Postnatal development of palatal and laryngeal taste buds in the hamster.

Mammalian taste buds are distributed within several distinct subpopulations, innervated by branches of three cranial nerves. These taste bud populations originate and mature at different times in various mammalian species and are thought to play differential roles in the control of taste-mediated behaviors. The hamster is a common animal for the electrophysiological study of the gustatory system, and it has been shown that taste buds innervated by the IXth nerve develop postnatally in this species. To delineate further the development of the gustatory system of hamsters, we quantified the numb…

Taste-responsive neurons and their locations in the solitary nucleus of the hamster.

The solitary nucleus (nucleus tractus solitarii), the first central relay for taste in mammals, was studied anatomically and physiologically in the golden hamster (Mesocricetus auratus). Activity of neurons to anterior tongue stimulation with sucrose, NaCl and KCl were extracellularly recorded. Electrolytic lesions or horseradish peroxidase deposits allowed subsequent localization of recording sites. Anterior tongue taste-responsive sites were restricted to a very small part of the rostral pole of the solitary nucleus, which is about 3% of the entire nucleus. Sites were confined to the rostral…

Increased aversion to bitter-tasting fluids in the Brattleboro rat.

The large daily water intake of Brattleboro rats with diabetes insipidus (BDI) is believed to be due to the volume of depletion caused by th … to drink. A two bottle preference test for quinine showed that the BDI rats actually have a higher taste threshold for the drug in solution in drinking water than the control LE rats. This suggests that the increased aversion to quinine in solution when it is the sole drinking source cannot be due to the BDI rats tasting the quinine at a lower concentr …

The effects of various barbiturates on LiCl induced taste aversion.

The effects of various barbiturates on LiCl induced taste aversion were examined. Rats were adapted to a 23 hr and 50 min water deprivation schedule. On the Treatment Day, animals were offered a novel 0.125% saccharin solution and then administered 3.0 mEq/kg LiCl. The saccharin solution was presented again on three subsequent Test Days. Fifteen minutes prior to drinking on the first Test Day animals were injected subcutaneously with either 10, 30, and 50 mg/kg Amobarbital, 3, 9, and 15 mg/kg Pentobarbital, 40, 80, and 120 mg/kg Barbital, or 1, 3, and 9 mg/kg Hexobarbital. Results indicate tha…

Taste-aversion-prone (TAP) rats and taste-aversion-resistant (TAR) rats differ in ethanol self-administration, but not in ethanol clearance or general consumption.

Taste-aversion (TA)-prone (TAP) rats and TA-resistant (TAR) rats have been developed by means of bidirectional selective breeding on the basis of their behavioral responses to a TA conditioning paradigm. The TA conditioning involved the pairing of an emetic-class agent (cyclophosphamide) with a novel saccharin solution as the conditioned stimulus. Despite the absence of ethanol in the selective breeding process, these rat lines differ widely in ethanol self-administration. In the current study, blood alcohol concentrations (BACs) were determined after 9 days of limited (2 h per day) access to …

Contribution of kelp dashi liquid to sustainable maintenance of taste sensation and promotion of healthy eating in older adults throughout the umami-taste salivary reflex.

INTRODUCTION: Taste decline, including taste loss in older adults, leads to malnutrition and frailty. In a super-aging society, improving taste decline and maintaining taste sensation are crucial for the wellbeing of older adults. Hyposalivation frequently affects older individuals and is the leading cause of taste decline in older adults. Treating taste decline, including taste loss, in older adults presents challenges due to the limited sustainable methods for increasing saliva production, except for drug therapy, which may lead to adverse effects. Umami-taste stimulation results in a prolon…

Expression profile of the zinc transporter ZnT3 in taste cells of rat circumvallate papillae and its role in zinc release, a potential mechanism for taste stimulation.

Zinc is an essential trace element, and its deficiency causes taste dysfunction. Zinc accumulates in zinc transporter (ZnT)3-expressing presynaptic vesicles in hippocampal neurons and acts as a neurotransmitter in the central nervous system. However, the distribution of zinc and its role as a signal transmitter in taste buds remain unknown. Therefore, we examined the distribution of zinc and expression profiles of ZnT3 in taste cells and evaluated zinc release from isolated taste cells upon taste stimuli. Taste cells with a spindle or pyriform morphology were revealed by staining with the fluo…

Taste-Masking Effect of Chlorogenic Acid (CGA) on Bitter Drugs Evaluated by Taste Sensor and Surface Plasmon Resonance on the Basis of CGA-Drug Interactions.

The purpose of this study was to evaluate the taste-masking effects of chlorogenic acid (CGA) on bitter drugs using taste sensor measurements and surface plasmon resonance (SPR) analysis of CGA-drug interactions. Six different bitter drugs were used: amlodipine besylate (AMD), diphenhydramine hydrochloride (DPH), donepezil hydrochloride (DNP), rebamipide (RBM), diclofenac sodium (DCF) and etodolac (ETD). Taste sensor outputs were significantly inhibited by the addition of CGA to all drugs. The inhibition ratio of the taste sensor output decreased in the following order DPH>DNP>AMD≈DCF≈RBM≈ETD.…

Taste Papillae-associated Salivary Gland Ducts Contribute to Immune Surveillance.

The lingual surface is composed of a tough epithelium that affords effective protection from the resident and food-borne microbiota. The circumvallate taste papillae (CVP) in the back of the tongue are associated with underlying minor salivary glands called von Ebner's gland (VEG) that drains its secretions into the trenches of the papillae. The taste buds are exposed to the lingual surface through taste pores, to enable interaction with tastants. These openings may be exploited by pathogens for infection. Mucosa-associated lymphoid tissue (MALT) such as those in the gut epithelium, tonsils an…

Taste cells depend on axon proximity to generate presynaptic sites.

The turnover and re-establishment of peripheral taste synapses is vital to maintain connectivity between primary taste receptor cells and the gustatory neurons which relay taste information from the tongue to the brain. Despite the importance of neuron-taste cell reconnection, the mechanisms governing synapse assembly in the taste bud are largely unknown. To determine whether nerve fiber connectivity is an initiating factor for the recruitment of presynaptic machinery in taste receptor cells, we use the expression of CALHM1 and Bassoon to identify presynaptic sites in type II (sweet, umami, bi…

Bitter Taste Receptor Agonists Induce Apoptosis in Papillary Thyroid Cancer.

BACKGROUND: Papillary thyroid carcinoma (PTC) is the most common thyroid malignancy, with a 20% recurrence rate. Bitter taste receptors (T2Rs) and their genes (TAS2Rs) may regulate survival in solid tumors. This study examined T2R expression and function in PTC cells. METHODS: Three PTC cell lines (MDA-T32, MDA-T68, and MDA-T85) were analyzed for expression using RT-qPCR and immunofluorescence. Live cell imaging measured calcium responses to six bitter agonists. Viability and apoptosis effects were assessed using crystal violet and caspase 3/7 activation assays. Genome analysis of survival wa…

Lineage tracing of Twist2-expressing cells in mouse taste buds.

OBJECTIVES: It is widely accepted that Taste bud cells originate from the epithelium; however, evidence indicates that these cells are also derived from the mesenchyme beneath the epithelium. In this study, the cell lineage expressing Twist2, a transcription factor that is specifically expressed in the mesenchyme within taste buds, were examined to gain a deeper understanding of its biological characteristics. METHODS: Immunohistochemical analyses and taste organoid cultures were performed using Twist2 lineage-tracing reporter mice (Twist2-Cre mice crossed with tdTomato mice) to identify Twis…
