upper airway, sporadic hypoxic episodes, and sympathetic activity are symptoms of obstructive sleep apnea (OSA). Obstructive sleep apnoea is due to a combination of altered upper airway structure and muscular function, a low arousal threshold and increased loop gain. Although recurrent upper airway (UA) collapse during sleep is the most frequent clinical hallmark of OSA, the exact cause of this collapse is unknown. Furthermore, while continuous positive airway pressure aids in the management of OSA, many patients find it intolerable. As a result, a bette

Rapid plasticity of dendritic spine: hints to possible functions?

Contrary to a century-old belief that dendritic spines are stable storage sites of long term memory, the emerging picture from a recent flurry of exciting observations using novel high resolution imaging methods of living cells in culture is that of a dynamic structure, which undergoes fast morphological changes over periods of hours and even minutes. Concurrently, the nature of stimuli which cause formation or collapse of dendritic spines has changed from a mysterious Hebbian-governed plasticity producing stimulus to the more trivial activation of the synapse by strong/weak stimulation. The m

Scaphoid Nonunion Arthroscopic-Assisted Reduction and Percutaneous Fixation With Distal Radius Autologous Bone Graft.

Scaphoid fractures are common and often progress to nonunion due to the unique vascularity. When nonunion occurs, patients are at risk of developing carpal collapse and arthritis if not managed appropriately with traditional surgical management. However, open surgical management puts the fragile vascular supply and extrinsic soft tissue stabilizers of the carpus at risk, compromising healing and clinical outcomes. Arthroscopy allows for the preservation of these structures and improved visualization of fracture debridement, reduction, and alignment. We describe an arthroscopic-assisted reducti

Disease-Specific Finite element Analysis of the Foot and Ankle.

Finite-element analysis is a computational modeling technique that can be used to quantify parameters that are difficult or impossible to measure externally in a geometrically complex structure such as the foot and ankle. It has been used to improve our understanding of pathomechanics and to evaluate proposed treatments for several disorders, including progressive collapsing foot deformity, ankle arthritis, syndesmotic injury, ankle fracture, plantar fasciitis, diabetic foot ulceration, hallux valgus, and lesser toe deformities. Parameters calculated from finite-element models have been widely
