GSDME-Mediated Pyroptosis and the mtDNA-cGAS-STING Pathway Drive Deoxynivalenol-Induced Porcine Intestinal Inflammatory Injury.

Deoxynivalenol (DON) is a prevalent mycotoxin that compromises intestinal barrier integrity, yet the mechanisms underlying DON-induced chronic inflammation remain unclear. This study investigated these mechanisms using a 28-day piglet model and porcine intestinal epithelial cells (IPEC-J2). In vivo, DON exposure caused intestinal morphological damage and inflammation, accompanied by caspase-3/Gasdermin E (GSDME)-mediated pyroptosis. In vitro experiments confirmed that DON triggers pyroptosis via the caspase-3/GSDME pathway. Mechanistically, activated GSDME permeabilized mitochondrial membranes

Selenium nanoparticle-modified probiotics attenuate deoxynivalenol-induced intestinal injury in chickens via glutathione-mediated metabolic pathway.

Deoxynivalenol (DON), one of the most prevalent mycotoxins, primarily targets the intestine causing oxidative stress and inflammatory storm. Meanwhile, Selenium nanoparticles (SeNPs) have been demonstrated to possess multiple physiological functions, including anti-inflammatory and antioxidant activities. We successfully synthesized biogenic SeNPs with zero-valent and 142 ± 23.5 nm particle size using the intrinsic oxidoreductase enzymes and metabolic machinery of Lactobacillus rhamnosus CICC 20257 (Lr) to efficiently reduce sodium selenite through a biological process. This study intends to i

Co-occurrence of Mycotoxin-Induced Hepatotoxicity in Mice Inhibited by Lycopene: Mitochondrial Impairment and Early Hepatic Fibrosis.

SCOPE: Mycotoxins co-contamination of agricultural products poses a serious threat to human and animal health, especially hepatic dysfunction. Zearalenone (ZEN), deoxynivalenol (DON), and aflatoxin B1 (AFB1) are three commonly co-occurring mycotoxins. This study is to determine whether lycopene (LYC) can alleviate hepatic toxicity induced by the co-occurrence of ZEN, DON, and AFB1 in mice. METHODS AND RESULTS: Eighty 6-week-old male ICR mice are divided into four groups: CON group (solvent control), LYC group (10 mg kg-1 LYC), Co-M group (10 mg kg-1 ZEN + 1 mg kg-1 DON + 0.5 mg kg-1 AFB1), an

Protective effects of mannan/β-glucans from yeast cell wall on the deoxyniyalenol-induced oxidative stress and autophagy in IPEC-J2 cells.

The aim of this study was to investigate the effects of biomacromolecules mannan/β-glucans from yeast cell wall (BYCW) to alleviate Deoxynivalenol(DON)-induced injury. Considering that DON has strong oxidizing effect and stimulates autophagy and apoptosis, we examined the effects of BYCW on consequent oxidative stress damage indicators, cells autophagy and apoptosis induced by DON using the porcine jejunum epithelial cell lines (IPEC-J2) as a cell culture model. The results showed that application of BYCW could reverse the decrease of cell viability by DON significantly, and suppress the level

The mycotoxin deoxynivalenol facilitates allergic sensitization to whey in mice.

Intestinal epithelial stress or damage may contribute to allergic sensitization against certain food antigens. Hence, the present study investigated whether impairment of intestinal barrier integrity by the mycotoxin deoxynivalenol (DON) contributes to the development of whey-induced food allergy in a murine model. C3H/HeOuJ mice, orally exposed to DON plus whey once a week for 5 consecutive weeks, showed whey-specific IgG1 and IgE in serum and an acute allergic skin response upon intradermal whey challenge, although early initiating mechanisms of sensitization in the intestine appeared to be

The PGC-1α/SIRT3 pathway mediates the effect of DON on mitochondrial autophagy and liver injury in mice.

Deoxynivalenol (DON)-induced liver injury is closely associated with mitochondrial dysfunction, yet it remains unclear whether this injury is mediated by mitochondrial autophagy via the PGC-1α/SIRT3 pathway. This study aimed to ascertain whether DON triggers mitochondrial autophagy, thereby causing liver injury in mice through the PGC-1α/SIRT3 pathway. Mice were orally administered DON at doses of 1.2 and 2.4 mg/kg once daily for 28 consecutive days. The results indicated that DON treatment significantly elevated the activity levels of two key liver enzymes and increased oxidative stress in th

Glucuronolactone improves lung injury caused by PRRSV and DON co-challenge by enhancing the Nrf2-mediated antioxidant capacity in weaning piglets.

In the pig industry, both deoxynivalenol (DON) challenge and porcine reproductive and respiratory syndrome virus (PRRSV) infection can negatively impact the growth performance of pigs, thereby impairing the healthy development of the industry. Glucuronolactone (GLU) has been shown to be a potent antioxidant that helps alleviate oxidative stress. Therefore, this study aimed to investigate the effect of GLU on oxidative stress and lung injury induced by co-challenge with PRRSV and DON. Eighteen weaned piglets were randomly divided into three groups: control (CON), DON, and DON + GLU. After DON a

Cellular responses to T-2 toxin and/or deoxynivalenol that induce cartilage damage are not specific to chondrocytes.

The relationship between T-2 toxin and deoxynivalenol (DON) and the risk of Kashin-Beck disease is still controversial since it is poorly known about their selectivity in cartilage damage. We aimed to compare the cytotoxicity of T-2 toxin and DON on cell lines representative of cell types encountered in vivo, including human chondrocytes (C28/I2), human hepatic epithelial cells (L-02) and human tubular epithelial cells (HK-2). In addition, we determined the distribution of T-2 toxin and DON in Sprague-Dawley (SD) rats after a single dose exposure. T-2 toxin or DON decreased proliferation in a

The Reproductive Toxicity Valuation of Deoxynivalenol: An Integrated Study from Network Toxicology, Molecular Docking, Molecular Dynamics Simulation and Single-Cell RNA Sequencing.

Deoxynivalenol (DON), a Fusarium-derived mycotoxin widely found in grain-based feed, has become a major global environmental contaminant. Reproductive toxicity is one of its most important toxic effects, yet systematic investigations covering both male and female reproductive injury remain limited. This study aimed to establish a combined strategy of network toxicology, molecular docking, molecular dynamics simulation, and single-cell RNA sequencing to evaluate the reproductive toxicity of DON. AKT1, EGFR, PIK3CA, PIK3R1, and SRC were identified as key targets involved in DON-induced reproduct

Effects of Deoxynivalenol and Fumonisins on Broiler Gut Cytoprotective Capacity.

Mycotoxins are a crucial problem for poultry production worldwide. Two of the most frequently found mycotoxins in feedstuffs are deoxynivalenol (DON) and fumonisins (FUM) which adversely affect gut health and poultry performance. The current knowledge on DON and FUM effects on broiler responses relevant for gut detoxification, antioxidant capacity, and health is still unclear. The aim of this study was to assess a range of selected molecular intestinal biomarkers for their responsiveness to the maximum allowable European Union dietary levels for DON (5 mg/kg) and FUM (20 mg/kg) in broilers. Effects of vomitoxin (deoxynivalenol) on transcription factor NF-kappa B/Rel binding activity in murine EL-4 thymoma and primary CD4+ T cells.

The trichothecene mycotoxin vomitoxin (VT, deoxynivalenol) superinduces the gene expression of IL-2 and several other cytokines in both cellular and murine models. Because transcription factor NF-kappaB/Rel has been shown to play a crucial role in control of cytokine gene transcription, we assessed the in vitro effects of VT on NF-kappaB/Rel binding activity by electrophoretic mobility shift assay using both cloned (EL-4) and primary (CD4+) murine T cells. When EL-4 thymoma cells were stimulated with phorbol 12-myristate 13-acetate plus ionomycin in the presence of 500 ng/ml VT, DNA binding ac

The acute toxicopathy of intravenous diacetoxyscirpenol (anguidine) administration in swine.

Diacetoxyscirpenol (DAS, anguidine) was given intravenously to swine at 0.0, 0.5, and 1.0 mg/kg body wt. In mitotically and metabolically active tissues such as gastrointestinal epithelium and lymphoid aggregates the effects of DAS mimicked radiation poisoning. A quadratic dose-response relationship between the cytotoxicity of DAS and damage to enterocytes was found. Enterocytes in different anatomical regions of the bowel had differing susceptibilities to the toxic effects of DAS. In lymphoid tissues, DAS was preferentially cytotoxic to B-lymphocyte-rich tissues as compared to T-lymphocyte-ri

Medical aspects of bio-terrorism.

INTRODUCTION: Bioterrorism is a terrorist action involving the intentional release or dissemination of a biological warfare agent (BWA), which includes some bacteria, viruses, rickettsiae, fungi or biological toxins. BWA is a naturally occurring or human-modified form that may kill or incapacitate humans, animals or plants as an act of war or terrorism. BWA is a weapon of choice for mass destruction and terrorism, because of the incubation period, less effective amount than chemical warfare agents, easily distribution, odorless, colorless, difficult to detect, no need of specialized equipment

MeJA inhibits fungal growth and DON toxin production by interfering with the cAMP-PKA signaling pathway in the wheat scab fungus Fusarium graminearum.

Deoxynivalenol (DON), a mycotoxin primarily produced by Fusarium species, is commonly found in cereal grains and poses risks to human and animal health, as well as global grain trade. This study demonstrates that methyl jasmonate (MeJA), a natural plant hormone, inhibits the growth and conidiation of Fusarium graminearum. Importantly, MeJA significantly reduces DON production by suppressing TRI gene expression and toxisome formation. To explore the molecular mechanism, we identified MeJA-tolerant mutants, including a transcription factor MRT1 and cAMP-PKA pathway-related genes (FgGPA1 and FgSN

Silanized Luminescent Quantum Dots for the Simultaneous Multicolor Lateral Flow Immunoassay of Two Mycotoxins.

A critical point for the successful development of a fluorescent quantum dot (QD)-based immunoassay is maintaining the high fluorescence quantum yield of QDs during hydrophilization and bioconjugation. In this paper, we carefully designed CdSe/CdS and CdSe/CdS/ZnS core-shell heterostructures and extended them with silica coating of different surface composition allowing preservation of fluorescence quantum yield as high as 70% in aqueous media. The silanized QDs containing epoxy and carboxy surface groups were bioconjugated with monoclonal antibodies. The synthesized fluorescent conjugates wer

Gene expression profiles and molecular mechanism of cultured human chondrocytes' exposure to T-2 toxin and deoxynivalenol.

T-2 toxin and deoxynivalenol (DON) are secondary metabolites produced by Fusarium fungi and are commonly found on food and feed. Although T-2 toxin and DON have been suggested as the etiology of Kashin-Beck disease (KBD), an endemic osteochondropathy, little is known about the mechanism when human chondrocytes are exposed to T-2 toxin and DON. The purpose of this study is to identify the gene expression differences and underlying molecular changes modulated by T-2 toxin and DON in vitro in human chondrocytes. After the experiments of cell viability, the gene expression profiles were analyzed i

[Mycotoxin research in humans].

This study investigates the occurrence of aflatoxins in Ecuador. Early investigators proved the presence of aflatoxins in human and animal food, but the disturbing data lead to the formation of two research teams at Guayaquil University and the Agrarian University of Ecuador to investigate aflaxotins and other mycotoxins in food and their relationship to human health. Because the concept of mycotoxicosis as a result of the secondary metabolites produced by different species of moulds could cause different clinical patterns, the research team includes Aspergillus metabolites found in the urine

Eucommia ulmoides flavonoids alleviate intestinal oxidative stress damage in weaned piglets by regulating the Nrf2/Keap1 signaling pathway.

This study examined how Eucommia ulmoides flavonoids (EUF) protect against intestinal oxidative stress induced by deoxynivalenol (DON) in weaned piglets. Forty weaned piglets were randomly assigned to four dietary groups for a period of 14 days. The piglets were fed a control diet (Control) or the Control diet supplemented with 100 mg EUF/kg (EUF group), 4 mg DON/kg diet (DON group) or both (EUF+DON group) in a 2×2 factorial design. DON-challenged piglets on the EUF-supplemented diet showed significant improvements in growth performance. They also had notably lower serum levels of alkaline pho
