Eupatilin ameliorates spinal cord injury by inhibiting damage-associated microglia and optimizing the regenerative microenvironment.

Microglia represent critical therapeutic targets in spinal cord injury (SCI), with damage-associated microglia (DAM) playing key roles in neuroinflammation and tissue repair. Through integrated in-silico analysis of single-cell RNA sequencing (scRNA-seq) and microarray datasets, we identified DAM subsets specific to acute SCI characterized by hub genes Fcer1g, Grn, and Gusb. Using a C57BL/6 mouse spinal cord contusion model, we validated increased DAM accumulation post-injury and demonstrated their propensity to transition toward homeostatic microglia (MG2). Eupatilin treatment promoted DAM-to

Molecular characterization of Pseudomyxoma peritonei with single-cell and bulk RNA sequencing.

Pseudomyxoma peritonei (PMP), a rare condition characterized by mucinous ascites in the peritoneal cavity, often leads to a poor prognosis. However, omics profiling of this disease remains significantly underexplored. Here, we present single-cell transcriptomic profiling of five PMP cases to identify cell type-specific gene features associated with PMP pathogenesis. Additionally, we provide bulk RNA-seq datasets from two independent cohorts: 19 fresh frozen tissue samples (12 PMPs) and 34 formalin-fixed paraffin-embedded (FFPE) samples (25 PMPs). We also offer protein expression data from a ti

Prenatal diagnosis of partial duplication chromosome 21q11.2-21q21.1: A case report and literature review.

OBJECTIVE: We report the prenatal diagnosis for a small supernumerary marker chromosome derived from chromosome 21q11.2-q21.1. CASE REPORT: A 40-year-old woman presenting at 13 weeks of gestation underwent noninvasive prenatal testing (NIPT) due to advanced maternal age. A positive finding for trisomy 21 urged genetic counseling and subsequent amniocentesis. Amniocentesis revealed a karyotype of 47, XX,+mar. The parental karyotypes were normal, and prenatal ultrasound findings were unremarkable. At a follow-up genetic counseling session, additional prenatal genetic testing was undertaken, spe

Duplication of Chromosome 22q as a Key Event in Hermaphroditism and Hypospadias.

Hermaphroditism can be triggered by 22q11.2 duplication in 46,XX individuals without SRY, indicating alternate routes regulating sexual diff … s. Despite their contributions, genes such as ZNF74 likely have little effect. Although chromosomal microarray analysis (CMA) and fluorescence in situ hybridization (FISH) improve the diagnosis of 22q11.2-associated hermaphroditism, the syndrome's varied expressivity makes clinical evaluation and prediction more difficult. These uncommon rearrangements are caused by …

Mycobacterium tuberculosis stimulates cuproptosis by regulating Lnc-Gm5532 to target FDX1 for bacteria intracellular survival.

Cuproptosis is a novel cell death modality but its regulatory role in tuberculosis remains obscure. This study is committed to explore the specific mechanism of cuproptosis induced by Mycobacterium tuberculosis (M.tb) infection. We confirmed that M.tb induced cuproptosis contributes to its intracellular survival. Then microarray analysis was employed to screen M.tb-infected DE mRNAs and LncRNAs which may be potential regulators of cuproptosis. We focus on a key cuproptosis gene FDX1, and 7 LncRNAs that can target FDX1. Among them, LncRNA-Gm5532 upregulated the expression of FDX1 by sponging mi…

Circulating Autoantibody Profiling Identifies LIMS1 as a Potential Target for Pathogenic Autoimmunity in pathologic Myopia.

High myopia is a leading cause of blindness worldwide, among which pathologic myopia, characterized by typical myopic macular degeneration, is the most detrimental. However, its pathogenesis remains largely unknown. Here, using a HuProt array, we first initiated a serological autoantibody profiling of high myopia and identified 18 potential autoantibodies, of which anti-LIMS1 autoantibody was validated by a customized focused microarray. Further subgroup analysis revealed its actual relevance to pathologic myopia, rather than simple high myopia without myopic macular degeneration. Mechanistica

Genomic structural variants analysis in leukemia by a novel cytogenetic technique: Optical genome mapping.

Genomic structural variants (SVs) play a pivotal role in driving the evolution of hematologic malignancies, particularly in leukemia, in which genetic abnormalities are crucial features. Detecting SVs is essential for achieving precise diagnosis and prognosis in these cases. Karyotyping, often complemented by fluorescence in situ hybridization and/or chromosomal microarray analysis, provides standard diagnostic outcomes for various types of SVs in front-line testing for leukemia. Recently, optical genome mapping (OGM) has emerged as a promising technique due to its ability to detect all SVs id…

Glycosylation profiles of breast cancer cells may represent clonal variations of multiple organ metastases.

Glycosylation changes of cancer cells are known to be associated with malignant progression and metastases and potentially determine the organ-selective nature of metastasis as theorized by Paget (Lancet 1:571-573, 1889). Cellular glycans play a variety of roles in the processes of metastasis and may be unique to the cells that metastasize to different organs. We analyzed the glycosylation profiles of the primary tumor and tumors metastasized to lymph node, liver, lung, brain, bone, thyroid, kidney, adrenal, small intestine and pancreas in an autopsy case of breast cancer employing a lectin mi

FZD7: A potential biomarker for endometriosis.

BACKGROUND: Endometriosis is a chronic inflammatory, benign disorder that often co-occurs with adenomyosis and/or leiomyoma. The overall incidence of endometriosis in reproductive period women was nearly 10%. However, the exact mechanisms of endometriosis-associated pathogenesis are still unknown. METHODS: In this study, we aimed to investigate whether Frizzled-7 (FZD7) would effectively promote the development of endometriosis. The microarray-based data analysis was performed to screen endometriosis-related differentially expressed genes. This process uncovered specific hub genes, and the ne

An Asymptomatic, Ectopic Mass as a Presentation of Adrenocortical Carcinoma Due to a Novel Germline TP53 p.Phe338Leu Tetramerisation Domain Variant.

Adrenocortical carcinoma (ACC) is a rare cancer in childhood. ACC is frequently associated with germline TP53 variants, with founder effects especially due to the p.Arg337His mutation. ACC leads to the secretion of adrenocortical hormones, resulting in endocrine syndromes, which is the usual trigger for establishing the diagnosis. We present a surprising ACC pathology in a non-secreting, ectopic retroperitoneal tumour in a 4-year-old boy, successfully controlled with chemotherapy and mitotane after microscopically incomplete tumour resection with spillage. Genomic analysis (gene panel sequenci

GLIS1 in Cancer-Associated Fibroblasts Regulates the Migration and Invasion of Ovarian Cancer Cells.

A cancer-associated fibroblasts (CAFs) are the most important players that modulate tumor aggressiveness. In this study, we aimed to identify CAF-related genes in ovarian serous carcinomas (OSC) that account for the high incidence and mortality of ovarian cancers (OCs) and to develop therapeutic targets for tumor microenvironment modulation. Here, we performed a microarray analysis of CAFs isolated from three metastatic and three nonmetastatic OSC tissues and compared their gene expression profiles. Among the genes increased in metastatic CAFs (mCAFs), GLIS1 (Glis Family Zinc Finger 1) showed …
