CD40, a cell surface molecule found on B lymphocytes and other antigen presenting cells, can, when engaged by CD40 ligand (CD40L), induce gene rearrangements and isotype switching. We report here that CD40 is also expressed on thymocytes and on up to 50% of peripheral T cells from autoimmune prone strains of mice. In normal animals, CD40 is present on a small population of T cells and thymocytes. CD40 is expressed on most T cell hybridomas. We demonstrate that CD40 engagement on peripheral T cells, T cell hybridomas and thymocytes results in altered TCRValpha expression. That induced expressio…

The capacity of minocycline and tetracycline for modulation of IL-1 secretion by LPS-stimulated human monocytes was investigated in vitro. Both minocycline and tetracycline suppressed the murine thymocyte co-mitogenic bioassay of IL-1 at 2 and 4 mg/l respectively. IL-1 beta secretion by LPS-stimulated human monocytes cultured for 24h at 1 x 10(6)/ml with 0, 5, 10 and 50 mg/l minocycline or tetracycline was therefore determined by ELISA. Monocytes from five different individuals served as replicates. LPS-stimulated monocytes secreted significantly more IL-1 beta in the presence of minocycline (

Post-transplant cyclophosphamide (PTCy) and anti-thymocyte globulin (ATG) are mainstay prophylactic treatment options for graft-versus-host disease (GVHD), widely used in haploidentical stem cell transplantation. Due to a lack of prospective studies, a number of retrospective comparisons have yielded different conclusions as to which prophylaxis regimen is superior. We performed a meta-analysis of these studies to get more informed and comprehensive decisions from clinicians. Nine studies were eligible, and a total of 1674 patients were included. The combined hazard ratio (HR), relative risk (

INTRODUCTION: Autologous haematopoietic stem cell transplantation improves event-free survival and lung function and reduces skin thickening in patients with progressive diffuse cutaneous systemic sclerosis. Anti-thymocyte globulin is a key lymphoablative constituent of conditioning protocols and is administered in a weight-based dosage. However, whether anti-thymocyte globulin exposure contributes to response to autologous haematopoietic stem cell transplantation and lymphocyte reconstitution in diffuse cutaneous systemic sclerosis patients is unknown. We aimed to explore the relationship bet

Retinoic‑acid‑receptor‑related orphan nuclear hormone receptor gamma t (RORγt), a critical transcriptional factor of Th17 cells, is a potential therapeutic target for Th17-mediated autoimmune diseases. In addition, RORγt is essential for thymocyte survival and lymph node development, and RORγt inhibition or deficiency causes abnormal thymocyte development, thymus lymphoma, and lymph node defect. Recent study demonstrated that specific regulation of Th17 differentiation related to the hinge region of RORγt. In this research, we investigated the effect of RORγt inhibitor, 5,6,7,8-tetrahydrobenzo

BACKGROUND: Anti-thymocyte globulin (ATG) is a treatment option for steroid-resistant T-cell-mediated rejection after kidney transplantation. However, the extent to which immune-cell subsets can repopulate the peripheral blood is unknown.
METHODS: Six patients with steroid-resistant T-cell-mediated rejection were recruited and underwent analysis of their immune cells for 1 year after ATG administration. Multicolor flow cytometric analysis was used to evaluate the proportions of T cells, B cells, natural killer cells, and monocytes.
RESULTS: T-cell repopulation from 24% to 75% occurred in the

We describe 47 patients with lymphoma and failed prior autologous hematopoietic cell transplantation (HCT) who received TLI-ATG (anti-thymocyte globulin) conditioning followed by allogeneic HCT. Thirty-two patients had non-Hodgkin lymphoma (NHL; diffuse large B-cell lymphoma (n=19), T-cell NHL (n=6), mantle cell lymphoma (n=4) or other B-cell subtypes (n=3)), and 15 had Hodgkin lymphoma. The median follow-up was 4.9 (range, 2.1-11.9) years. The cumulative incidence of grade II-IV acute GvHD at day +100 was 12%, and the cumulative incidence of extensive chronic GvHD at 1 year was 36%. The ability of thymocytes to assess T cell receptor (TCR) signaling strength and initiate the appropriate downstream response is crucial for determining their fate. We have previously shown that a c-Cbl RING finger mutant knock-in mouse, in which the E3 ubiquitin ligase activity of c-Cbl is inactivated, is highly sensitive to TCR-induced death signals that cause thymic deletion. This high intensity signal involves the enhanced tyrosine phosphorylation of the mutant c-Cbl protein promoting a marked increase in the activation of Akt. Here we show that this high intensity signal in c-Cbl RING fin…

PURPOSE: The appearance of tumor suppressor protein 53 (p53) -/- thymocytes at an early stage of radiation-induced lymphomagenesis was investigated in the p53 heterozygous (+/-) B10 mice following a single dose of irradiation, since most thymic lymphomas manifested the loss of the wild-type p53 allele and the loss of heterozygosity was thought to be an early event critical for radiation-induced thymic lymphomagenesis in p53 +/- mice.
MATERIALS AND METHODS: The mice were exposed to a single dose (6 Gy) of irradiation to induce thymic lymphomas and, at various times after irradiation, treated w…

Thymocyte selection involves signaling by TCR engaging diverse self-peptide:MHC molecule ligands on various cell types in the cortex and medulla. Here we separately analyze early and late stages of selection to better understand how presenting cell type, ligand quality, and the timing of TCR signaling contribute to intrathymic differentiation. TCR transgenic CD4+CD8+ thymocytes (double positive (DP)) from MHC-deficient mice were stimulated using various presenting cells and ligands. The resulting CD69high cells were isolated and evaluated for maturation in reaggregate cultures with wild-type o…

The LMO2 and TAL1 genes were first identified via chromosomal translocations and later found to encode proteins that interact during normal erythroid development. Some T cell leukaemia patients have chromosomal abnormalities involving both genes, implying that LMO2 and TAL1 act synergistically to promote tumorigenesis after their inappropriate co-expression. To test this hypothesis, transgenic mice were made which co-express Lmo2 and Tal1 genes in T cells. Dimers of Lmo2 and Tal1 proteins were formed in thymocytes of double but not single transgenic mice. Furthermore, thymuses of double transg…

Peripheral T-cell lymphomas (PTL) are morphologically and immunophenotypically heterogeneous. We have examined a series of cases to determine whether this heterogeneity is reflected at the level of developmentally specific T-cell receptor (TCR) gene rearrangement. 4 of 5 cases had clonal rearrangements of TCR beta and gamma chain genes; one of these also had a probable DQ52-J immunoglobulin heavy chain gene rearrangement. 2 of the 4 TCR gamma gene rearrangements involved the most 5' J region, JP1, a characteristic of immature thymocytes. These 2 cases also had immunophenotypic features of imma…

It is known that many immunologic responses to IL-1 are antagonized by the neuropeptide alpha-melanocyte stimulating hormone (alpha-MSH). This led us to investigate the possible reciprocal effects of IL-1 and the functionally related epidermal cytokines, epidermal cell-derived thymocyte activating factor (ETAF) and IL-6, on the melanogenic effect of alpha-MSH on murine Cloudman melanoma cells. When these cells were treated with ETAF in combination with alpha-MSH or its potent analog [Nle4,D-Phe7]-alpha-MSH, the melanotropin induced increase in tyrosinase activity, and thus melanin synthesis, w

Cytokine release syndrome (CRS) is a rare but serious complication of rabbit anti-thymocyte globulin, a common induction medication for solid organ transplant recipients. We present a case of grade 4 cytokine release syndrome in a renal transplant patient in whom tocilizumab successfully salvaged the graft function and stabilized the patient. A 22-year-old woman with end-stage renal disease on hemodialysis and a complex history including systemic lupus erythematosus nephritis underwent a deceased donor kidney transplant with thymoglobulin induction. Hours postoperatively, she developed hemodyn

T cell differentiation and selection in the thymus are pivotal for establishing antigen specificity and shaping the functional repertoire of peripheral T cells. Here, we present an in vitro protocol to investigate the roles of specific target genes during early thymic development in mice utilizing retroviral vectors. We describe steps for retroviral packaging, the isolation of primary murine thymic cells, retroviral transduction, and the subsequent in vitro differentiation of thymocytes. For complete details on the use and execution of this protocol, please refer to Fan Zhao et al.1.

Graft versus host disease (GVHD) is a serious side effect of allogenic stem cell transplantation (allo-SCT). Anti-thymocyte globulin (ATG) reduces GVHD but raises the risk of infections following allo-SCT. Preventive therapy is recommended for cytomegalovirus (CMV), which is a major pathogen. Prophylactic administration of letermovir has shown a significant reduction in CMV reactivation. We hypothesized letermovir could reduce CMV reactivation and affect the outcome of allo-SCT. We retrospectively studied 88 patients who received allo-SCT with ATG for acute leukemia or myelodysplastic syndrome

BACKGROUND: Rabbit anti-thymocyte globulin (rATG) is widely used for induction immunosuppression in kidney transplantation; however, its administration carries some adverse effects. This study aimed to detect the incidence of thromboembolic events (TEEs) after kidney transplantation and the causality relationship between these events and rATG administration as induction therapy.
METHODS: This multicenter retrospective study included 654 kidney recipients from six Iranian centers receiving [Thymoglobulin® (rATG-A) or TGlobulin25™ or (rATG-B)]. Outcomes included TEEs within 30 days post-transpl

INTRODUCTION: Anti-Thymocyte Globulin (ATG) is a cornerstone in immune suppression for solid organ transplantation. The treatment is a delicate balance between complications arising from over-immunosuppression such as infections and cancer versus rejection stemming from under-immunosuppression. CD3+ T-lymphocyte measurements are frequently employed for treatment monitoring. However, this analysis is costly and not always accessible. The aim of this study was to investigate whether the total count of lymphocytes could replace CD3+ T-lymphocyte measurements based on data from our transplantation
