Facultative predation expands the ecological repertoire of Streptomyces.

Microbial predators obtain energy from killing other living cells. Streptomyces are soil bacteria that are known to produce numerous catabolic enzymes and antimicrobial compounds to defend against competing organisms. Here, we demonstrate that Streptomyces are predatory bacteria that prey on Saccharomyces cerevisiae. Time-lapse fluorescence microscopy and scanning electron microscopy revealed that predation is initiated by physical contact between Streptomyces lavendulae YAKB-15 and yeast cells. Comparative transcriptomics indicated that the interaction triggered the production of numerous ext

Key role of TLR3 in type I IFN expression and apoptosis induction in IBDV-infected chicken fibroblast cells.

Infectious Bursal Disease Virus (IBDV) (Avibirnavirus genus, Birnaviridae family) is a non-enveloped virus with a double-stranded RNA (dsRNA … reovirus (ARV). Our findings also suggest that TLR3 may also play a role in viral release into the extracellular space. Additionally, receptor interacting protein kinase 1 (RIPK1), a protein that interacts with TLR3 through the adaptor Toll/IL-1 receptor (TIR) domain-containing adaptor-inducing interferon-β (TRIF), was shown to contribute to both IFN production and …

Lymphatic Malformations With Activating KRAS Mutations Impair Lymphatic Valve Development Through Matrix Metalloproteinases.

BACKGROUND: Lymphatic malformations are lesions that can be due to inherited or somatic mutations, and they lead to a defective lymphatic vasculature. Activating KRAS (Kirsten rat sarcoma viral oncogene homolog) mutations have been identified recently in patients with lymphatic malformation with lymphedema, chylous ascites, or life-threatening chylothorax. In a lymphatic malformation mouse model, KRAS mutations are associated with a loss of lymphatic valves, which has been proposed to cause chylothorax via retrograde lymph flow into the pleural space. However, the mechanisms underlying the los

Large mammalian herbivores influence the deployment of microbial extracellular enzymes to control soil carbon cycling.

UNLABELLED: Large mammalian grazers have regulated biogeochemical cycles across 40% of the Earth's terrestrial surface for millennia. While grazer effects on organic matter entering the soil are becoming clear, their effect on the downstream microbial foraging through extracellular-enzymes (EE), which link processes at the cellular level to ecosystem-level flow of matter remain less clear. We assessed how large mammalian grazers influence microbial foraging in soil using two lines of evidence. First, utilising a 14-year-old long-term grazer-exclusion experiment in the Trans-Himalayan ecosystem

White matter microstructure of language pathways in non-verbal autism: insights from diffusion tensor imaging and myelin water imaging.

INTRODUCTION: Absence of language development is a condition encountered across a large range of neurodevelopmental disorders, including a significant proportion of children with autism spectrum disorder. The neurobiological underpinnings of non-verbal ASD (nvASD) remain poorly understood. METHODS: This study employed multimodal MRI to investigate white matter (WM) microstructural abnormalities in nvASD, focusing on language-related pathways. We analyzed diffusion tensor imaging (DTI) metrics-fractional anisotropy (FA), mean diffusivity (MD), and radial diffusivity (RD)-alongside myelin water

Analysis of Kallikrein 6, Acetyl-α-Tubulin, and Aquaporin 1 and 2 Expression Patterns During Normal Human Nephrogenesis and in Congenital Anomalies of the Kidney and Urinary Tract (CAKUT).

Background/Objectives: The human kallikrein-related peptidase 6 (KLK6), a serine protease with trypsin-like properties, belongs to the 15-member kallikrein (KLK) gene family and is predominantly recognized for its role in oncogenesis, neurodegenerative disorders, and skin conditions. Aquaporins (AQPs) are integral membrane proteins that facilitate water transport across cell membranes. AQP1 is constitutively active in the kidneys and plays a crucial role in reabsorbing filtered water, while AQP2 is regulated by vasopressin and is essential for maintaining body fluid homeostasis. The primary ob

Early host-parasite interaction models reveal a key role for fibrinolysis in Fasciola hepatica intestinal migration.

BACKGROUND: Fasciola hepatica is the most common etiologic agent of fasciolosis, a parasitic disease that affects millions of ruminants worldwide and a zoonotic human infection of public health concern. Upon ingestion of infective metacercariae, F. hepatica newly excysted juveniles (FhNEJ) emerge in the duodenum and cross the intestinal wall to initiate a migration route that culminates with their establishment within the hepatic bile ducts. The ability of FhNEJ to exploit the broad-spectrum activities of host plasmin, the central protease of the fibrinolytic system, has been proposed as a str

Activation of the COX-2/mPGES-1/PGE-2 cascade through the NLRP3 inflammasome contributes to Angiostrongylus cantonensis-induced eosinophilic meningoencephalitis.

Prostaglandin E2 (PGE-2) is synthesised by cyclooxygenase-2 (COX-2) and microsomal prostaglandin E synthase 1 (mPGES-1). PGE-2 exhibits pro-inflammatory properties in inflammatory conditions. However, there remains limited understanding of the COX-2/mPGES-1/PGE-2 pathway in Angiostrongylus cantonensis-induced meningoencephalitis. This study revealed several key findings regarding the activation of the COX-2/mPGES-1/PGE-2 pathway and its correlation with eosinophilic meningoencephalitis induced by A. cantonensis infection. Immunostaining revealed an increase in the expression of COX-2 and mPGES

Development of properly-polarized trophoblast stem cell-derived organoids to model early human pregnancy.

UNLABELLED: The development of human trophoblast stem cells (hTSC) and stem cell-derived trophoblast organoids has enabled investigation of placental physiology and disease and early maternal-fetal interactions during a stage of human pregnancy that previously had been severely restricted. A key shortcoming in existing trophoblast organoid methodologies is the non-physiologic position of the syncytiotrophoblast (STB) within the inner portion of the organoid, which neither recapitulates placental villous morphology in vivo nor allows for facile modeling of STB exposure to the endometrium or the

MicroRNA-224-5p nanoparticles balance homeostasis via inhibiting cartilage degeneration and synovial inflammation for synergistic alleviation of osteoarthritis.

MicroRNAs play a crucial role in regulating cartilage extracellular matrix (ECM) metabolism and are being explored as potential therapeutic targets for osteoarthritis (OA). The present study indicated that microRNA-224-5p (miR-224-5p) could balance the homeostasis of OA via regulating cartilage degradation and synovium inflammatory simultaneously. Multifunctional polyamidoamine dendrimer with amino acids used as efficient vector to deliver miR-224-5p. The vector could condense miR-224-5p into transfected nanoparticles, which showed higher cellular uptake and transfection efficiency compared to

Aldosterone-Induced Sarco/Endoplasmic Reticulum Ca2+ Pump Upregulation Counterbalances Cav1.2-Mediated Ca2+ Influx in Mesenteric Arteries.

In mesenteric arteries (MAs), aldosterone (ALDO) binds to the endogenous mineralocorticoid receptor (MR) and increases the expression of the voltage-gated L-type Cav1.2 channel, an essential ion channel for vascular contraction, sarcoplasmic reticulum (SR) Ca2+ store refilling, and Ca2+ spark generation. In mesenteric artery smooth muscle cells (MASMCs), Ca2+ influx through Cav1.2 is the indirect mechanism for triggering Ca2+ sparks. This process is facilitated by plasma membrane-sarcoplasmic reticulum (PM-SR) nanojunctions that drive Ca2+ from the extracellular space into the SR via Sarco/End…

MicroRNA-196a-5p overexpression in Wharton's jelly umbilical cord stem cells promotes their osteogenic differentiation and new bone formation in bone defects in the rat calvarium.

The peri-tooth root alveolar loss often does not have sufficient space for repair material transplantation and plasticity. Mesenchymal stem cell (MSC) sheets have an advantage in providing more extracellular matrix (ECM) and may prove to be a new therapeutic consideration for this bone defect repair. The identification of key regulators that stimulate MSCs' osteogenic potential and sheet-derived ECM deposition is the key to promoting its application. In this study, we found that inhibition or overexpression of miR-196a-5p led to a decline or enhancement, respectively, in the alkaline phosphata

Geometry of the Carotid Artery and Its Association With Pathologic Changes in a Chinese Population.

OBJECTIVES: Carotid artery geometry influences blood flow disturbances and is thus an important risk factor for carotid atherosclerosis. Extracellular matrix (ECM) and yes-associated protein (YAP) expression may play essential roles in the pathophysiology of carotid artery stenosis, but the effect of blood flow disturbances of carotid bifurcation location on the ECM is unknown. We hypothesized that carotid artery anatomy and geometry are independently associated with the ECM and YAP expression. METHODS: In this cross-sectional study, 193 patients were divided into two groups: an asymptomatic
