An inactivated yellow fever 17DD vaccine cultivated in Vero cell cultures.

Yellow fever is an acute infectious disease caused by prototype virus of the genus Flavivirus. It is endemic in Africa and South America where it represents a serious public health problem causing epidemics of hemorrhagic fever with mortality rates ranging from 20% to 50%. There is no available antiviral therapy and vaccination is the primary method of disease control. Although the attenuated vaccines for yellow fever show safety and efficacy it became necessary to develop a new yellow fever vaccine due to the occurrence of rare serious adverse events, which include visceral and neurotropic di

The efficacy of inactivated West Nile vaccine (WN-VAX) in mice and monkeys.

BACKGROUND: West Nile virus (WNV) belonging to the genus Flavivirus of the family Flaviviridae causes nervous system disorder in humans, horses and birds. Licensed WNV vaccines are available for use in horses but not for humans. We previously developed an inactivated West Nile virus vaccine (WN-VAX) using a seed virus from West Nile virus (WNV NY99) that was originally isolated in New York City in 1999. In this study, we report the immunogenicity of WN-VAX in both mice and non-human primates. FINDINGS: The WN-VAX immunized mice showed protection against lethal infection with WNV NY99. The cha

Inactivated polio vaccination using a microneedle patch is immunogenic in the rhesus macaque.

The phased replacement of oral polio vaccine (OPV) with inactivated polio vaccine (IPV) is expected to significantly complicate mass vaccination campaigns, which are an important component of the global polio eradication endgame strategy. To simplify mass vaccination with IPV, we developed microneedle patches that are easy to administer, have a small package size, generate no sharps waste and are inexpensive to manufacture. When administered to rhesus macaques, neutralizing antibody titers were equivalent among monkeys vaccinated using microneedle patches and conventional intramuscular injecti

Clinical and immune responses to inactivated influenza A(H1N1)pdm09 vaccine in children.

BACKGROUND: As the influenza A H1N1 pandemic emerged in 2009, children were found to experience high morbidity and mortality and were prioritized for vaccination. This multicenter, randomized, double-blind, age-stratified trial assessed the safety and immunogenicity of inactivated influenza A(H1N1)pdm09 vaccine in healthy children aged 6 months to 17 years. METHODS: Children received 2 doses of approximately 15 or 30 µg hemagglutin antigen 21 days apart. Reactogenicity was assessed for 8 days after each dose, adverse events through day 42, and serious adverse events or new-onset chronic illne

Trivalent inactivated influenza vaccine is not associated with sickle cell crises in children.

BACKGROUND AND OBJECTIVES: Children with sickle cell disease are considered at high risk for complications from influenza infection and are recommended to receive annual influenza vaccination. However, data on the safety of influenza vaccination in children with sickle cell anemia are sparse. METHODS: Using a retrospective cohort of children aged 6 months to 17 years in 8 managed care organizations that comprise the Vaccine Safety Datalink and who had a diagnosis of sickle cell anemia from 1999 to 2006, we conducted matched case-control and self-controlled case series studies to examine the a

Field evaluation of an inactivated vaccine to control raccoon rabies in Ontario, Canada.

Since raccoon rabies first appeared in Ontario in 1999, >90,000 raccoons (Procyon lotor) have received IMRAB3 inactivated rabies vaccine via intramuscular (IM) injection and were released at the point of capture as part of a multiyear rabies control program, trap-vaccinate-release (TVR). Of the 132 confirmed cases infected with raccoon-variant rabies virus in Ontario between 1999 and 2005, two were from that vaccinated group, as indicated by the presence of identifying ear tags. During ongoing rabies control programs in 2003, sera were collected from 172 wild raccoons that had received IMRAB3

Live attenuated versus inactivated influenza vaccine in infants and young children.

BACKGROUND: Universal vaccination of children 6 to 59 months of age with trivalent inactivated influenza vaccine has recently been recommended by U.S. advisory bodies. To evaluate alternative vaccine approaches, we compared the safety and efficacy of intranasally administered live attenuated influenza vaccine with those of inactivated vaccine in infants and young children. METHODS: Children 6 to 59 months of age, without a recent episode of wheezing illness or severe asthma, were randomly assigned in a 1:1 ratio to receive either cold-adapted trivalent live attenuated influenza vaccine (a ref

Randomized trial of inactivated and live polio vaccine schedules in Guatemalan infants.

BACKGROUND: The immunogenicity of inactivated poliovirus vaccine (IPV) in developing countries is not well documented. This study compared the immune response to IPV with that to oral poliovirus vaccine (OPV) in Guatemalan infants. METHODS: This was an open-label, randomized comparison of IPV only, OPV only, or IPV followed by OPV in Guatemalan public health clinics. Serum samples were tested for neutralizing antibodies, and stool samples were tested for Sabin strain polioviruses. RESULTS: Seropositivity rates 2 months after 2 doses of IPV were 98%-100% for polio types 1, 2, and 3 and were 9

Age- and dose-interval-dependent antibody responses to inactivated poliovirus vaccine.

Antibody responses were studied in five groups of children immunized with different three-dose schedules of inactivated poliovirus vaccine (IPV). The age of the child at the first dose (1-4 months) and the interval between the first and second doses (2-4 months) influenced the initial responses in a serotype-dependent manner. All the groups attained sufficient antibody level after three doses but the third dose given at 18 months resulted in higher persisting antibody levels than that given at 12 months. The highest persisting antibody titers against PV1 and PV2 (but not against PV3) at the ag

Immunostimulatory effects of polar glycopeptidolipids of Mycobacterium chelonae for inactivated rabies vaccine.

Humoral and cellular immune responses were analyzed with Fuenzalida-Palacios rabies vaccine associated with pGPL-Mc, polar glycopeptidolipids extracted from Mycobacterium chelonae, aiming at its use as adjuvant. These results were compared to those obtained with BCG, a well-known immunostimulator, under the same conditions. Rabies vaccine plus pGPL-Mc (2.5 mg/kg) induced a significant increase in serum neutralizing activity, in vitro lymphocyte proliferation (spontaneous, specific and mitogen stimulation) and delayed type hypersensibility. In addition, pGPL-Mc, as well as BCG, enhanced the vac

[The evaluation of the reactogenicity and immunogenic activity of a new concentrated inactivated leptospirosis vaccine].

In the controlled field trial the reactogenicity, safety and antigenic activity of a new concentrated inactivated leptospirosis vaccine after its administration in one and two injections of 0.5 ml were studied in comparison with those of the existing commercial vaccine, introduced in two injections in doses of 2.0 and 2.5 ml. The new experimental vaccine exhibited low reactogenicity and was found to be safe and highly immunogenic when introduced in a single injection of 0.5 ml. As shown in this trial, the immunogenic characteristics of immunization made in a single injection were not inferior

Immunogenicity and safety of a new inactivated hepatitis A vaccine in Thai young adults.

In view of the increasing median age of hepatitis A virus (HAV) infection observed recently in Asia, and the resulting increased number of symptomatic cases occurring in adults, with the concomitant risk of outbreaks, immunization against this agent on a national scale might be considered. An open clinical trial was conducted in Thai adolescents and young adults in order to establish the immunogenicity and safety of a new inactivated hepatitis A vaccine. At 24-week intervals, two doses (primary dose and booster) of the hepatitis A vaccine (160 antigenic units per dose) were administered to 80

Developed countries should not use inactivated polio vaccine for the prevention of poliomyelitis.

Taking into account the global status of polio, it seems evident that the continuing use of oral poliovaccine in all countries is the most obvious and prudent public health policy for the foreseeable future. Possible exceptions might include those countries which are not troubled by the added cost of the inactivated vaccine; whose health services are able to guarantee high levels of vaccine coverage; and which can expect to experience comparatively few importations of wild poliovirus. An important question is whether it is warranted at this time to recommend a combined schedule of inactivated

Severe systemic reactions attributed to the acetone-inactivated parenteral typhoid vaccine.

Typhoid vaccine is recommended for people traveling to areas where they may be exposed to contaminated food and/or water. Two different parenteral forms of the vaccine have been utilized. We report five active duty patients with severe systemic reactions to the acetone-inactivated form of typhoid vaccine. The patients presented with very similar physical exam findings and laboratory abnormalities. Alternative typhoid vaccines that are less reactogenic are now available for use.

Evaluation of the potency and safety of inactivated Japanese encephalitis vaccine in US inhabitants.

In 1981 and 1982, two US citizens died from Japanese encephalitis (JE) acquired in China. In 1983, the Centers for Disease Control initiated an evaluation of a purified, inactivated, mouse-brain-derived JE vaccine produced and used in Japan since 1966. Two doses of this vaccine given 1-2 weeks apart evoked neutralizing antibody titers greater than or equal to 8 in only 77% of recipients. After three JE vaccine doses administered 1-2 weeks apart, 99% developed titers greater than or equal to 8. When a third dose was given to 29 participants 6-12 months after the primary series, all developed ti

[Immunogenicity and stability of a Soviet inactivated cultured vaccine against Japanese encephalitis].

Six immunologically active vaccine batches inducing a specific antibody to Japanese encephalitis (JE) virus were obtained in serial manufacture of the preparation. In HI tests, the minimal antibody titre was 1:80, the maximal 1:320, neutralization index 1g was 3.7 to 5.2. The data on the stability of the antigenic potency of the vaccines in relation to the duration of storage at 4 degrees-6 degrees C are presented (the follow-up period 3 years). A certain relationship was found between the antigenic potency of the preparation and the titre of the initial infectious tissue culture virus. Also,
