Prevalence of Extended-Spectrum β-Lactamases CTX-M-8 and CTX-M-2-Producing Salmonella Serotypes from Clinical and Nonhuman Isolates in Brazil.

We characterized extended-spectrum β-lactamases (ESBL) enzymes among Salmonella strains isolated in Brazil from 2009 to 2014. Salmonella recovered from both clinical and nonhuman (food, poultry, and environment) sources were subjected to antimicrobial susceptibility testing. β-lactamases genes were detected by polymerase chain reaction/sequencing; plasmid profiles and transferability were assessed by S1-pulsed field gel electrophoresis (PFGE). Genetic diversity was evaluated by XbaI-PFGE. Out of 630 Salmonella strains screened, 46 displayed ESBL phenotype, distributed across 11 different serot

Autoactivated KlPDR1 gene in the control of multidrug resistance in Kluyveromyces lactis.

The KlPDR1 gene encodes a zinc finger transcription factor that has recently been shown to be involved in the control of multidrug resistance of Kluyveromyces lactis . In this work, we provide evidence that the K. lactis KlPDR1 gene is under positive autoregulation by KlPdr1p, which plays a role in the activation of the main multidrug resistance transporter gene KlPDR5. Electrophoretic mobility shift assays, as well as the use of gusA reporter constructs, enabled us to identify the 5'-tataTCCGGGTAactt-3' sequence motif in the KlPDR1 promoter (in the position -326 to -319 bp) as the PDRE (pleio

Global in silico genomic epidemiology of tetracycline resistance in Staphylococcus aureus: one health dynamics and genotype-phenotype associations.

Tetracyclines remain widely used in human and veterinary medicine for the treatment of Staphylococcus aureus, including methicillin-resistant strains, and are key agents at the human–animal–environment interface. However, the global genomic epidemiology of tetracycline resistance mechanisms, their temporal stability, host structuring, and genotype–phenotype relationships across the tetracycline class remain incompletely resolved. We performed a large-scale in silico analysis to characterize tetracycline resistance determinants, minimum inhibitory concentration (MIC) patterns, and One Health dy

Comparative genomic and functional analysis of Staphylococcus caprae clinical isolates: insights into genetic diversity and opportunistic pathogenic potential.

Staphylococcus caprae, a coagulase-negative staphylococcus (CoNS), is an opportunistic microorganism increasingly identified in clinical specimens. However, its genomic and functional characterization in human infection have not yet been fully elucidated. In this study, we characterized the genomic and phenotypic diversity of two clinical S. caprae isolates, M89 and S13, to elucidate their opportunistic traits. Whole-genome sequencing was performed to evaluate genomic differences in antimicrobial resistance, virulence determinants, and protein-coding sequences, while their pathogenic potential

Differential perception of virulence factors of uropathogenic Escherichia coli at the level of chromatin dynamics of infected host cells.

INTRODUCTION: Uropathogenic Escherichia coli (UPEC) evades the innate immune response in the urinary tract through the coordinated action of various virulence factors encoded within distinct pathogenicity islands (PAIs). We have demonstrated that UPEC infection leads to the epigenetic regulation of host gene expression; however, the specific role of PAI-encoded virulence factors in this process remains largely unexplored. METHODS: In this follow-up study, we infected Galleria mellonella larvae with individual PAI deletion mutants of UPEC strain 536 to investigate the relationship between UPEC

Genomic and Phenotypic Insights Into Antibiotic Resistance and Virulence of Klebsiella pneumoniae From the Environment in Southern Taiwan.

BACKGROUND: Klebsiella pneumoniae is a major human pathogen responsible for healthcare- and community-associated infections and a critical contributor to the global antimicrobial resistance (AMR) crisis. Although widely present in the environment, its role in harboring multidrug-resistant (MDR) and hypervirulent strains (hvKp) remains insufficiently characterized. METHODS: This study assessed the prevalence, genomic diversity, and pathogenic potential of K. pneumoniae isolated from surface waters in southern Taiwan. A total of 62 sites were sampled, yielding 68 environmental isolates. Whole-g

ArcA modulates multidrug resistance and compound susceptibility in Klebsiella pneumoniae through ArcB-independent regulation of the SMR efflux pump kpnEF.

Klebsiella pneumoniae has become a major clinical and public health threat due to the increasing prevalence of healthcare-associated infections caused by multidrug-resistant strains. In this study, we demonstrated that the deletion of arcA of ArcAB two-component system diminished the susceptibility of K. pneumoniae to antibiotics, osmotic agents, disinfectants, and structural compounds, which was independent of arcB. RNA-seq analysis revealed a marked upregulation of SMR efflux pump genes kpnEF in the ΔarcA strain compared to the wild-type strain, while the ΔarcB strain exhibited no significan

Targeting epigenetic regulators to overcome drug resistance in the emerging human fungal pathogen Candida auris.

The rise of drug-resistant fungal species, such as Candida auris, poses a serious threat to global health, with mortality rates exceeding 40% and resistance rates surpassing 90%. The limited arsenal of effective antifungal agents underscores the urgent need for novel strategies. Here, we systematically evaluate the role of histone H3 post-translational modifications in C. auris drug resistance, focusing on acetylation mediated by Gcn5 and Rtt109, and methylation mediated by Set1, Set2, and Dot1. Mutants deficient in these enzymes exhibit varying degrees of antifungal drug sensitivity. Notably,

Host obesity impacts genetic variation in influenza A viral populations.

Obesity is well established as a risk factor for many noncommunicable diseases; however, its consequences for infectious disease are poorly understood. Here, we investigated the impact of host obesity on influenza A virus (IAV) genetic variation using a diet-induced obesity ferret model and the A/Hong Kong/1073/1999 (H9N2) strain. Using a co-caging study design, we investigated the maintenance, generation, and transmission of intrahost IAV genetic variation by sequencing viral genomic RNA obtained from nasal wash samples over multiple days of infection. We found evidence for an enhanced role o

Delineating the Acquired Genetic Diversity and Multidrug Resistance in Alcaligenes from Poultry Farms and Nearby Soil.

Alcaligenes faecalis is one of the most important and clinically significant environmental pathogens, increasing in importance due to its isolation from soil and nosocomial environments. The Gram-negative soil bacterium is associated with skin endocarditis, bacteremia, dysentery, meningitis, endophthalmitis, urinary tract infections, and pneumonia in patients. With emerging antibiotic resistance in A. faecalis, it has become crucial to understand the origin of such resistance genes within this clinically significant environmental and gut bacterium. In this research, we studied the impact of an

Characterization of a carbapenem-resistant Citrobacter amalonaticus coharbouring bla IMP-4 and qnrs1 genes.

Introduction. Members of the genus Citrobacter are facultative anaerobic Gram-negative bacilli belonging to the Enterobacterales [Janda J Clin Microbiol 1994; 32(8):1850-1854; Arens Clin Microbiol Infect 1997;3(1):53-57]. Formerly, Citrobacter species were occasionally reported as nosocomial pathogens with low virulence [Pepperell Antimicrob Agents Chemother 2002;46(11):3555-60]. Now, they are consistently reported to cause nosocomial infections of the urinary tract, respiratory tract, bone, peritoneum, endocardium, meninges, intestines, bloodstream and central nervous system. Among Citrobacte

Ten years of molecular epidemiology surveillance of Listeria monocytogenes in Chile 2008-2017.

Listeria monocytogenes causes severe diseases in humans, including febrile gastroenteritis and systemic infections that has a high mortality despite antibiotic treatment. This pathogen may cause massive outbreaks associated to the consumption of contaminated food products, which highlight its importance in public health. In the last decade, L. monocytogenes has emerged as a foodborne pathogen of major importance in Chile. A previous work showed that in Chile during 2008 and 2009, L. monocytogenes serotypes 1/2a, 1/2b and 4b were the most frequently identified in food and clinical strains. Here

Culture-dependent and metagenomic analysis of lesser horseshoe bats' gut microbiome revealing unique bacterial diversity and signatures of potential human pathogens.

Bats are highly diverse and ecologically important mammals. They harbor various bacteria, viruses, and fungal communities that are either beneficial or potentially pathogenic. Extensive metagenomic studies in bats are limited, particularly for the gut, and to date, there are no reports on the bacterial diversity of Rhinolophus monoceros from Meghalaya, India. There are limited studies on the isolation of potential harmful or beneficial bacteria and their interactions with the environment through culture-dependent approaches. Therefore, high-throughput screening was used to understand the popul
