Gastrointestinal parasites in the immunocompromised.

PURPOSE OF REVIEW: Parasites and other infections have many effects on the gastrointestinal tract of individuals who are immunocompromised. … nd abnormal immune systems with helminth infections such as hookworm and strongyloidiasis, and with trematode infections such as schistosomiasis, are receiving increasing attention. These are discussed, together with the novel concept of using live helminths to treat inflammatory bowel disease. SUMMARY: Parasitic infections remain a significant problem for immunocomp …

Schistosomin from the snail Biomphalaria glabrata: expression studies suggest no involvement in trematode-mediated castration.

By inhibiting reproductive hormones, the neuropeptide schistosomin produced by the snail Lymnaea stagnalis plays an essential role in parasitic castration mediated by the schistosome parasite Trichobilharzia ocellata during late stage infection. Here we report on the presence and expression of schistosomin in the snail Biomphalaria glabrata, a prominent intermediate host of the parasite Schistosoma mansoni, one of the causative agents of human schistosomiasis. The deduced amino acid (aa) sequences from complementary DNAs (cDNAs) from B. glabrata contain a 17 aa signal peptide and a 79 aa matur

Diagnosis of cattle fasciolosis by the detection of a circulating antigen using a monoclonal antibody.

A monoclonal antibody (MoAb) 1C12 that reacts with a 66 kDa surface tegumental (ST) antigen of adult worms of Fasciola gigantica was used to detect circulating antigen in sera of experimentally and naturally infected cattle. A combination of rabbit anti ST-antigens and MoAb 1C12 were used to capture and detect the circulating antigen in sandwich ELISA. The dilutions of 1:1,000 of rabbit anti ST-antigens and 1:100 for MoAb 1C12 were used to reduce cross-reactivity with other trematodes' antigens. The circulating antigen of F. gigantica was demonstrated in sera of all experimentally infected ani

Trematode Proteomics: Recent Advances and Future Directions.

Trematodes cause disease in millions of people worldwide, but the absence of commercial vaccines has led to an over-reliance on a handful of monotherapies to control infections. Since drug-resistant fluke populations are emerging, a deeper understanding of parasite biology and host interactions is required to identify new drug targets and immunogenic vaccine candidates. Mass spectrometry-based proteomics represents a key tool to that end. Recent studies have capitalised on the wider availability of annotated helminth genomes to achieve greater coverage of trematode proteomes and discover new a

Temperature does not influence functional response of amphipods consuming different trematode prey.

Direct consumption on free-living cercariae stages of trematodes by non-host organisms interferes with trematode transmission and leads to reduced infections in the next suitable hosts. Consumer functional responses provide a useful tool to examine relationships between consumption rates and ecologically relevant prey densities, whilst also accounting for abiotic factors that likely influence consumption rates. We investigated how temperature influences the consumer functional response of the amphipod Gammarus lacustris towards the cercariae of three freshwater trematodes (Diplostomum, Apatemo

The life cycle and molecular phylogeny of a gorgoderid trematode recorded from the mussel Nodularia douglasiae in the Yodo River, Japan.

In 2009, a novel larval trematode of the family Gorgoderidae was found in the gonads of Nodularia douglasiae (Unionidae) from the lower reaches of the Yodo River, Osaka Prefecture, Japan. This is the first collection of trematodes in a unionid mussel in Japan. We investigated the morphology and life cycle of the trematode, and conducted a molecular phylogenetic analysis with other gorgoderid species, both those collected in the Yodo River water system and those reported in the literature. Immature adult worms were obtained from the ureters of the common carp Cyprinus carpio, the first known in

High prevalence of urinary schistosomiasis in a desert population: results from an exploratory study around the Ounianga lakes in Chad.

BACKGROUND: Researching a water-borne disease in the middle of the Sahara desert might not seem the most relevant concern. However, nomadic Sahelian pastoralists health concerns regarding their livestock and anecdotal reports about trematode infections of Fasciola spp. and Schistosoma spp. in desert-raised animals justified an exploratory study focusing on the lakes of Ounianga in Northern Chad. The aim was to test whether trematode parasites such as Schistosoma spp. occur in human populations living around the Sahara desert lakes of Ounianga Kebir and Ounianga Serir in northern Chad. METHODS…

The antigens of Paragonimus westermani, Schistosoma mansoni, and Fasciola hepatica adult worms. Evidence for the presence of cross-reactive antigens and for cross-protection to Schistosoma mansoni infection using antigens of Paragonimus westermani.

The presence of cross-reacting antigens between Paragonimus westermani, Schistosoma mansoni, and Fasciola hepatica adult worms was demonstrated by Ouchterlony immunodiffusion and enzyme-linked immunosorbent assay (ELISA). A serum bank was developed against the three trematode genera to serve as probes to determine the presence of cross-reacting antibodies to P. westermani worm extracts. In this manner, it was possible to demonstrate that antigens common to F. hepatica and S. mansoni tegument were also present in P. westermani worm extracts. Likewise, it was possible to demonstrate that the F.

Coinfection with Clonorchis sinensis modulates murine host response against Trichinella spiralis infection.

Concomitant infections of different species of parasites are common in the field. Infection with one parasite species likely triggers host responses that may influence the subsequent infection of another species and alter disease outcomes. So far, the majority of studies have focused on single species parasite infection, and the mechanisms of protection induced by the first parasite infection against the secondary infection remain poorly defined. In this study, we assess the impact of trematode Clonorchis sinensis infection on the course of another tissue nematode Trichinella spiralis challeng

North American paragonimiasis: epidemiology and diagnostic strategies.

Paragonimiasis is a zoonotic, food-borne trematode infection that affects around 23 million people in Asia, Africa and the Americas. North American paragonimiasis, caused by Paragonimus kellicotti, is a common infection of crustacean-feeding mammals in parts of the USA and Canada. Although infection rates in crayfish are very high in some areas, human infections are rare and depend on the consumption of raw or undercooked crayfish. Human infections can be easily prevented and treated, but proper diagnosis of paragonimiasis is a problem. Paragonimus lung flukes often cause serious disease sympt

Observations on the biology of Postharmostomum ntowi Hodasi, 1967 (Trematoda: Brachylaimidae) based on intermediate and definitive hosts found in Nigeria.

Following the recovery of the metacercariae of a brachylaimid trematode from the rectum of the frog Amnirana galamensis from Ase in Delta State, Nigeria, we investigated the land snails in the locality to establish their roles in the life cycle of the parasite. Of the four land snails investigated from Ase (Limicolaria aurora, Archachatina marginata, A. papyracea, and Thapsia oscitans), and a Limicolaria sp. from Tombia (Bayelsa State), four harboured larval stages of the bracylaimid. Only L. aurora and the Limicolaria sp. harboured cercariogenous sporocysts and are therefore presumed to serve

Broader Geographic Sampling Increases Extent of Intermediate Host Specificity for a Trematode Parasite (Notocotylidae: Quinqueserialis quinqueserialis).

Knowledge of helminth life cycles is essential to understanding their host specificity, geographic distribution, and transmission. Many helminth life cycle descriptions are based on field collections in a limited part of the parasite's range. However, it is important to determine whether helminth life cycles and host specificity remain consistent across their geographic range so that we may better understand their life history and transmission ecology. Here, we investigated whether the life cycle of a widespread trematode, Quinqueserialis quinqueserialis (Notocotylidae) varies across its geogr

Outbreak of Philophthalmus gralli in four greater rheas (Rhea americana).

Using slit-lamp biomicroscopy, conjunctival biopsy, and morphological identification, a flock of four Greater rheas (Rhea americana) in Arizona were diagnosed with conjunctivitis secondary to Philophthalmus gralli (P. gralli) infection. Aquatic snails from the exhibit's water source were identified as Melanoides tuberculatus, a known vector for P. gralli. Comparison of partial sequences of DNA regions from P. gralli adults removed from the rheas and metacercariae from the aquatic snails demonstrated a 100% match, confirming the source of infection. The flock was divided into two treatment grou

Trematodes recovered in the small intestine of stray cats in the Republic of Korea.

In 2005, we reported the infection status of 438 stray cats with various species of intestinal helminths, including nematodes (4 species), trematodes (23 species), and cestodes (5 species) in the Republic of Korea. However, morphologic details of each helminth species have not been provided. In the present study, we intended to describe morphologic details of 13 trematode species which were either new fauna of cats (10 species) or new fauna of not only cats but also all animal hosts (3 species). The worms were fixed in 10% neutral buffered formalin under a cover slip pressure, stained with Sem

Genetic identification of Fasciola hepatica by ITS-2 sequence of nuclear ribosomal DNA in Turkey.

The trematodes of the genus Fasciola are the common liver flukes of a range species of animals and have a global geographical distribution. They can generally be distinguished on the basic of their morphology. ITS-2 ribosomal DNA sequences have been used to characterize the liver flukes as a specific marker from different geographical regions which include F. hepatica, F. gigantica, and an intermediate Fasciola. To determine the phylogenetic location of F. hepatica of Turkey origin based on ITS-2 rDNA molecular data, adult F. hepatica trematodes were collected from the liver naturally infected

First report of Euryhelmis parasites (Trematoda, Heterophyidae) in Africa: conservation implications for endemic amphibians.

In this study, we report, through molecular identification, the first African records of a digenean trematode parasite of the genus Euryhelmis. We recovered metacercariae encysted in an anuran, the endemic Moroccan painted frog (Discoglossus scovazzi), and a vulnerable caudate, the North African fire salamander (Salamandra algira), from four localities in North Africa (Morocco). Our records go back to the past century and have been confirmed in successive fieldwork seasons thereafter. Metacercarial stages of these parasites require amphibians as the last intermediate host, but the exact identi

Site-level variation and molecular identification of dual Microphallus infections in shore crabs (Carcinus maenas).

Digenean trematodes commonly use marine crustaceans as intermediate hosts, yet their distribution and host-specific dynamics remain underexplored in temperate European systems. In this study, metacercarial stages of Microphallus similis and M. primas were identified in the hepatopancreas of Carcinus maenas collected from two distinct locations in South Wales, UK across a 4-month period. M. similis exhibited consistently high prevalence (~ 76%) at both sites, while M. primas occurred only as a co-infection, with lower prevalence (2-9%) and infection intensity. At Oxwich Bay, crab size and color

Fascioloides magna infection in a captive impala (Aepyceros melampus) in Florida, USA.

Parasitic diseases are associated with diverse clinical signs, and veterinary professionals must be familiar with the presentations of infections in a variety of hosts. As human activity introduces new and exotic host species, expanding our understanding of pathogen effects in new species becomes integral to effective surveillance, diagnosis, and treatment. This article presents the expansion of the known host range of the American liver fluke, Fascioloides magna, with the infection of a farmed impala (Aepyceros melampus) in Marion County, Florida, USA.
