Biosponges Embedded with GDNF Enhance Neuromuscular Recovery Following Volumetric Muscle Loss.

Volumetric muscle loss (VML) injuries result in an irrecoverable loss of muscle mass and function. VML injury causes loss of both contractile tissue and associated neuromuscular junctions (NMJs). Biosponge (BSG) scaffolds, composed of gelatin, collagen, and laminin-111, have improved recovery following VML. However, improvements in NMJ quantity were not observed. Glial cell line-derived neurotrophic factor (GDNF) is known to promote motor unit survival and stimulate neurite outgrowth. In this work, BSG scaffolds were electrostatically coupled with GDNF via gelatin nanoparticles (GNPs) to suppo

An R83W mutation in Rab3A causes autosomal-dominant cerebellar ataxia.

Spinocerebellar ataxias (SCAs) are a group of progressive neurodegenerative disorders caused by pathogenic variants in more than 40 genes with diverse cellular functions. In this study, we identified the c.247C > T p.(Arg83Trp) variant in RAB3A, encoding a small GTPase involved in membrane-associated regulated exocytosis, in two families with cerebellar ataxia. Affected individuals presented with adult-onset, gradually progressive cerebellar symptoms, often accompanied by mild gait spasticity and tremors. Variable features of neurodevelopmental disorders were also observed. Brain MRI consisten

Structural Destabilization of FRMD3 by a FERM Domain Mutation Causes Hypomyelinating Disease via Oligodendrocyte Dysfunction.

Hypomyelinating diseases are a heterogeneous group of neurodevelopmental disorders caused by genetic anomalies that impair myelin formation or maintenance. Here, we investigate a novel homozygous missense variant in FRMD3 (NM_174938.6:c.898T > C; p.C300R) in a 2-year-old male presenting with global developmental delay, hypotonia, mild ataxia, and MRI features consistent with hypomyelination. The variant affects a highly conserved cysteine within the FERM domain of FRMD3, and predicted to be deleterious by multiple in silico tools. Molecular dynamics simulations and biophysical analyses reveale

A Multi-parametric MRI and Machine Learning Study of Cerebellar Structure in Youth with Neurofibromatosis Type 1.

BACKGROUND: Neurofibromatosis type 1 (NF1) is a genetic condition caused by pathogenic variants of the NF1 gene. While alterations in cerebral structural and microstructural differences have been reported in NF1, the cerebellum remains largely unexplored. Since individuals with NF1 are at risk for cognitive difficulties, which are in turn associated with cerebellar processes, understanding the underlying neural mechanisms is critical for intervention development. METHODS: Youth (5-16 years) with NF1 (n = 30) and unaffected youth (n = 40) participated in neuropsychological (i.e., neurocognitiv

Electroactive Dual-Drug Hydrogel-PLA Conduit for Nerve Regeneration.

Peripheral nerve injury (PNI) remains a major clinical challenge due to the limitations of autologous nerve grafting. Herein, a multifunctional composite nerve conduit was developed by integrating an oriented collagen-grafted polylactic acid (PLA) electrospun scaffold with a conductive and anti-inflammatory hydrogel to enhance peripheral nerve regeneration. The hydrogel was fabricated by chemically cross-linking carboxymethyl chitosan (CMCS) and sodium carboxymethyl cellulose (CMC-Na), incorporating polyaniline (PANI) to impart electrical conductivity and loading rhein (RH) for sustained anti-

Puerarin attenuates binge ethanol-induced cortical neurotoxicity in association with AKT/mTOR signaling and autophagy-related responses.

Puerarin (Pue), a major isoflavone derived from Pueraria lobata, has demonstrated neuroprotective potential in multiple neurological disorders; however, its effects on ethanol (EtOH)-induced cortical injury and the associated molecular responses remain incompletely understood. In the present study, network pharmacology was combined with in vivo and in vitro experiments to investigate molecular responses associated with the effects of Pue on EtOH-induced neurotoxicity. Public databases were used to predict targets of Pue and alcohol-related brain injury, followed by protein-protein interaction

Novel Neurotrophin-3 peptidomimetic synthetic neurotrophin promotes neurological recovery after spinal cord injury.

With 500,000 people paralyzed by traumatic spinal cord injuries (SCI) each year worldwide, there is a strong need for treatments that improve mobility and overall life quality after such injuries. There are currently no FDA-approved drugs that reliably improve locomotor function after injury. This is due to the complex nature of injury and our lack of ability to regenerate damaged areas of our central nervous system. Neurotrophin-3 (NT-3) is a neuroprotective, neuritogenic, pro-synaptogenic, and pro-myelinating neurotrophic factor. By binding to and activating the Tropomyosin receptor kinase (

Distinct patterns of microstructural brain changes in Alzheimer's disease subtypes: Diffusion MRI metrics in braak stage-related regions.

BackgroundAlzheimer's disease (AD) is pathologically defined by the accumulation of amyloid-β and phosphorylated tau and subsequent neuronal death. These neuropathological changes typically accumulate in the medial temporal lobe before spreading to other cortical regions (Braak staging pattern). Recent studies using postmortem brain tissue and neuroimaging data have revealed AD subtypes with distinct neuropathological patterns. Hippocampal-sparing (HpSp) subtype of AD shows the preservation of medial temporal lobe volume, compared to typical AD; however, underlying changes in the brain microst

Nitric oxide refines retinal circuit architecture independently of retinal wave dynamics.

How diffusible neuromodulators control neural circuit assembly remains a key open question in neuroscience. While nitric oxide (NO) is known to regulate mature synaptic plasticity, its role during development, specifically whether it shapes circuits through activity-dependent or activity-independent pathways, has not been fully understood. Here, we combine ultrasensitive electron paramagnetic resonance (EPR) spectroscopy, 4096-channel high-density multielectrode array (HD-MEA) recordings, and advanced graph-theoretical analysis to study how NO contributes to retinal network formation during a

Cell-specific expression of APP and GABABR1 isoforms in wild-type and the APP NL-G-F mouse model of Alzheimer's disease.

Amyloid precursor protein (APP) is a type I transmembrane protein that undergoes proteolytic processing to generate amyloid-β, the main component of amyloid plaques found in brains with Alzheimer's disease (AD). APP proteolytic processing also generates soluble APP alpha (sAPPα) which modulates synaptic transmission and neurite outgrowth through the γ-aminobutyric acid type B receptor (GABABR). sAPPα specifically binds the 1a isoform of GABABR subunit 1 (R1a) which contains two sushi domains absent in R1b. Here, we define the cell type-specific expression patterns of APP and GABABR1a in the br

Hopx(+) optic nerve head-astrocytes counter neuronal stress and glaucoma damage.

Retinal ganglion cell (RGC) axons form the optic nerve (ON). Numerous age-related ON diseases, including glaucoma, the second most common cause of worldwide blindness, result from multiple RGC stressors. Nearly all ON astrocytes in the optic nerve head (ONH): the junctional region between the ON and the retina in young-adult rodents expresses the homeodomain only (Hopx) protein. Hopx(+) ONH astrocytes are depleted during aging. ONH primary cultures which include Hopx(+) astrocytes secrete extracellular vesicles (ONH-EVs) which selectively enhance RGC survival and neurite extension in culture,

A Novel 3D Semi-Automated Full Quantification Technique for Detection of Intraneural Phospho-α-Synuclein in Skin Biopsies.

BACKGROUND: Parkinson's disease (PD) and multiple system atrophy (MSA) are synucleinopathies marked by α-synuclein aggregation, while progressive supranuclear palsy (PSP) is a tauopathy. Clinical overlap between these diseases complicates diagnosis. Detection of intraneural S129 phospho-α-synuclein (pαSyn) via immunofluorescence staining (IF) in skin biopsies shows diagnostic promise. However, prior studies rarely addressed differentiation between synucleinopathies and tauopathies and lacked assessment of varying pαSyn burden-particularly relevant for this aim. METHODS: In this cross-sectiona

Isolation and functional characterization of novel neuropeptides regulating hippocampal neurogenesis.

Neuropeptides (NPs) are small peptides, produced and secreted by neurons, serving as neuromodulators and chemical messengers that influence neuronal growth, proliferation, and differentiation. Despite their diverse and important roles, the discovery and functional characterization of many NPs remain incomplete. This study aimed to extract and identify novel NPs from rat brain tissue. For this purpose, NPs were purified using gel filtration chromatography and sequenced by mass spectrometry (MS). After characterization, one candidate peptide (NP1) was selected for functional evaluation in primar

Aligned fibrous scaffolds containing glycosaminoglycan mimetics enhance Schwann cell-supported axon growth and myelination.

Objective.Traumatic spinal cord injury causes functional impairments in large part due to the limited ability of damaged axons to regenerate. During development, the extracellular matrix (ECM) plays a critical role in providing molecular cues that promote and direct axon growth. Biomaterials that mimic the physicochemical properties of the ECM may be promising for promoting axon growth in the injured spinal cord. Glycosaminoglycans (GAGs) in the ECM during development are known to direct axon growth and guidance. We previously reported that GAG-mimetic cellulose sulfate, which can be synthesiz

cAmbly, a peptide derived from the star tick Amblyomma sculptum salivary gland transcriptome, attenuates inflammatory pain by activating endogenous opioid pathways.

Pain affects more than 1.5 billion people worldwide and remains a leading cause of disability, but currently available analgesics often have limited efficacy and significant adverse effects. In this context, bioactive molecules derived from animal venoms and secretions have emerged as promising novel therapeutic agents. Here, we investigated the antinociceptive and anti-inflammatory properties of cAmbly, a cell-penetrating peptide derived from the C-terminal region of Amblyomin-X, a protein from the salivary gland transcriptome of the Amblyomma sculptum tick. FUS inactivation promotes leptomeningeal metastasis of lung cancer through the up-regulation of CD36.

The prognosis of non-small cell lung cancer (NSCLC) with leptomeningeal metastasis (LM) is dismal. The regulators of LM progression remain elusive, thus impeding effective clinical intervention. Here, we performed in vivo genome-wide CRISPR-based screens and found that fused in sarcoma (FUS) ablation promoted LM in both PC9 and A549 cells. FUS repressed CD36 expression by directly interacting with and destabilizing PPARA (peroxisome proliferator-activated receptor α) messenger RNA. CD36 augmented fatty acid uptake and oxidative phosphorylation in NSCLC cells. Matrix metallopeptidase 2 (MMP2) w

The class-IIa HDAC inhibitor TMP269 promotes BMP-Smad signalling and is neuroprotective in in vitro and in vivo 6-hydroxydopamine models of Parkinson's disease.

Degeneration of midbrain nigrostriatal dopaminergic neurons is a pathological hallmark of Parkinson's disease (PD). Peripheral delivery of a compound(s) to arrest or slow this dopaminergic degeneration is a key therapeutic goal. Pan-inhibitors of histone deacetylase (HDAC) enzymes, key epigenetic regulators, have shown therapeutic promise in PD models. However as there are several classes of HDACs (ClassI-IV), class-specific inhibition will be important to ensure target specificity. Here we examine the neuroprotective potential of the Class-IIa HDAC inhibitor, TMP269. We show that TMP269 prote

Targeting Nerve Fiber Outgrowth Inhibition After Experimental Spinal Cord Injury: A Systematic Review and Meta-analysis of Chondroitinase ABC.

BACKGROUND: Spinal cord injury (SCI) can impair motor, sensory, and autonomic function. The formation of the glial scar comprises protective as well as inhibitory neurite outgrowth properties operated by the deposition of chondroitin sulfate proteoglycans (CSPG). Chondroitinase ABC (ChABC) can degrade CSPG and foster neuroaxonal plasticity as a therapeutic approach to restore locomotor function after SCI. OBJECTIVES: To systematically review experimental ChABC treatments after SCI and assess their efficacy for locomotor function a comprehensive literature search was conducted following pre-re

The synthetic cannabinoids ADB-FUBINACA and AMB-FUBINACA enhance in vitro neurodifferentiation of NG108-15 cells, along with PGC-1α dysregulation and mitochondrial dysfunction.

There is growing concern regarding the use of Synthetic Cannabinoids (SCs) by young adults (including pregnant and breastfeeding women, and women of childbearing age), due to their potential to cause neurodevelopmental disorders. Here, we first-hand assessed the in vitro impact of two indazole-derived SCs, ADB-FUBINACA and AMB-FUBINACA, on the neurodifferentiation of NG108-15 cells, especially focusing on their modulation of mitochondrial function during such process. Both SCs tested enhanced neurite outgrowth in NG108-15 cells at biologically-relevant concentrations (< 1 μM), a process that w
