N-glycolylneuraminic acid and N-acetylneuraminic acid define feline blood group A and B antigens.

Blood group incompatibility causes transfusion reactions and neonatal isoerythrolysis in cats. We investigated the molecular nature of the blood group antigens from cats that had blood type A, B, and AB erythrocytes. Naturally occurring anti-type B antibodies, Triticum vulgaris lectin, monoclonal antibody (MoAb) 32-27, and MoAb R-24 were used in agglutination tests, Western blots, and thin-layer chromatography (TLC) enzyme immunostaining. Type A erythrocytes had NeuGc-NeuGc-Galactose-Glucose-Ceramide ([NeuGc]2GD3) where NeuGc represents N-glycolylneuraminic acid, and NeuAc-NeuGc-GD3, where Neu…

Changes in Phospholipid Composition of a Winter Wheat Cultivar during Germination at 2 C and 24 C.

Evaluation of various solvent systems for lipid extraction of wheat Triticum aestivum L. cv. Rideau seeds showed that boiling 2-propanol followed by the Bligh-Dyer procedure was the most efficient method, with respect to lipid yield and ability to inactivate lipolytic enzymes. Ten phospholipids were identified in dry seeds; the major components being phosphatidylcholine, lysophosphatidylcholine, N-acyl lysophosphatidyl-ethanolamine, N-acylphosphatidylethanolamine, and phosphatidylethanolamine. After growth for 1 week (2 C) or 31 hours (24 C), the proportions of phosphatidylethanolamine + lysop…

Novel but stable endosymbionts have contrasting effects on aphid dispersal and plant feeding damage in the cereal pest Diuraphis noxia.

Endosymbiotic bacteria can affect many ecological attributes of their insect hosts, including (in herbivorous insects) how they interact with plants where they feed. This raises the issue of whether deliberate endosymbiont introductions could be used to decrease crop damage caused by insect pests. Here, we investigate how transinfecting Rickettsiella viridis and Regiella insecticola endosymbionts into a novel pest aphid host, the Russian wheat aphid (Diuraphis noxia), influences population growth, alate production, dispersal ability, and crop damage. Both the Rickettsiella (originating from pe

Association of dumpling wrapper quality with property of starch of three wheat varieties isolated from different flour millstreams.

The objective of this study was to investigate quality of dumpling wrappers (DWs) modulated by starch of three wheat varieties isolated from different flour millstreams. Starch from break section of the milling system (BS) had more small B-type granules and lower relative crystallinity (RC) and swelling power (SP) than that from reduction section (RS). This improved starch-gluten interactions and enhanced stability and elasticity of interior network in DWs containing BS. It could be indicated by higher elastic modulus and zero shear viscosity and lower maximum creep compliance. Thus, DWs conta

Elevated CO2 promotes faster wheat starch digestion through coordinated changes in starch and protein.

Rising atmospheric CO2 affects the nutritional functionality of wheat grains, but how coordinated changes in starch and protein under elevated CO2 (eCO2) contribute to starch digestibility remains unclear. Winter wheat was grown under ambient (415 ppm) and eCO2 (600 ppm) to investigate eCO2 effects on grain quality traits and in vitro starch digestion kinetics. Results indicated that eCO2 generally increased amylose content, the proportion of B-type starch granules, the enthalpy of gelatinization and the relative crystallinity of wheat grain starch, while decreasing R1047/1022 and the proporti

Exogenous porcine blood polypeptide alleviates drought damage in wheat.

Previous studies have demonstrated that porcine blood polypeptide (PBP) could mitigate plant damage under adverse conditions. This study was conducted to systematically investigate the possible role of PBP in enhancing drought resistance in wheat through comprehensive physiological and biochemical analyses, as well as transcriptomic analysis. Morphological observations revealed that PBP-primed seedlings exhibited improved growth, biomass accumulation and root system under drought stress. Physiological and biochemical analyses demonstrated that primed seedlings existed significantly higher valu

Field trial analyses of wheat and cassava benefit from spatial correction.

Spatial variation is a major source of error in agricultural field experiments affecting genotype performance prediction. Implementing statistical models that account for spatial effects can improve the prediction of genotype performance. This study evaluated the impact of the P-spline spatial correction method on the estimation of genetic parameters and AIC values in two distinct crops, wheat and cassava, using four models: Block, Block + Spatial, Block + Marker, and Block + Marker + Spatial. Analyses were performed on data from 115 and 68 trials obtained from the T3/WheatCAP and Cassavabase

Quantum Light Imaging of the Plant Root─Rhizobacteria─System Using iLOV Fluorescence.

It has been demonstrated recently that the use of specific quantum states of light for two-photon microscopy can lead to a substantial reduction of the intensity of light needed for the imaging of biological objects. This opens new ways to better understand the delicate mechanisms of the interactions of living organisms in the rhizosphere on an extended time scale without optical damage or alteration of their native behavior. Herein, we report a successful realization of entangled two-photon excited fluorescence microscopy of wheat plant roots and a set of biological samples, including purifie

High-titer microbial lipid fermentation and extractive recovery by cell wall-weakened Trichosporon cutaneum strain and wheat straw sugar syrup feedstock.

Lignocellulosic biomass is a promising feedstock for sustainable microbial lipid production; however, its utilization is hindered by low fermentation performance and inefficient lipid recovery. In this study, an integrated strategy combining enzyme-mediated adaptive evolution combined with centrifugal fractionation was employed to generate the oleaginous yeast strain Trichosporon cutaneum YY52E, exhibiting enhanced lipid accumulation and reduced cell wall integrity. The lipid titer reached 60.1 g/L in fed-batch fermentation, with a yield of 0.3 g/g sugars, using wheat straw sugar syrup prepare

Optimized sowing rate and fertilization regulate photosynthetic characteristics, grain yield and quality of winter wheat.

The Loess Plateau is a principal dryland winter wheat production region in China, where unreasonable sowing rate and fertilization management limit yield and quality. A two-factor split-plot experiment was conducted during 2019-2021 with three sowing rates (150 kg ha-1, 180 kg ha-1, 210 kg ha-1) and three fertilization type (CK, OPT, OPT-N). Increasing sowing rate to 210 kg ha-1 significantly improved population tiller, secondary root number, and canopy structure. OPT reduced N input by > 50% but maintained plant growth and delayed leaf senescence. The 180 kg ha-1 rate enhanced canopy light in

Harnessing genomics approaches for heat stress resilience in wheat: from discovery to deployment.

Global warming poses a critical threat to wheat (Triticum aestivum L.) production, particularly during sensitive reproductive stages. This review synthesizes current understanding of how heat stress affects wheat and the adaptive mechanisms that confer tolerance, with emphasis on recent advances in genomics and biotechnology. Heat stress impairs morphological, physiological, biochemical, and molecular processes, reducing photosynthetic efficiency, accelerating senescence, disrupting assimilate partitioning, and damaging cellular structures. Adaptive responses include optimized water relations,…

Physiological characterization of Cochliobolus bicolor (sin. Bipolaris bicolor) from wheat BRS 264 seeds.

Several diseases cause a reduction on the wheat (Triticum aestivum L.) yield, among which is helminthosporiosis, which infects the plant in all its stages of development. The objective of this work was to carry out the physiological characterization, under different temperature regimes of Cochliobolus bicolor isolates from BRS 264 wheat seeds. The six isolates of C. bicolor were recovered from the mycoteca stored in the Plant Pathology Laboratory of the Goiás State University. For the physiological evaluation, C. bicolor isolates were inoculated in Petri dishes containing PDA medium and subjec…

Microbial co-occurrence network and spatial metabolism underlie wheat cultivar effects on medium-high temperature Daqu fermentation.

Although wheat cultivar is widely accepted to shape Daqu quality, it remains unclear which cultivar-linked microbial property - taxonomic composition, spatial niche differentiation, or the topology of cross-domain interactions - is the proximate driver of flavor-precursor accumulation. To address this gap, four wheat cultivars (GS19, QM725, QM838 and ZM) were used as natural perturbations, and depth-resolved (core/surface) 16S/ITS profiling, untargeted metabolomics and volatilomics were combined across the 90-day fermentation-maturation trajectory of medium-high-temperature Daqu. Of the four c

Construction and expression of a transglucosidase-hexose fusion enzyme for oxidating starch: The intrinsic correlation between molecular structure and oxidation degree.

Genetic engineering was utilized to fuse transglucosidase (TG) and hexose oxidase (HOX), producing a fusion enzyme TGHOX. This novel enzyme eenabled a sequence reaction involving starch chain transformation and oxidation in a single system. Compared with native wheat starch (24.07% amylose), TGHOX treatment increased the amylose content of wheat starch, with the TGHOX-16 treatment reaching a maximum amylose content of 41.77%. The A-chain content of wheat starch increased from 31.31% to 41.68% after TGHOX treatment. Higher A-chain content provided more highly accessible non-reducing end substra

Empowering Gliadin Detection: A Visible-Code Semiquantitative Lateral Flow System for Rapid and Reliable Results.

Wheat is a global staple, but its main protein, "gluten," can trigger severe reactions in people with celiac disease, an autoimmune disorder affecting around 1% of the population. The only treatment is a lifelong gluten-free diet. Gliadin, a gluten component, is the primary trigger and is difficult to detect due to its low solubility. This makes it challenging for celiac patients to safely eat out or to enjoy diverse foods. To address this, we developed LEO (Lateral flow Enhanced by Optical imaging), a portable gluten detection system combining lateral flow assays with smartphone-based image a

Stripe Rust Virulence and Host Reaction Assessment Protocols: From Lab to Field.

Puccinia striiformis, the causal agent of stripe rust disease, poses a significant threat to cereal crops worldwide. Since 2000, the emergence of heat-adapted races/lineages of the wheat-infecting form P. striiformis f. sp. tritici (Pst) has enabled the disease to spread beyond its traditional high-elevation, cool, and moist regions. It can now occur wherever wheat is cultivated, making it a pandemic concern. This chapter provides an overview of the methodologies and practices established in our lab for working with this pathogen. These methodologies include isolate collection, recovery, purif

Novel technique to enhance phosphorus availability in sewage sludge biochar using wood ash extract to produce an effective circular economy product.

Sewage sludge (or biosolids) produced during wastewater treatment contain significant amounts of phosphorus. Pyrolysis treatment can produce a safe material while sequestering carbon; however, it underutilises the phosphorus in the resulting biochar, leaving it partially inaccessible to plants. Here the effect of doping calcium-rich sewage sludge (14 % calcium) with potassium acetate, combustion wood ash and its water-extract on phosphorus availability, and carbon and nitrogen release during pyrolysis are investigated at 500 and 700 °C. The aim was to produce an effective circular economy mate

Lunar magnetism impairs wheat seedling photosynthesis: A simulated environment study.

Plants evolved under Earth's stable geomagnetic field (GMF), a condition sharply contrasting with the near-absence of a global magnetic field on the Moon. However, the effects of this stark magnetic disparity on fundamental plant processes like photosynthesis remain underexplored, particularly in the context of future lunar agriculture. This study rigorously investigated the physiological and biochemical mechanisms underpinning the photosynthetic response of wheat seedlings - a staple crop selected for its centrality in closed-loop life support - to a simulated lunar weak magnetic field (WMF,

Assessing ozone pollution and climate change impacts on winter wheat: flux modeling vs. dose-response modeling.

Surface ozone is a phytotoxic pollutant that damages photosynthetic systems, reduces gas exchange, retards vegetation growth, and decreases yield. In this study, we developed a new ozone flux module within an agroecosystem model framework to enhance our ability to understand, measure, and predict the impact of surface ozone on agricultural productivity. The new module was calibrated and evaluated against historical observational data from multiple sites. It was then applied to predict winter wheat yield and gross primary productivity (GPP) in response to future ozone changes under different cl
