Printing the Pathway Forward in Bone Metastatic Cancer Research: Applications of 3D Engineered Models and Bioprinted Scaffolds to Recapitulate the Bone-Tumor Niche.

Breast cancer commonly metastasizes to bone, resulting in osteolytic lesions and poor patient quality of life. The bone extracellular matrix (ECM) plays a critical role in cancer cell metastasis by means of the physical and biochemical cues it provides to support cellular crosstalk. Current two-dimensional in-vitro models lack the spatial and biochemical complexities of the native ECM and do not fully recapitulate crosstalk that occurs between the tumor and endogenous stromal cells. Engineered models such as bone-on-a-chip, extramedullary bone, and bioreactors are presently used to model cellu

A Novel Bespoke Hypertrophic Scar Treatment: Actualizing Hybrid Pressure and Silicone Therapies with 3D Printing and Scanning.

The treatment of hypertrophic scars (HSs) is considered to be the most challenging task in wound rehabilitation. Conventional silicone sheet therapy has a positive effect on the healing process of HSs. However, the dimensions of the silicone sheet are typically larger than those of the HS itself which may negatively impact the healthy skin that surrounds the HS. Furthermore, the debonding and displacement of the silicone sheet from the skin are critical problems that affect treatment compliance. Herein, we propose a bespoke HS treatment design that integrates pressure sleeve with a silicone sh

Sustainable release of vancomycin from micro-arc oxidised 3D-printed porous Ti6Al4V for treating methicillin-resistant Staphylococcus aureus bone infection and enhancing osteogenesis in a rabbit tibia osteomyelitis model.

Elimination of infection and enhancement of osteogenesis by orthopaedic implants are two critical factors in the treatment of complex bone infections. A prolonged and expensive procedure requiring two surgical steps and a 6-8-week period of joint immobilisation is utilised as a primary treatment for revision arthroplasty of an infected prosthesis, greatly affecting long-term patient care for the ageing population. Here, we evaluated the effects of vancomycin-loaded in micro-arc oxidised (MAO) three-dimensional (3D) printed porous Ti6Al4V scaffolds on osteogenesis. This system showed a high loa

Do 3-D Printed Handheld Models Improve Surgeon Reliability for Recognition of Intraarticular Distal Radius Fracture Characteristics?

BACKGROUND: For fracture care, radiographs and two-dimensional (2-D) and three-dimensional (3-D) CT are primarily used for preoperative planning and postoperative evaluation. Intraarticular distal radius fractures are technically challenging to treat, and meticulous preoperative planning is paramount to improve the patient's outcome. Three-dimensionally printed handheld models might improve the surgeon's interpretation of specific fracture characteristics and patterns preoperatively and could therefore be clinically valuable; however, the additional value of 3-D printed handheld models for fra

3D Artificial Skin Model As a Novel Strategy for the Detection of Pyroptosis-Cascade Activation in Amyotrophic Lateral Sclerosis.

Amyotrophic lateral sclerosis (ALS) is a severe adult-onset neurodegenerative disease with limited treatment approaches. Evidence has shown that degeneration of cutaneous nerves may reflect neurodegenerative processes occurring within the central nervous system. Although skin biopsy is widely adopted in clinical practice, the procedure is invasive and requires multiple patients' tissue removals. Therefore, we developed a 3D innervated skin model by combining 3D printing of methacrylated hyaluronic acid as an innovative tool for better reproducing the dermis and epidermis and electrospinning of

Multifunctional 3D-printed Mg-doped bioactive glass/PLLA scaffolds with quercetin delivery for remodeling the osteoporotic microenvironment via osteogenic-angiogenic coupling and osteoclast inhibition.

Osteoporotic bone defects remain challenging to repair due to a dysregulated microenvironment characterized by impaired osteogenesis, insufficient angiogenesis, and excessive osteoclast activity. Herein, we report a multifunctional three-dimensional (3D)-printed scaffold that integrates quercetin-loaded magnesium-doped mesoporous bioactive glass (Mg-BG-Que) into a poly (L-lactic acid) (PLLA) matrix, enabling simultaneous modulation of these pathological features. The composite scaffold affords sustained co-delivery of quercetin and bioactive ions (Mg, Si, Ca, and P), thereby providing both str

3D Bioprinting of High-Grade Serous Ovarian Cancer Cells: Workflow for Gelatin-Based Bioink Formulation with Hyaluronic Acid and Printability Assessment.

Ovarian cancer (OC) is one of the leading causes of cancer-related mortality in women, primarily due to the late-stage diagnosis and limited therapeutic efficacy. Three-dimensional (3D) bioprinting enables the fabrication of cell-laden scaffolds that better recapitulate the native spatial organization and may support the investigation of tumor cell behavior. However, high-grade serous ovarian cancer (HG-SOC) cell subtypes remain largely unexplored in bioprinting, and it remains unclear whether a different scaffold composition is required to provide an optimal niche for each subtype. To address
