ACPA-positive and ACPA-negative rheumatoid arthritis differ in their requirements for combination DMARDs and corticosteroids: secondary analysis of a randomized controlled trial.

INTRODUCTION: UK guidelines recommend that all early active rheumatoid arthritis (RA) patients are offered combination disease-modifying antirheumatic drugs (DMARDs) and short-term corticosteroids. Anti-citrullinated protein antibody (ACPA)-positive and ACPA-negative RA may differ in their treatment responses. We used data from a randomized controlled trial - the Combination Anti-Rheumatic Drugs in Early RA (CARDERA) trial - to examine whether responses to intensive combination treatments in early RA differ by ACPA status. METHODS: The CARDERA trial randomized 467 early active RA patients to

Clinical correlations with Porphyromonas gingivalis antibody responses in patients with early rheumatoid arthritis.

INTRODUCTION: Prior studies have demonstrated an increased frequency of antibodies to Porphyromonas gingivalis (Pg), a leading agent of periodontal disease, in rheumatoid arthritis (RA) patients. However, these patients generally had long-standing disease, and clinical associations with these antibodies were inconsistent. Our goal was to examine Pg antibody responses and their clinical associations in patients with early RA prior to and after disease-modifying antirheumatic drug (DMARD) therapy. METHODS: Serum samples from 50 DMARD-naïve RA patients were tested using an enzyme-linked immunoso

Glucagon-like peptide-2 (GLP-2) increases net amino acid utilization by the portal-drained viscera of ruminating calves.

Glucagon-like peptide-2 (GLP-2) increases small intestinal mass and blood flow in ruminant calves, but its impact on nutrient metabolism across the portal-drained viscera (PDV) and liver is unknown. Eight Holstein calves with catheters in the carotid artery, mesenteric vein, portal vein and hepatic vein were paired by age and randomly assigned to control (0.5% bovine serum albumin in saline; n = 4) or GLP-2 (100 μg/kg BW per day bovine GLP-2 in bovine serum albumin; n = 4). Treatments were administered subcutaneously every 12 h for 10 days. Blood flow was measured on days 0 and 10 and included

Dual role for nitric oxide in dynorphin spinal neurotoxicity.

The pharmacological effects of nitric oxide synthase (NOS) inhibitors, NO donor, and NOS substrate on dynorphin(Dyn) A(1-17) spinal neurotoxicity were studied. Intrathecal (i.t.) pretreatment with both 7-nitroindazole 1 micromol, a selective neuronal constitutive NOS (ncNOS) inhibitor, and aminoguanidine 1 micromol, a selective inducible NOS (iNOS) inhibitor, 10 min prior to i.t. Dyn A(1-17) 20 nmol significantly ameliorated Dyn-induced neurological outcome. Both 7-nitroindazole and aminoguanidine significantly antagonized the increases of cNOS and iNOS activities measured by conversion of 3H-

Genetic and epigenetic alteration of the programmed cell death 1 in rheumatoid arthritis.

BACKGROUND: Rheumatoid arthritis (RA) is an autoimmune disease where both genetics and epigenetics are contributing factors. In order to discover genetic and epigenetic associations with RA and its phenotypes, we analysed RNA expression, DNA variations and DNA methylation of programmed cell death 1 (PDCD1) in a cohort of RA patients and healthy controls. METHODS: RA patients (n = 206) and healthy controls (n = 234) were included for analysis of PDCD1 expression, PDCD1 polymorphisms and PDCD1 methylation. Differences in continuous variables between groups were compared by applying t tests. Ass

Bacterial infection induces nitric oxide synthase in human neutrophils.

The identification of human inflammatory cells that express inducible nitric oxide synthase and the clarification of the role of inducible nitric oxide synthase in human infectious or inflammatory processes have been elusive. In neutrophil-enriched fractions from urine, we demonstrate a 43-fold increase in nitric oxide synthase activity in patients with urinary tract infections compared with that in neutrophil-enriched fractions from noninfected controls. Partially purified inducible nitric oxide synthase is primarily membrane associated, calcium independent, and inhibited by arginine analogue

Transcriptomic signatures of classical monocytes reveal pro-inflammatory modules and heterogeneity in polyarticular juvenile idiopathic arthritis.

INTRODUCTION: Polyarticular juvenile idiopathic arthritis (pJIA) is a childhood-onset autoimmune disease. Immune cells contribute to persistent inflammation observed in pJIA. Despite the crucial role of monocytes in arthritis, the precise involvement of classical monocytes in the pathogenesis of pJIA remains uncertain. Here, we aimed to uncover the transcriptomic patterns of classical monocytes in pJIA, focusing on their involvement in disease mechanism and heterogeneity. METHODS: A total of 17 healthy subjects and 18 premenopausal women with pJIA according to ILAR criteria were included. Cla

Nitric oxide production contributes to Bacillus anthracis edema toxin-associated arterial hypotension and lethality: ex vivo and in vivo studies in the rat.

We showed previously that Bacillus anthracis edema toxin (ET), comprised of protective antigen (PA) and edema factor (EF), inhibits phenylephrine (PE)-induced contraction in rat aortic rings and these effects are diminished in endothelial-denuded rings. Therefore, employing rat aortic ring and in vivo models, we tested the hypothesis that nitric oxide (NO) contributes to ET's arterial effects. Compared with rings challenged with PA alone, ET (PA + EF) reduced PE-stimulated maximal contractile force (MCF) and increased the PE concentration producing 50% MCF (EC50) (P < 0.0001). Compared with pl

A NET-associated immunothrombotic phenotype is associated with coronary no-reflow and myocardial injury after primary PCI in ST-segment elevation myocardial infarction: a real-world translational cohort.

BACKGROUND: Coronary no-reflow after primary percutaneous coronary intervention (PCI) reflects failure of myocardial reperfusion despite treatment of the culprit epicardial lesion. Neutrophil extracellular traps (NETs) and immunothrombosis provide a plausible link between sterile inflammation, thrombus biology, and microvascular obstruction. We evaluated whether a clinically measurable NET-associated immunothrombotic phenotype (NET-IP) was associated with no-reflow and myocardial injury after primary PCI. METHODS: This single-center translational cohort included 742 patients with ST-segment e

Overexpression of Soluble Fibrinogen-like Protein 2 in MSCs Ameliorates Renal Ischemia-Reperfusion Injury in Mice by Modulating Neutrophils.

BACKGROUND: Renal ischemia-reperfusion (I/R) injury is a major cause of graft dysfunction and failure, driving inflammation and tissue damage. Mesenchymal stem cells (MSCs) possess therapeutic potential due to their immunomodulatory properties. Notably, neutrophils express the inhibitory receptor CD32b, which is a specific target of the immunosuppressive molecule soluble fibrinogen-like protein 2 (sFgl2). AIMS: To investigate the therapeutic efficacy and underlying mechanisms of genetically engineered MSCs expressing sFgl2 (sFgl2-MSCs) in treating renal I/R injury, with a focus on neutrophil

S-propargyl-cysteine remodels the HCC microenvironment and potentiates anti-PD-1 therapy through CSE/H2S-linked vascular and immune reprogramming.

BACKGROUND: Hepatocellular carcinoma (HCC) is characterized by aberrant angiogenesis and an immunosuppressive tumor microenvironment, both of which limit durable responses to PD-1 blockade. Strategies that concurrently target vascular dysfunction and immune inhibition may improve therapeutic efficacy. METHODS: The interaction between S-propargyl-cysteine (SPRC) and cystathionine-gamma-lyase (CSE) was assessed by surface plasmon resonance. In human umbilical vein endothelial cells, NF-κB p65 and STAT3 phosphorylation, VEGFA expression, extracellular H2S production, CSE enzymatic activity, and

Profiling immune check point inhibitors - Induced arthritis: A systematic review and pooled analyses of cohort studies.

BACKGROUND: The growing use of immune checkpoint inhibitors (ICIs) in the oncology field along with the potential development of the so-called immune-related adverse events (irAEs) has introduced new diagnostic and therapeutic challenges for rheumatologists. Rheumatic irAEs (Rh-irAEs), particularly those involving the musculoskeletal system, are relatively common and sometimes underdiagnosed, with an estimated prevalence of up to 7% of ICIs treated patients. In this context, we conducted a systematic review and pooled analyses of cohort studies to better characterize ICIs-induced-arthritis (IC

Platelet-derived HMGB1 induces NETosis, exacerbating brain damage in the photothrombotic stroke model.

Following cerebral ischemia, neutrophil extracellular traps (NETs) contribute significantly to brain damage by exacerbating delayed immune cell infiltration and vascular injury. They are detected both in brain tissue and within blood vessels. Danger-associated molecular pattern (DAMP) molecules have been implicated in inducing NETosis after cerebral ischemia. This study investigated the role of High mobility group box 1 (HMGB1), a prototype DAMP molecule, in NETosis induction following photothrombotic stroke (PTS), with a particular focus on neutrophil-platelet interactions. In PTS, thrombi co

Neutrophil extracellular trap markers predict thrombotic risk in patients with hyperhomocysteinemia: development and validation of a predictive model.

BACKGROUND: Venous thromboembolic diseases pose a serious threat to human health, and their pathogenesis is closely associated with the abnormal activation of neutrophils and neutrophil extracellular traps (NETs). However, the mechanisms underlying the role of NETs in thrombosis related to Hyperhomocysteinemia (hHcy) remain unclear. In this study, we systematically investigated the correlation between NETs and hHcy-associated thrombosis by measuring key NETs markers (myeloperoxidase, MPO; citrullinated histone H3, CitH3) and cell-free DNA (cfDNA) levels. Based on multivariate analysis, we deve
